TPX is a web-based PubMed search enhancement tool that enables faster article searching using analysis and exploration features. These features include identification of relevant biomedical concepts from search results with linkouts to source databases, concept based article categorization, concept assisted search and filtering, query refinement. A distinguishing feature here is the ability to add user-defined concept names and/or concept types for named entity recognition. The tool allows contextual exploration of knowledge sources by providing concept association maps derived from the MEDLINE repository. It also has a full-text search mode that can be configured on request to access local text repositories, incorporating entity co-occurrence search at sentence/paragraph levels. P1 nuclease is one of the most extensively used single-strand DNA specific nucleases in molecular biology. In modern biology, it is used as an enzymatic probe to detect altered DNA conformations. It is well documented that P1 cleaves single-stranded nucleic acids and single-stranded DNA regions. The fact that P1 can act under a wide range of conditions, including physiological pH and temperature make it the most commonly used enzymatic probe in DNA structural studies. Surprisingly, to this date, there is no study to characterize the influence of length of mismatches on P1 sensitivity. Using a series of radioactively labeled oligomeric DNA substrates-containing mismatches, we find that P1 nuclease cleavage is dependent on the length of mismatches. P1 does not cleave DNA when there is a single-base mismatch. P1 cleavage efficiency is optimum when mismatch length is 3 nt or more. Changing the position of the mismatches also does not make any difference in cleavage efficacy. These novel findings on P1 properties have implications for its use in DNA structure and DNA repair studies.  相似文献

Boosted by technology advancements, government and commercial interest, ad-hoc wireless networks are emerging as a serious platform for distributed mission-critical applications. Guaranteeing QoS in this environment is a hard problem because several applications may share the same resources in the network, and mobile ad-hoc wireless networks (MANETs) typically exhibit high variability in network topology and communication quality. In this paper we introduce DYNAMIQUE, a resource management infrastructure for MANETs. We present a resource model for multi-application admission control that optimizes the application admission utility, defined as a combination of the QoS satisfaction ratio. A method based on external adaptation (shrinking QoS for existing applications and later QoS expansion) is introduced as a way to reduce computation complexity by reducing the search space. We designed an application admission protocol that uses a greedy heuristic to improve application utility. For this, the admission control considers network topology information from the routing layer. Specifically, the admission protocol takes benefit from a cluster network organization, as defined by ad-hoc routing protocols such as CBRP and LANMAR. Information on cluster membership and cluster head elections allows the admission protocol to minimize control signaling and to improve application quality by localizing task mapping.  相似文献

Phosphoenolpyruvate carboxylase (PEPC) was purified from leaves of four species of Alternanthera differing in their photosynthetic carbon metabolism: Alternanthera sessilis (C3), A. pungens (C4), A. ficoides and A. tenella (C3-C4 intermediates or C3-C4). The activity and properties of PEPC were examined at limiting (0.05 mM) or saturating (10 mM) bicarbonate concentrations. The Vmax as well as Km values (for Mg2+ or PEP) of PEPC from A. ficoides and A. tenella (C3-C4 intermediates) were in between those of C3 (A. sessilis) and C4 species (A. pungens). Similarly, the sensitivity of PEPC to malate (an inhibitor) or G-6-P (an activator) of A. ficoides and A. tenella (C3-C4) was also of intermediate status between those of C3 and C4 species of A. sessilis and A. pungens, respectively. In all the four species, the maximal activity (Vmax), affinity for PEP (Km), and the sensitivity to malate (KI) or G-6-P (KA) of PEPC were higher at 10 mM bicarbonate than at 0.05 mM bicarbonate. Again, the sensitivity to bicarbonate of PEPC from C3-C4 intermediates was in between those of C3- and C4-species. Thus the characteristics of PEPC of C3-C4 intermediate species of Alternanthera are intermediate between C3- and C4-type, in both their kinetic and regulatory properties. Bicarbonate could be an important modulator of PEPC, particularly in C4 plants. This revised version was published online in August 2006 with corrections to the Cover Date.  相似文献
