The occurrence of a penis in female gastropods has commonly been accepted as an indicator of tributyltin (TBT) pollution. During the surveys performed in Patagonian waters between 2002 and 2010, individuals of Buccinanops deformis with a short penis and an external vas deferens were initially considered as showing imposex. Doubt arose when different manifestations of the phenomenon were identified at the two sites with the densest populations of the species (San Antonio Bay, SAB, and Villarino). This led us to search at both locations for: (a) histological confirmation of sex, (b) butyltin measurements in sediments and snail tissues, and (c) incidence of trematode infestation. We found that both populations have individuals with a short penis. While all the inspected individuals from SAB were histologically identified as females (imposex), those from Villarino were identified as males. Trematodes were absent in individuals from SAB and showed very low prevalence in those from Villarino, with metacercariae found in the digestive gland. In snail tissues, butyltins were found mainly in the form of dibutyltin (DBT) in SAB and at negligible levels in Villarino. In sediments, butyltins were found only in the form of TBT, at higher levels in SAB than in Villarino. Thus, it became clear that our finding of a short penis is a manifestation of imposex on females from SAB related to the high TBT levels, whereas in Villarino it is a male trait, which is not related to TBT or a trematode infestation but may be a Dumpton syndrome-like abnormality.  相似文献

Targeted muscle reinnervation (TMR) is a surgical procedure used to redirect nerves originally controlling muscles of the amputated limb into remaining muscles above the amputation, to treat phantom limb pain and facilitate prosthetic control. While this procedure effectively establishes robust prosthetic control, there is little knowledge on the behavior and characteristics of the reinnervated motor units. In this study we compared the m. pectoralis of five TMR patients to nine able-bodied controls with respect to motor unit action potential (MUAP) characteristics. We recorded and decomposed high-density surface EMG signals into individual spike trains of motor unit action potentials. In the TMR patients the MUAP surface area normalized to the electrode grid surface (0.25 ± 0.17 and 0.81 ± 0.46, p < 0.001) and the MUAP duration (10.92 ± 3.89 ms and 14.03 ± 3.91 ms, p < 0.01) were smaller for the TMR group than for the controls. The mean MUAP amplitude (0.19 ± 0.11 mV and 0.14 ± 0.06 mV, p = 0.07) was not significantly different between the two groups. Finally, we observed that MUAP surface representation in TMR generally overlapped, and the surface occupied by motor units corresponding to only one motor task was on average smaller than 12% of the electrode surface. These results suggest that smaller MUAP surface areas in TMR patients do not necessarily facilitate prosthetic control due to a high degree of overlap between these areas, and a neural information—based control could lead to improved performance. Based on the results we also infer that the size of the motor units after reinnervation is influenced by the size of the innervating motor neuron.  相似文献

A study of a stocked upland reservoir trout fishery, using gill nets, revealed the presence of a large stock of pike and poor trout numbers. The overall growth rate of pike was slow. Two year-classes of pike dominated, but the broad age and length frequency ranges, and the high average weight of individuals were indicative of an unexploited stock. Higher than normal water levels during the spawning period and into the summer period in both 1985 and 1986, coincided with the dominant pike year-classes recorded several years later. Freshly isolated human mucosal T lymphocytes in vitro can markedly diminish an important property of intestinal epithelium—its barrier function. On the other hand, cytokines and their cellular receptors, which maintain homeostasis of epithelia, limit epithelial permeability, and preserve barrier function, are not well characterized. Using a described human colonic epithelial cell monolayer system, we found that transforming growth factor-β1 (TGF-β1) preserved 75% or more of epithelial barrier function, quantitated electrophysiologically, even in the presence of cytokines generated by a high density of barrier-disruptive mucosa-derived mononuclear cells. In opposing the TGF-β1 effect, cytokines able to reduce barrier function were spontaneously secreted by mucosal T cells and were increased in their barrier effect after T-lymphocyte activation. Further, neutralization of individual cytokines with specific monoclonal antibodies abrogated the lymphocyte-induced reduction in epithelial barrier function, and identified interferon gamma (IFN-γ), interleukin (IL)-4, and IL-10, but not IL-6, as the primary cytokines whose barrier effects were curtailed by TGF-β1. Receptors (RI and RII) for TGF-β1 were found to be localized primarily to the apical and basal membranes of surface epithelium in colonic crypts. These findings provide the scientific basis for new strategies to pharmacologically enhance the barrier function of epithelia in mucosal organs regularly exposed to environmental antigens and to T-lymphocyte products. The adhesive interactions of nerve growth cones stabilize elongating nerve fibers and mediate transmembrane signaling to regulate growth cone behaviors. We used interference reflection microscopy and immunocytochemistry to examine the dynamics and composition of substratum contacts that growth cones of chick sensory neurons make with extracellular adhesive glycoproteins, fibronectin and laminin. Interference reflection microscopy indicated that sensory neuronal growth cones on fibronectin-treated substrata, but not on laminin, make contacts that have the appearance and immobility of fibroblastic focal contacts. Interference reflection microscopy and subsequent immunocytochemical staining showed that β1 integrin and phosphotyrosine residues were concentrated at growth cone sites that resemble focal contacts. Two other components of focal contacts, paxillin and zyxin, were also co-localized with concentrated phosphotyrosine residues at sites that resemble focal contacts. Such staining patterns were not observed on laminin-treated substrata. Growth cone migration on fibronectin-treated substrata was inhibited by herbimycin A, a tyrosine kinase inhibitor. We conclude that sensory neuronal growth cones distinguish fibronectin from laminin by making contacts with distinct organization and regulation of cytoskeletal components at the adhesive sites. This finding suggests that growth cone interactions with different adhesive molecules lead to distinctive transmembrane organization and signaling to regulate nerve fiber elongation. © 1996 John Wiley & Sons, Inc.  相似文献
