Oxidation of Acetate by Various Strains of Bacillus popilliae   总被引:5,自引:3,他引:2       下载免费PDF全文
A number of strains of Bacillus popilliae were examined for their ability to oxidize acetate. Some of these would not sporulate in vitro, and some were oligosporogenous. The ability to oxidize acetate varied widely among the strains tested. A culture derived from spores of the parent strain produced in vivo and one of the asporogenous strains derived from it failed to produce any significant levels of (14)CO(2) from [(14)C]acetate. Oligosporogenous strains derived from the same parent culture all produced (14)CO(2) from both [1-(14)C] and [2-(14)C]acetate but at relatively low rates. The highest rates of acetate oxidation were observed with three strains which did not produce spores in vitro. When cultured under appropriate conditions, one of these strains displayed a secondary growth response concomitant with a decrease in the titratable acidity and an increase in the pH of the medium. The data indicate that B. popilliae has a complete citric acid cycle but that the activity of the cycle is strongly repressed in wild-type strains under the usual conditions used for in vitro cultivation.  相似文献

The changes in activities of glyoxysomal and peroxisomal enzymes have been correlated with the fine structure of microbodies in cotyledons of the cucumber (Cucumis sativus L.) during the transition from fat degradation to photosynthesis in light-grown plants, and in plants grown in the dark and then exposed to light. During early periods of development in the light (days 2 through 4), the microbodies (glyoxysomes) are interspersed among lipid bodies and contain relatively high activities of glyoxylate cycle enzymes involved in lipid degradation. Thereafter, these activities decrease rapidly as the cotyledons expand and become photosynthetic, and the activity of glycolate oxidase rises to a peak (day 7); concomitantly the microbodies (peroxisomes) become preferentially associated with chloroplasts.  相似文献

A motile, predacious bacterium which degraded Pythium debaryanum was strongly attracted to substances released into the medium by the fungus. A nonpredacious bacterium was not attracted to these substances. The predator bacterium was specifically attracted to cellulose and its oligomers which are known to be components of the cell wall of Pythium. Ethanol inhibited chemotaxis of the bacterium without affecting either its motility or its ability to degrade cellulose. A second predacious bacterium was isolated for the alga, Skeletonema costatum. The role of chemoreception in the detection of microbial prey by bacterial predators in natural habitats is discussed.  相似文献

The motion of a population of chemotactic bacteria in a radial exponential gradient of attractant in a cylindrical container has been calculated using a mathematical model suggested by Keller and Segel. Numerical solutions for the equations of bacterial migration have been found which give for all times the cell density at distances from the center of the cylinder. The ultimate distribution of bacteria is a simple stationary exponential function of the distance. Experiments to verify the theoretical predictions are suggested.  相似文献

A coupling constant-dihedral angle correlation for the H? Cα? Cβ? H system of amino acid residues in peptides has been derived from a set of model compounds covering the full range of dihedral angles. The expression obtained, J = 11.0 cos2 θ ?1.4 cos θ + 1.6 sin2θ, is close to those already used in pmr studies of peptide conformation, and provides a firmer foundation for them. Analogues of d-glucose modified at C-3, and in some cases at a second position, were prepared and tested for active accumulation by everted segments of hamster intestine. Their relative affinity for the sugar carrier was measured by tissue/medium ratio, Michaelis-Menten kinetics and competitive inhibition of d-galactose or methyl alpha-d-glucoside transport. d-Glucose and its 3-deoxy-3-fluoro, 3-chloro-3-deoxy and to a smaller extent its 3-bromo-3-deoxy derivatives, bound and were transported more strongly than 3-deoxy-d-glucose and other sugars not containing an electronegative atom in the gluco configuration at C-3. 3-Deoxy-d-galactose, 3,6-dideoxy-d-glucose and d-gulose, which have two alterations from the d-glucose structure, were not, or only very weakly, transported. The results are interpreted as indicating the presence of a hydrogen bond from the carrier to the hydroxyl group at C-3 of d-glucose. Spatial requirements are also discussed. New syntheses are reported for 3-chloro-3-deoxy- and 3-bromo-3-deoxy-d-glucose and 3,6-dideoxy-d-glucose.  相似文献

The effect of several controlled variables on the peak titer and fold increase of Rift Valley fever virus grown in suspension culture on two variants of Earle's L cell, L-DR and L-MA clone 1-1, was studied. No significant amount of cell-associated virus was found at 24 hr, indicating a release of virus soon after its formation. Mild sonic treatment of the virus produced in serum-free medium increased the infective titer about 10x. This difference was not observed with virus produced in medium supplemented with serum. Peak titer was not affected by medium used during the infection period, by multiplicity of inoculum (MOI), or by initial cell concentration within the test range of 10(4) to 2 x 10(6) cell/ml. Cell strain employed influenced titer, because the L-DR cell did not produce virus efficiently at low MOI and low initial cell concentration. The time of peak titer and fold replication was dependent on MOI and initial cell concentration. Differences in virus propagation in monolayer and suspension systems are discussed.  相似文献
