The activity of various lipases was compared, in both free and immobilized forms, using the kinetics of the hydrolysis reaction of p-nitrophenyl butyrate, which was followed with in situ UV/Vis diode array spectrophotometry. Several enzymes were used to catalyze the reaction, namely Candida antarctica lipase B and Fusarium solani pisi cutinase wildtype and three single-mutation variants. The enzymes were tested in three different forms: free, immobilized as cross-linked aggregates and supported on zeolite NaY. A simple kinetic model was used to allow a quantitative comparison of the behavior of the different catalysts. It was concluded that although immobilization reduces the activity of the enzyme, the zeolite offers a much higher specific activity when compared to the cross-linked aggregates, thus supplying a heterogeneous catalyst with promising catalytic properties. 相似文献
Summary A possible cause of non-disjunction of chromosome 21 in Down Syndromes has been cytogenetically evaluated by examining the parents by Ag-staining technique. In all the cases studied so far, the contributing parents have active ribosomal cistrons on both chromosomes 21 i.e. both chromosomes are stained positively by silver staining. These results show that the active NORs might play an essential role in meiotic non-disjunction. Furthermore, the preliminary results demonstrate that the acrocentric associations of homologous and non-homologous nature involving chromosome 21 are the most frequent in the contributing parent which may further indicate the role of multiple cellular factors affecting the associations in promoting the non-disjunction in addition to active NORs. Wing imaginal disc cells in Drosophila develop by using information received from a signaling center associated with the anterior/posterior compartment border. We show here that disc cells have thin, actin-based extensions (cytonemes) that project to this signaling center. Cytonemes can be induced when cells from the lateral flanks of a wing disc are cultured next to cells from the A/P border or next to a source of fibroblast growth factor. Mouse limb bud cells also grow projections during a brief culture period, indicating that cytonemes are an attribute of both vertebrate and invertebrate cells. We suggest that cytonemes may be responsible for some forms of long-range cell-cell communication. 相似文献
Three- to four-hour exposure to fluorescnt light, one to three times weekly, reproducibly enhanced the proliferation rate of human diploid fibroblasts. This enhancement was observed in WI-38 and a line from whole embryo mince at late population doubling level (PDL) as well as in a line from adult skin at early PDL. Single or multiple exposures of short duration stimulated proliferation, whereas exposures of long duration were cytotoxic. This proliferative response is reversible, and is mediated through the culture medium, Dulbecco Vogt's supplemented with 10% fetal bovine serum. Apparently light produces some mitogenic substance(s) in the culture medium that accumulates in the cells and is toxic or growth-stimulatory depending on its concentration per cell. Another possibility is that light produces in the medium both cytotoxic and growth-stimulatory substances. 相似文献

Seriola lalandi is an economically important species that is globally distributed in temperate and subtropical marine waters. Aquaculture production of this species has had problems associated with intensive fish farming, such as disease outbreaks or nutritional deficiencies causing high mortality. Intestinal microbiota are involved in many processes that benefit a host, such as disease control, stimulation of the immune response, and the promotion of nutrient metabolism. The aim of this study is to evaluate the in vitro probiotic properties of bacteria isolated from the intestinal content of wild Seriola lalandi. The probiotic potential was evaluated in terms of (i) the antimicrobial activity against vibrios causing outbreaks in farmed fish; (ii) the ability to stimulate genes related to an innate immune response in fish; and (iii) antibiotic resistance. Nineteen isolates identified as Pseudomonas, Shewanella, Psychrobacter, and Acinetobacter showed antimicrobial activity and significant relative expression of cytokines, serum amyloid A protein (SAA), hepcidin, and lysozyme. A positive correlation was observed between the levels of expression and the bacterial load after 24 h of exposure. Pseudomonas isolates showed a level of antibiotic resistance. In conclusion, isolates of the genera Shewanella, Psychrobacter, and Acinetobacter could serve as potential probiotics in S. lalandi culture.
Picrorhiza kurroa is a medicinal perennial herb of economic importance due to its hepatoprotective properties mainly accounted by picroside I and picroside II. To fulfill the current demand of the market indiscriminate collection from its natural habitat pose a great threat to this endangered species. To strategize the conservation of natural populations, a set of 20 highly informative novel genic SSR markers were identified. The utility of these makers was successfully tested for the genetic diversity characterization of P. kurroa populations (n = 28) from three geographical locations. These markers produced 136 alleles (average of 6.8) with mean observed heterozygosity, expected heterozygosity, Shannon’s information index, and PIC value of 0.971, 0.798, 1.681, and 0.737 respectively, revealing a higher extent of genetic diversity in P. kurroa. Further, clustering of all the individuals according to their geographical locations indicates at a spatial population structure in P. kurroa. The current study suggests that informative SSR makers identified here can be potentially used for diversity characterization targeting wider geographical locations for selection of elite/quality genotypes for commercial cultivation and genetic rescue of this endangered species. 相似文献
