Effects on the male endocrine system of long-term treatment with gonadotropin-releasing hormone agonists and estrogens in male-to-female transsexuals.

We studied hormonal changes resulting from long-term treatment with gonadotropin-releasing hormone agonist and 17beta estradiol valerate in 40 healthy middle-aged male-to-female transsexuals over a period of two years. All of the participants received injections of 3.8 mg goserelin acetate every four weeks in combination with 6 mg oral 17beta estradiol valerate per day for cross-sex hormone treatment for male-to-female transsexuals. There was a significant reduction in the levels of serum luteinizing hormone and follicle-stimulating hormone to the hypogonadal stage. Mean testosterone levels decreased by 97% to 0.52 and 0.59 nmol/l after 12 months and 24 months, respectively. There was a significant reduction in dehydroepiandrosterone sulfate by 37% after 12 months and 43% after 24 months, and androstendione by 29% after 12 months and 27% after 24 months, respectively. Cortisol levels were reduced by 43% and 50%, respectively. Estrogen levels were significantly increased from 77.51 to 677 after 12 months and 661 pmol/l after 24 months. Sex hormone-binding globulin and corticoid-binding globulin levels were significantly increased after 12 and 24 months. There was a significant decrease in all measured androgen fractions and cortisol during long-term treatment with gonadotropin-releasing hormone agonist and 17beta estradiol valerate. Apart from suppression of testicular hormone production, one possible interpretation is that treatment with long-term gonadotropin-releasing hormone agonist and 17beta estradiol valerate influences adrenal hormone levels in healthy middle-aged male-to-female transsexuals. Cortisol serum levels may be decreased due to estrogen-induced increase in corticoid-binding globulin.     

Spilanthol-related amides from Acmella ciliata

From the flower heads of Acmella ciliata amides closely related to spilanthol were isolated. They possess isobutylamine, 2-phenylethylamine and 2-methylbutylamine parts. In the acidic part deca-2E,6Z,8E-trienoic acid, known from spilanthol, and its 2,3-dihydro derivative were found. Two compounds bear ester groups, a novelty for unsaturated amides. The structures were elucidated by means of high field ¹H NMR and high resolution mass spectrometry.     

Involvement of human MCM8 in prereplication complex assembly by recruiting hcdc6 to chromatin

The MCM2-MCM7 complex is an essential component of the prereplication complex (pre-RC), which is recruited by the cdc6 and cdt1 proteins to origins of DNA replication during G(1) phase. Here, we report that the accumulation on chromatin of another member of the MCM protein family, human MCM8 (hMCM8), occurs during early G(1) phase, before the hMCM2-hMCM7 complex binds. hMCM8 interacts in vivo with two components of the pre-RC, namely, hcdc6 and hORC2. Depletion of endogenous hMCM8 protein by RNA interference leads to a delay of entry into S phase, suggesting a role for hMCM8 in G(1) progression. Furthermore, down-regulation of hMCM8 also leads to a reduced loading of hcdc6 and the hMCM2-hMCM7 complex on chromatin. These results suggest that hMCM8 is a crucial component of the pre-RC and that the interaction between hMCM8 and hcdc6 is required for pre-RC assembly.     

The reefal margin and slope of a Middle Triassic carbonate platform: the Latemar (Dolomites, Italy)

The Latemar is a mainly aggrading platform, but shows repeated backstepping during its entire development. The behaviour of the slope does not reflect accommodation changes and lateral consistencies of the lagoonal interior; the Latemar contemporaneously reveals different, even contrasting depositional characteristics. The slope of the late stage platform evolution corresponds at least partially to the base-of-slope apron model. Controlling factors on slope evolution are of tectonic (proximity of the Stava Line) and autocyclic (repeated oversteepening) nature. Other factors are insignificant and/or overprinted. The reef-facies at Latemar reveals a complex facies pattern; it varies along and across the margin and is rich in encrusting sponges, corals, biogenic crusts and Microproblematica. Some biota or fossil assemblages—e.g. foraminifers (Abriolina mediterranea, Turriglomina scandonei) or Tubiphytes multisiphonatus thrombolites—have not been described in the Dolomites before. Biostratigraphic evidence from the uppermost reef-facies confirms a mainly Anisian age of the outcropping platform interior.     

Use of flow-cytometric analysis to optimize cell banking strategies for production of biopharmaceuticals from mammalian cells

Production of biopharmaceuticals from mammalian cells requires generation of master, working and post-production cell banks of high quality under GMP conditions. An optimal cryopreservation strategy is needed for each new production cell line, particularly with regard to establishing production processes that are completely devoid of serum or even any animal components and to ensuring robust thaw performance for reliable production. Here, we describe a novel strategy employing flow-cytometric (FC) analysis of Annexin V-stained cells for high-throughput characterization of cell banks. Our data show that this method enables predictive evaluation of a cryopreservation strategy as early as 6h after thawing of cells. Furthermore, a broad study is presented characterizing various factors that may influence the quality of serum-free production cell banks from NSO and CHO cell lines. These results demonstrate how FC-based analysis can be used for development of future state-of-the-art cryopreservation strategies.
