Summary A technique for observing the reduction of potassium tellurite byCandidas was developed. Various species of these yeasts reduce it at different concentrations, but that which is most useful for differentiation is 0.02 % added to Sabouraud dextrose agar basal medium. Of the yeasts studied,C. albicans, C. parapsilosis andC. tropicalis all reduced potassium tellurite to the concentration mentioned before, while the growth ofC. krusei, C. parakrusei andC. pseudotropicalis was inhibited. Without exception,C. pseudotropicalis reduced this salt at lower concentrations. The two strains ofC. guilliermondii tested gave contradictory results: one of them grew and reduced potassium tellurite, while the growth of the other was inhibited.Professor of Microbiology.  相似文献

The correlation between pollen/ovule (P/O) ratio and breeding system has generally been accounted for either on the basis that P/O reflects pollination efficiency, or in terms of the sex allocation theory. The following were assessed for taxa belonging to genus Astragalus subgenus Epiglottis: 1) Degree of correlation between P/O and the breeding system, measured by means of autofertility; 2) The absence or existence of correlation between P/O and pollen grain size; and 3) The ability of various theories to account for the results obtained. Results showed a minimal correlation between P/O and autofertility, and between P/O and pollen grain size in the taxa studied. Analysis of these results in terms of the sex allocation theory enabled this correlation to be explained as a function of the variation existing between taxa with respect to the resources invested in each pollen grain and in each ovule. The predictive capacity of this theory, which has moreover proven valuable in explaining the structural peculiarities of the androecium in these taxa, was also highlighted. The type of self-pollination applicable was also discussed, as was the phenotypic model of selection of self-fertilization considered most plausible for these taxa.  相似文献

We conducted visual fish surveys in coexisting mangrove-coral (CMC) habitats in Panama to analyze the effect of coral presence in mangrove habitats on the fish assemblage. Our study revealed that CMC habitats harbor distinct fish assemblages compared to mangrove habitats without coral, with greater species richness and increased herbivore abundance. Massive elevations of serum phospholipase A2 activity have been documented in patients with septic shock. Serum PLA2 activity correlated to the degree and duration of circulatory collapse, while purified native PLA2 reproduced hypotension in experimental animals. In a prospective study of patients with septic shock, we have determined the relationship of PLA2 enzyme activity to PLA2 immunoreactivity using radiolabelled E. coli phospholipid substrate and an ELISA specific for group II human nonpancreatic PLA2. In all patients, there was a clear concordance of the two assays. Maximal PLA2 concentration was increased a mean of 554-fold over normal levels. We found no evidence to support the presence of activating or inhibitory proteins. These data confirm that the observed increase in serum PLA2 activity in septic shock is due to intravascular release of group II nonpancreatic PLA2.  相似文献

Four xylanase preparations that are commercially available, namely Cartazyme from Sandoz, Ecopulp from Alko-ICI, Irgazyme from Ciba-Genencor and Pulpzyme HB from Novo Nordisk, were tested in bleaching experiments of kraft pulps from Pinus radiata. The main objective of this study was to optimize a reduction in the consumption of chlorine dioxide in the bleaching sequences C90/D10EoDED, C70/D30EoDED and D100EDED. Enzymatic treatments led to savings of ClO2 between 3.5 and 3.9 kg per air-dried tons (ADT) in the three bleaching sequences, without affecting the target brightness of the pulps. In these assays, some minor although reproducible differences in the performance of the enzymes were observed. In most cases, xylanase treatment partially affected the beatability of the pulps, measured as the number of revolutions in the PFI mill required to reach the same tensile index as the respective controls.  相似文献

The complete nucleotide sequence of the mitochondrial DNA of the rainbow trout, Onchorynchus mykiss, has been determined. The total length of the molecule is 16,660 bp. The rainbow trout mitochondrial DNA has the same organization described in eutherian mammals, the clawed frog (Xenopus laevis), and the two fish species, Oriental stream loach (Crossotoma lacustre) and carp (Cyprinus carpio). Alignment and comparison of the deduced amino acid sequences of the 13 proteins encoded by rainbow trout and other vertebrate mitochondrial genomes allowed us to estimate that COI is the most conserved mitochondrial subunit (amino acid identity ranging from 85.6% to 94.8%) whereas ATPase 8 is the most variable one (amino acid identity ranging from 30.8% to 70.4%). Putative secondary structures for the 22 tRNAs found in the molecule are given along with an extensive comparison of tRNA sequences among representative species of each major group of vertebrates. In this sense, an unusual cloverleaf structure for the tRNASer(AGY) is proposed. A stem-loop structure inferred for the origin of the L-strand replication (OL) and the presence of a large polycytidine tract in the OL loop is described. The existence of this stretch instead of the usual T-rich sequence reported so far in mammal mtDNAs is explained in terms of a less-strict template dependence of the RNA primase involved in the initiation of L-strand replication. Abstract: Peroxynitrite is a powerful oxidant formed by the near-diffusion-limited reaction of nitric oxide with superoxide. Large doses of peroxynitrite (>2 m M ) resulted in rapid cell swelling and necrosis of undifferentiated PC12 cells. However, brief exposure to lower concentrations of peroxynitrite (EC50 = 850 µ M ) initially (3–4 h) caused minimal damage to low-density cultures. By 8 h, cytoplasmic shrinkage with nuclear condensation and fragmentation became increasingly evident. After 24 h, 36% of peroxynitrite-treated cells demonstrated these features associated with apoptosis. In addition, 46% of peroxynitrite-treated cells demonstrated DNA fragmentation (by terminal-deoxynucleotide transferase-mediated dUTP-digoxigenin nick end-labeling) after 7 h, which was inhibited by posttreatment with the endonuclease inhibitor aurintricarboxylic acid. Serum starvation also resulted in apoptosis in control cells (23%), the percentage of which was not altered significantly by peroxynitrite treatment. Although peroxynitrite is known to be toxic to cells, the present study provides a first indication that peroxynitrite induces apoptosis. Furthermore, pretreatment of cells with nerve growth factor or insulin, but not epidermal growth factor, was protective against peroxynitrite-induced apoptosis. However, both acidic and basic fibroblast growth factors greatly increased peroxynitrite-initiated apoptosis, to 63 and 70%, respectively. Thus, specific trophic factors demonstrate differential regulation of peroxynitrite-induced apoptosis in vitro.  相似文献

Synopsis The Río Bayano in eastern Panama is one of many tropical rivers in which bull sharks (Carcharhinus leucas) and largetooth sawfish (Pristis perotteti) have been known to occur. Since both species can osmoregulate in fresh water throughout life, theoretically, both could survive in landlocked situations for many years.P. perotteti reproduces in fresh water, butC. leucas ordinarily does not, so only the former would appear to have the potential for establishing a breeding stock in such a landlocked freshwater body.The damming of the Rio Bayano and the creation of a large impoundment in 1976 provides a test of the ability of members of both species trapped there to survive and to establish breeding stocks. In 1980 and 1981 three mature femaleC. leucas were found dead in Lake Bayano and three sub-adult femaleP. perotteti were taken by trammel net. These events confirm the ability of both to survive in fresh water for long periods, but the establishment of breeding stocks appears doubtful and the question may not be answered for many years.  相似文献

Rhodospirillum rubrum strain F24.1 is a spontaneous revertant of nonphototrophic mutant F24 derived from wild-type strain S1. Strain F24 shows no detectable photochemical activity and contains, at most, traces of the photoreaction center polypeptides. Strain F24.1 has a phototrophic growth rate close to that of the wild-type strain (Picorel, R., del Valle-Tascón, S. and Ramírez, J.M. (1977) Arch. Biophys. Biochem. 181, 665–670) but shows little photochemical activity. Light-induced absorbance changes in the near-infrared, photoinduced EPR signals and ferricyanide-elicited absorbance changes indicate that strain F24.1 has a photoreaction center content of 7–8% as compared to strain S1. Polyacrylamide gel electrophoresis of isolated F24.1 chromatophores shows the photoreaction center polypeptides to be present in amounts compatible with this value. Photoreaction center was prepared from strain F24.1 and showed no detectable difference with that of strain S1. It is concluded that strain F24.1 photosynthesis is due entirely to its residual 7–8% of typical photoreaction center.  相似文献
