We determined the crystal structures of three nucleosome core particles in complex with site-specific DNA-binding ligands, the pyrrole-imidazole polyamides. While the structure of the histone octamer and its interaction with the DNA remain unaffected by ligand binding, nucleosomal DNA undergoes significant structural changes at the ligand-binding sites and in adjacent regions to accommodate the ligands. Our findings suggest that twist diffusion occurs over long distances through tightly bound nucleosomal DNA. This may be relevant to the mechanism of ATP-dependent and spontaneous nucleosome translocation, and to the effect of bound factors on nucleosome dynamics.  相似文献

Circulating nucleic acids (CNA) are present in small amounts in the plasma of healthy individuals. However, increased levels of plasma CNA have been reported in a number of clinical disorders like cancer, stroke, trauma, myocardial infarction, autoimmune disorders, and pregnancy-associated complications. CNA has received special attention because of its potential application as a non-invasive, rapid and sensitive tool for molecular diagnosis and monitoring of acute pathologies and the prenatal diagnosis of fetal genetic diseases. This review throws light on the current status of blood CNA as a diagnostic marker and its potential as a powerful tool in the future.  相似文献

Mannheimia haemolytica is a leading causative agent of pasteurellosis in ruminants. Genome of M. haemolytica strains from different hosts has been sequenced worldwide to understand its pathogenesis. There are only few reports on the isolation of M. haemolytica in India with limited information on its molecular characteristics. The present study focuses on genome sequence analysis of a M. haemolytica strain isolated from pneumonic sheep. Mannheimia haemolytica A2 strain NIVEDI/MH/1 was isolated and identified by species and serotype-specific PCRs. Whole genome sequencing was performed using the Ion Torrent Personal Genome Machine. A comparative genomic analysis was performed to understand the virulence determinants of the Indian strain and its phylogenetic relationship with other global strains. Sequence data revealed a draft genome of 2,211,426 bp size with 41.3% GC content, assembled into 17 contigs, and contained 2379 genes. Five genomic islands identified in the genome showed high sequence identity with other respiratory pathogens of the Pasteurellaceae family. Phylogenetic analysis showed M. haemolytica A2 NIVEDI/MH/1 is very close to a M. haemolytica A2 strain from pneumonic calf. Further, the analysis revealed the presence of virulence, metal-, and multidrug resistance genes needed for pathogenesis and survival of the bacteria during infection. Also, we identified the presence of type I-C and type II-C of CRISPR-Cas arrays in the present sequenced genome. The study emphasizes the role of M. haemolytica in respiratory infections of ruminants in the Indian subcontinent and indicates the role of vertical and horizontal gene pools in pathogenicity and survivability of the bacteria.  相似文献

Biologically inspired synthesis of nanoparticles was found to be more attractive in metal nanoparticle synthesis. The present study reported an in-situ biogenic synthesis of silver nanoparticles (AgNPs) using Solanum trilobatum aqueous leaf extract. On this basis, the aqueous leaf extract of S. trilobatum acted as a reducing agent and stabilizing agent to synthesize highly stable AgNPs at ambient temperature. Eventually, the synthesized and stabilized AgNPs surface plasmon resonance was near 430 nm through a UV–visible (UV–vis) spectrophotometer. Here, the stability of the silver colloids monitored through zeta potential and mean particle size was evaluated through diffraction light scattering (DLF). Further, the average particle size was found to be 27.6 nm and spherical, confirmed with transmission electron microscopy (TEM). Also, colloidal AgNPs and aqueous extract are found to be rich sources of antioxidants and exhibit higher free radical scavenging ability. Thus, efficient inhibition with COX1 and COX2 enzymes and the protective effect with human red blood cell (HRBC) membrane stability showed significant results. These features are promising, suggesting the possibility of the AgNPs to be useful to disease-modifying for treating inflammatory disorders and associated complications.  相似文献

The estimation of genetic instability by direct extent of DNA damage and repair is an important aspect of studies on mutagenesis, carcinogenesis, aging and evolution. Different methods have been introduced from time to time in an effort to meet this need. Single cell gel electrophoresis (SCGE) assay is a new, simple and sensitive method of evaluating DNA damage and repair at individual cell level. This assay can be performed on extremely small number of cells and results can be obtained within a relatively short time. The SCGE assay has the potential to play an important role not only in the understanding of some of the fundamental aspects of human biology but also can be helpful in many practical ways. Stem cells are unspecialized cells that can self renew indefinitely and differentiate into several somatic cells given the correct environmental cues. In the stem cell niche, stem cell-extracellular matrix (ECM) interactions are crucial for different cellular functions, such as adhesion, proliferation, and differentiation. Nitrosourea therapeutics occupies a definite place in cancer therapy but its exact mechanism of action has yet to be established. Nimustine, a chloroethyl nitrosourea derivative, is used to treat various types of malignancy including gliomas. The present work focuses on the understanding of nimustine interaction with DNA to delineate its mechanism at molecular level. Attenuated total reflection-Fourier transform infrared (ATR-FTIR) has been used to determine the binding sites of nimustine on DNA. Circular dichroism (CD) spectroscopy has been used to confirm conformational variations in DNA molecule upon nimustine-DNA interaction. Thermodynamic parameters of nimustine-DNA reaction have been calculated by isothermal titration calorimetry. Results of the present study demonstrate that nimustine is not a simple alkylating agent rather it causes major grove-directed-alkylation. Spectroscopic data suggest binding of nimustine with nitrogenous bases guanine (C6 = O6) and thymine (C4 = O4) in DNA major groove. CD spectra of nimustine-DNA complexes point toward the perturbation of native B-conformation of DNA and its partial transition into C-form. Thermodynamically, nimustine-DNA interaction is an entropy driven endothermic reaction, which suggests hydrophobic interaction of nimustine in DNA-major groove pocket. Spectral results suggest base binding and local conformational changes in DNA upon nimustine interaction. Investigation of drug-DNA interaction is an essential part of rational drug designing that also provides information about the drug’s action at molecular level. Results, demonstrated here, may contribute in the development of new nitrosourea therapeutics with better efficacy and fewer side effects.  相似文献

Polyhydroxyalkanoates (PHAs) are natural polyesters produced by microbes, a potential alternative to synthetic plastics. Various methods ranging from gravimetry to spectrophotometry are routinely used for qualitative analysis of extracted PHA. There is a great need for accurate quantification of intracellular PHA during bioprocess. Hence, the present study aims to improvise the existing Nile red-based flow cytometry protocol. It was achieved using respective cells in a non-PHA accumulating state as gating control to minimize non-specific staining. The optimal Nile red concentration required for PHA staining is 5?×?103?pg?mL?1, which is ~103-fold less than that of earlier reports. Further, it was inferred that flow-based quantification was more accurate than the gravimetric method. The intracellular PHA content was highest in Pseudomonas sp. MNNG-S (52.06?%) among the Pseudomonas strains tested by the flow-based method. Both gravimetric and flow-based cell cycle analyses revealed that DNA synthesis (S phase) and PHA production (log phase) are synchronous at 24–48?h of culture. This study supports flow-based PHA quantification for real time online measurement of intracellular PHA for bioreactor monitoring, control and optimization enduing industrial applications.  相似文献
