One of the challenges encountered in microRNA (miRNA) studies is to observe their dual role in different conditions and cells. This leads to a tougher prediction of their behavior as gene expression regulators. miR-203 has been identified to play a negative role in the progression of malignant melanoma; however, it has been reported, with dual effect, as both an oncomiR and tumor suppressor miRNA in some malignancies, such as breast cancer, meanwhile, the role of miR-203 in melanoma stem cells or even metastatic cells is unclear. In the present study, after observation of upregulation of miR-203 in melanoma patient's serum and also melanospheres as cancer stem cells model, we examined its overexpression on the stemness potential and migration ability of melanoma cells. Our data demonstrated that the increased miR-203 level was significantly associated with significant increase in the ability of proliferation, colony and spheres formation, migration, and tumorigenesis in A375 and NA8 cells. All of these changes were associated with enhancement of BRAF, several epithelial to mesenchymal transition factors, and stemness genes. In conclusion, our results clearly determined that miR-203 could be down-regulateddownregulated in melanoma tissues but be overexpressed in melanoma stem cells. It has an important role as oncomiR and promote repopulation, tumorigenicity, self-renewal, and migration. Therefore, we suggested overexpression of miR-203 as biomarker for early detection of metastasis. However, more studies are needed to validate our data. 相似文献

Since sturgeons do not show clear sexual dimorphism particularly when are small in size, attempts were made to determine the best methods to identify early sex in farmed beluga sturgeon (Huso huso). The present study describes the ultrasonography, small surgery and plasma steroid hormone methods to determine gender at 18‐month fish, which no research has been conducted yet into the fish at such small ages. Twenty one cultured beluga sturgeon's gonad were imaged using an ultrasonograph unit with a 9–13 MHz linear transducer. Overall accuracy of sex determination using ultrasonography was 80.95%. Plasma testosterone (T) levels were significantly higher in males than in females whereas 17α,20βOH‐P levels were significantly higher in females than in males. Testosterone (T) and 17α,20βOH‐P were not correlated with morphometric parameters (TL, SL, W, CF) in 18‐month beluga sturgeon. Results of this study indicated that sex could be identified by each of ultrasonography, small surgery and analysis of blood plasma in such a small size (18‐month). Although direct observation was more efficient than the other methods, ultrasonography was the simplest and cost‐effective tool in sturgeon's sex determination compared to other methods, and the least invasive. 相似文献

The gene encoding GSTO2 was localized to human chromosome 10q24.3, a region that may harbor gene(s) involved in the developing of colorectal cancer.
To investigate the association between GSTO2 N142D genetic polymorphism and susceptibility to colorectal cancer the present study was done. We studied 63 (26 females,
37 males) colorectal cancer patients and 126 (52 females, 74 males) healthy individuals. The control subjects were frequency
matched for age and gender with the colorectal cancer group. The genotypes were performed using RFLP-PCR method. The ND and
DD genotypes were not associated with risk of colorectal cancer, in comparison with the NN genotype. Family history for cancer
in the first degree of relatives significantly differed between cases and controls (P = 0.012). The profiles of GSTO2 genotypes and family history in control and cancerous groups were compared to each other. Subjects with NN genotype and positive
family history significantly were at high risk to develop colorectal cancer in comparison with subjects with DD or ND genotypes
and negative family history (P = 0.003). Present findings indicating that GSTO2 NN genotype increase the risk of colorectal cancer in persons with positive family history for cancer in the first degree

The capacity of mesenchymal stem cells (MSCs) to survive and engraft in the target tissue may lead to promising therapeutic
effects. However, the fact that the majority of MSCs die during the first few days following transplantation complicates cell
therapy. Hence, it is necessary to strengthen the stem cells to withstand the rigors of the microenvironment to improve the
efficacy of cell therapy. In this study, we manipulated MSCs to express a cytoprotective factor, heme oxygenase-1 (HO-1),
to address this issue. Full-length cDNA of human HO-1 was isolated and cloned into TOPO vector by TOPO cloning reaction. Then,
the construct was ligated to gateway adapted adenovirus expression vector by LR recombination reaction. Afterwards, the recombinant
virus expressing HO-1 was produced in appropriate mammalian cell line and used to infect MSCs. The HO-1 engineered MSCs were
exposed to hypoxic and oxidative stress conditions followed by evaluation of the cells’ viability and apoptosis. Transient
expression of HO-1 was detected within MSCs. It was observed that HO-1 expression could protect MSCs against cell death and
the apoptosis triggered by hypoxic and oxidative stress conditions. The MSCs-HO-1 retained their ability to differentiate
into adipogenic, chondrogenic, or osteogenic lineages. These findings could be applied as a strategy for prevention of graft
cell death in MSCs-based cell therapy and is a good demonstration of how an understanding of cellular stress responses can

doi:10.1111/j.1741‐2358.2009.00313.x Prevalence of oral and dental disorders in institutionalised elderly people in Rasht, Iran Objective: The aim of this study was to determine the prevalence of dental and oral mucosal lesions in institutionalised elderly people in Rasht. Background: Oral health in the elderly people is important to tailor the health programmes for this increasing age group within the population. Methods: A total of 216 elderly people residing in a geriatric nursing home were included in the study. Subjects were interviewed and examined clinically and registered in a data collection form and analysed using spss version 11 program (SPSS Inc, Chicago, Illinois, USA). Results: The prevalence of dental and oral disorders was 44.9% and 86.1% respectively. The most frequent oral disorders were dry mouth (42.1%), fissure tongue (25.9%), atrophic of tongue (25%), sublingual varicosity (22.7%), burning sensation (16.7%) and varix (15.3%). The prevalence of edentulousness was 56% (95%CI: 49–63%). The mean level of retained teeth was 3.22. The odds of an oral mucosal disorder in females were significantly more than in males (OR = 2.79, 95%CI: 1.25–6.23). Discussion: The findings revealed a high prevalence of dental and oral disorders in institutionalised elderly people in Rasht in comparison to similar studies. The mean of retained teeth was also much lower than in other reports. Therefore, the necessity for regular oral examination of these rapidly expanding age groups of people is essential. 相似文献

Acute kidney injury (AKI) is one of the most common health-threatening diseases in the world. There is still no effective medical treatment for AKI. Recently, Mesenchymal stem cell (MSC)-based therapy has been proposed for treatment of AKI. However, the microenvironment of damaged kidney tissue is not favorable for survival of MSCs which would be used for therapeutic intervention. In this study, we genetically manipulated MSCs to up-regulate lipocalin-2 (Lcn2) and investigated whether the engineered MSCs (MSC-Lcn2) could improve cisplatin-induced AKI in a rat model. Our results revealed that up-regulation of Lcn2 in MSCs efficiently enhanced renal function. MSC Lcn2 up-regulates expression of HGF, IGF, FGF and VEGF growth factors. In addition, they reduced molecular biomarkers of kidney injury such as KIM-1 and Cystatin C, while increased the markers of proximal tubular epithelium such as AQP-1 and CK18 following cisplatin-induced AKI. Overall, here we over-expressed Lcn2, a well-known cytoprotective factor against acute ischemic renal injury, in MSCs. This not only potentiated beneficial roles of MSCs for cell therapy purposes but also suggested a new modality for treatment of AKI. 相似文献
