In alkaline calcareous waters, hydrophytes become encrusted with carbonate precipitates of a complex nature which alter bulk chemical analyses of the plants. The manganese, zinc and copper concentrations, and the ash content of two aquatic plants, Egeria densa and Potamogeton nodusus, differed considerably depending on the cleaning procedure used and, therefore, the amount of marl encrustation included in the analyses. Tapwater washed plants, retaining the marl, consistently had ash contents two to three times higher than acid-cleaned plants and exhibited higher trace metal concentrations, especially manganese.  相似文献

A major detoxification mechanism involving the binding of excess metals to inorganic, insoluble metalliferous granules occurs in many invertebrates. X-ray microanalysis of granules extracted from the barnacles Elminius modestus Darwin and Semibalanus balanoides (L.) (Crustacea: Cirripedia) indicated that one granule type was predominant. Examples of this granule type usually contained Fe and P, or Zn and S, or a combination of all four elements as major constituents. The predominant form of P in the granules was pyrophosphate, P2O4−7, with oprthophosphate and other polyphosphates present in lower amounts. The S was present as organic S. Biochemical analysis showed high levels of Ca, Fe, Mg, K and Zn in the granules with Cu and Ag present in smaller quantities. Considerable variation was shown between granules.  相似文献

We have used adenovirus as a molecular probe to examine the recombination of viral DNA following infection of mammalian cells. The technique gives a quantitative measure of homologous recombination between adenovirus type 2 (Ad2) and Ad5PyMTR3. Ad5PyMTR3 is an insertion mutant of Ad5 containing polyoma virus (Py) DNA inserted into a deleted E1 region of the Ad5 genome. Cells were coinfected with Ad2 and Ad5PyMTR3 and at an appropriate time after infection, viral DNA was extracted from the infected cells, digested with restriction endonuclease and electrophoresed through an agarose gel. Although Ad2 and Ad5 have more than 99% DNA homology, they differ sufficiently in their restriction endonuclease patterns, such that recombinant viral DNA molecules containing the Py insert could be detected and quantified by Southern blotting and hybridization to a radioactive Py DNA probe. Using this method we are able to detect and quantitate recombinant viral DNA molecules containing the Py insert which are present at frequencies down to at least 1 in 100. Recombination was detected in Chinese hamster ovary cells, monkey kidney cells, human HeLa cells, normal human fibroblasts and SV40 transformed human fibroblasts. In experiments using HeLa cells, the frequency of recombination between the Py insert on Ad5PyMTR3 and a number of unique restriction enzyme sites on Ad2 increased with the distance from the Py insert to the restriction site. Also in HeLa cells, recombination increased with increasing amounts of viral DNA synthesis and with increasing UV dose to the virus. UV-irradiation of both coinfecting viruses with 1500 J/m2 resulted in a more than 100-fold reduction in the amount of viral DNA synthesized and about a 3-fold increase in the frequency of recombination.  相似文献

Although non-human primates are widely used models of human diseases, often studied for long periods of time, their dietary requirements are not well defined. Over a two—three-year time period, female rhesus macaques were fed either a marginally zinc-deprived diet or a comparable zinc replete purified diet. The purified diets were based on current NRC recommendations. Interestingly poor pregnancy outcome was noted in both the zinc-deprived as well as in the control group. Twenty-eight percent of controls (N=16) and 20% of zinc-deprived animals (N=15) failed to conceive after at least seven matings. Pregnancy loss was also high in both groups of animals including 31% of conceptions in controls (N=11) and 33% of conceptions in zinc deficient animals (N=12). The majority of pregnancy losses occurred after animals had been fed purified diet for over two years. In contrast, normal colony pregnancy loss is estimated at 11%. Furthermore, 3 of 18 live-born infants died prior to 7 months of age and one third of surviving offspring were growth-retarded during the 1st year of life compared to established colony norms. Our data demonstrate the difficulty of constructing adequate purified diets for long-term studies in primates even when based upon available and recommended nutritional information.  相似文献

A brief review is given of experiments which are concerned with the hypothesis that brain RNA and protein synthesis are directly involved in the establishment of long-term memory. It is concluded that these experiments neither support or refute this hypothesis. A convincing demonstration is lacking of interanimal memory transfer by injection of macromolecular extracts. The majority of experiments which attempt to correlate increased macromolecular synthesis with learning use radioactive precursor methods and these studies do not exclude possible changes in precursor specific activity as the cause of the increased labeling. Although some studies find directly observable changes in brain macromolecules in response to training, their relationship to memory formation is unclear. It is possible that these changes represent only an enhanced production of constitutive macromolecules in response to an increase in cerebral metabolism during training, rather than molecular changes that are directly involved with modifying synaptic connectivity. Inhibitors of cerebral protein synthesis block memory formation, but these drugs are not pharmacologically specific and this complicates the interpretation of these studies.  相似文献

An enhanced reactivation of UV-irradiated adenovirus type 2 (Ad 2) was detected following irradiation of the host cells with γ-rays prior to infection. Non-irradiated and γ-irradiated normal human fibroblasts were infected immediately after irradiation with either non-irradiated or UV-irradiated Ad 2. At 48h after infection, cultures were examined by indirect immunofluorescence to determine the number cells in which the viral function of viral structural antigen (Vag) was expressed. Pre-irradiation of cells with 1 krad resulted in a 2–3-fold increase in the survival of this viral function following different UV doses to the virus up to 1.75 × 103 J/m2. For a fixed UV dose of 1.0 × 103 J/m2 to the virus this enhancement increased with preirradiation dose to the cells up to a maximum factor of 2–3 for a dose of 1 krad. An examination of Vag expression at various times after infection indicates that pre-irradiation of the cells with γ-rays prior to infection with UV-irradiated virus leads to an earlier onset and/or increased rate of Vag synthesis. This enhancement of Vag production from a UV-damaged template may result from an inducible DNA-repair mechanism in human fibroblasts which may or may not be error-prone.  相似文献

The alimentary tract of barnacles is made up of cuticle-lined foregut and hindgut with an intervening U-shaped midgut associated anteriorly with a pair of pancreatic glands and perhaps midgut caeca. Epithelial salivary glands secrete acid mucopolysaccharide, glycoprotein or both. Cells of all the midgut regions are capable of absorption which is carried out mainly by the anterior midgut and caeca. Midgut cells of Balanus balanoides (L.) show a seasonal variation in the distribution of intracellular lipid droplets. Midgut cells rest on an elastic basal lamina and secrete a peritrophic membrane which contains mucopolysaccharide and protein. Cells of the stratum perintestinale connect with the midgut epithelial cells via cell processes which probably translocate absorbed materials. Glycoprotein globules and lipid droplets accumulate in the body parenchyma of B. balanoides and are transported to the ovaries to form yolk (glycolipovitellin). The pancreatic gland cells of all barnacles are active secretory cells secreting proteinaceous material (probably digestive enzymes).  相似文献

Shiga toxin-producing Escherichia coli (STEC) is a food-borne pathogen that may be responsible for severe human infections. Only a limited number of serotypes, including O26:H11, are involved in the majority of serious cases and outbreaks. The main virulence factors, Shiga toxins (Stx), are encoded by bacteriophages. Seventy-four STEC O26:H11 strains of various origins (including human, dairy, and cattle) were characterized for their stx subtypes and Stx phage chromosomal insertion sites. The majority of food and cattle strains possessed the stx1a subtype, while human strains carried mainly stx1a or stx2a. The wrbA and yehV genes were the main Stx phage insertion sites in STEC O26:H11, followed distantly by yecE and sbcB. Interestingly, the occurrence of Stx phages inserted in the yecE gene was low in dairy strains. In most of the 29 stx-negative E. coli O26:H11 strains also studied here, these bacterial insertion sites were vacant. Multilocus sequence typing of 20 stx-positive or stx-negative E. coli O26:H11 strains showed that they were distributed into two phylogenetic groups defined by sequence type 21 (ST21) and ST29. Finally, an EspK-carrying phage was found inserted in the ssrA gene in the majority of the STEC O26:H11 strains but in only a minority of the stx-negative E. coli O26:H11 strains. The differences in the stx subtypes and Stx phage insertion sites observed in STEC O26:H11 according to their origin might reflect that strains circulating in cattle and foods are clonally distinct from those isolated from human patients.  相似文献

The plant circadian clock regulates the rhythms of plant metabolism. Many herbivore-induced plant volatiles(HIPVs) fluctuate, diurnally, but the role of the circadian clock in the emission of HIPVs and their ecological consequences remains largely unknown.Here, we show that the timing of herbivore attack can alter the outcome of tri-trophic interactions, and this is mediated by the circadian clock, under both field and glasshouse conditions. Although most HIPV emissions did not have a circadian rhythm, the circadian clock modulated HIPV emissions in a time-dependent manner. HIPVs mediate tri-trophic interactions, and the circadian clock may affect these interactions by modulating HIPV emission in nature.  相似文献
