Deoxyhaemocyanin, treated with NO under strictly anaerobic conditions, yielded methaemocyanin and N2O in a fast reaction. In a further slow reaction this methaemocyanin lost its triplet electron paramagnetic resonance (EPR) signal at g = 4 and yielded a nitrosyl derivative with a characteristic g = 2 Cu(II) EPR signal, indicating the binding of a single NO per copper pair. Thus under strictly anaerobic conditions deoxyhaemocyanin and methaemocyanin, treated with NO, gave the same derivative as shown by circular dichroism and EPR spectra. Methaemocyanin yielded, moreover, reversibly a nitrite derivative, characterized by a triplet signal at g = 4 with 7 hyperfine lines. 相似文献

The reaction of nitrite at pH 5.7 with deoxyhaemocyanin of Astacus leptodactylus yielded methaemocyanin in two one-electron steps, as nitrite was reduced to NO. This methaemocyanin could be almost fully regenerated by an anaerobic treatment with HONH2, in contrast with the methaemocyanin prepared with H2O2. A destruction of active sites on treating oxyhaemocyanin with HONH2 explains the partial regeneration of methaemocyanin under air, as traces of H2O2 are formed in the autoxidation of HONH2. The reaction rate of nitrite with deoxyhaemocyanin is almost 15 times that with oxyhaemocyanin. The slope of -1.0 for the logarithm of the pseudo-first-order rate constants plotted against pH indicates that HNO2 is the reacting species. Methaemocyanin was e.p.r.-undetectable, but a binuclear signal was observed at g = 2 on binding nitrite to methaemocyanin. This signal disappeared with a pKa of 6.50, suggesting that a mu-aquo bridging ligand, which can be replaced by nitrite, is deprotonated to a mu-hydroxo bridging ligand, which resists substitution by nitrite. The intensity of this triplet e.p.r. signal allowed the determination of the association constant of nitrite to the active site of Astacus methaemocyanin and yielded a value of 237 M-1 at pH 5.7. The interpretation by some authors of nitrosylhaemocyanin as a nitrite derivative of semimethaemocyanin is contradicted by this rapid reaction of nitrite with copper(I) in deoxyhaemocyanin and in semi-methaemocyanin and by the low binding constant of nitrite to the active site of methaemocyanin. 相似文献

Coffea arabica, the wild ancestor of all commercial Arabica coffee cultivars worldwide, is endemic to the montane rainforests of Ethiopia. These forests, which harbour the most important C. arabica gene pool, are threatened by increasing anthropogenic disturbance, potentially altering the mating patterns, pollen dispersal and maintenance of genetic diversity in C. arabica understorey populations. We genotyped 376 adult coffee shrubs and 418 progenies from three natural unmanaged, and three highly managed coffee populations, using 24 microsatellite markers. Mating system analysis of C. arabica yielded an overall multilocus outcrossing rate of 76%, which contrasts with the common knowledge that C. arabica is a predominantly selfing species. In highly managed coffee populations, paternity could be assigned to 78% of the progenies, whereas in the unmanaged natural coffee populations, only 57% of the progenies could be assigned to a father, indicating reduced long‐distance pollen dispersal in managed forests. Furthermore, the fraction of selfed progenies was significantly higher in managed (23%) than unmanaged (10%) coffee forests. Finally, the lack of spatial genetic structure in all studied populations suggests high seed dispersal in unmanaged populations, and intense berry harvesting and coffee planting in the managed populations. Our results imply that in situ conservation of the wild gene pool of C. arabica must focus on limiting intensification of coffee forest management, as decreased pollen dispersal and increased selfing in C. arabica in intensively managed populations may increase the risk of genetic erosion. © 2014 The Linnean Society of London, Biological Journal of the Linnean Society, 2014, 112 , 76–88. 相似文献

In the mouse, development of the lateral semicircular canal of the inner ear is sensitive to Bmp4 heterozygosity. In the C57BL6 background 30% of the heterozygotes display circling behavior, 66% have a specific defect in the vestibular part of the inner ear, namely the constriction, interruption or absence of the lateral semicircular canal. Only mice having both ears affected display circling behavior. In the (C57BL6xCBA)N1 background, the penetrance of the canal phenotype is greatly reduced, and bilateral lateral canal defect is not sufficient to induce circling. We found association of the canal phenotype with the genotype of markers on chromosome 14 and 4, co-localizing with Ecs and Eclb identified in the Ecl mouse with similar lateral canal defects. Candidate genes to contain the causal mutation are Bmp4 on chromosome 14, and Rere on chromosome 4. 相似文献

This paper deals with the economic viability of using energy maize as a phytoremediation crop in a vast agricultural area moderately contaminated with metals. The acceptance of phytoremediation as a remediation technology is, besides the extraction rate, determined by its profitability, being the effects it has on the income of the farmer whose land is contaminated. This income can be supported by producing renewable energy through anaerobic digestion of energy maize, a crop that takes up only relatively low amounts of metals, but that can be valorised as a feedstock for energy production. The effect on the income per hectare of growing energy maize instead of fodder maize seems positive, given the most likely values of variables and while keeping the basic income stable, originating from dairy cattle farming activities. We propose growing energy maize aiming at risk-reduction, and generating an alternative income for farmers, yet in the long run also generating a gradual reduction of the pollution levels. In this way, remediation is demoted to a secondary objective with sustainable risk-based land use as primary objective. 相似文献

This paper deals with remediation of the Campine soil, an agricultural area diffusely contaminated with metals where most farmers raise dairy cattle and grow fodder maize. In a previous study, we calculated the effect of switching from fodder to energy maize on the farmer's income. Selling this energy maize as feedstock for anaerobic digestion to produce renewable energy could lead to a significant increase in his income. This paper explores the economic opportunities for the farmer of digesting the harvested contaminated biomass himself, by performing a Net Present Value (NPV) analysis on the digestion activity and by calculating the probability of a positive NPV of income resulting from the digestion installation. We investigate the trade off between the maximum price for energy maize that can be paid by the digestion activity and the minimum price that the farming activity needs to compensate for covering its production costs. Integrating the previous study in the current analysis results in an increase of total extra income for the farmer (i.e., from both growing energy maize and performing digestion). 相似文献

The AMP-activated protein kinase (AMPK) and cAMP signaling systems are both key regulators of cellular metabolism. In this study, we show that AMPK activity is attenuated in response to cAMP-elevating agents through modulation of at least two of its alpha subunit phosphorylation sites, viz. alpha-Thr(172) and alpha1-Ser(485)/alpha2-Ser(491), in the clonal beta-cell line INS-1 as well as in mouse embryonic fibroblasts and COS cells. Forskolin, isobutylmethylxanthine, and the glucose-dependent insulinotropic peptide inhibited AMPK activity and reduced phosphorylation of the activation loop alpha-Thr(172) via inhibition of calcium/calmodulin-dependent protein kinase kinase-alpha and -beta, but not LKB1. These agents also enhanced phosphorylation of alpha-Ser(485/491) by the cAMP-dependent protein kinase. AMPK alpha-Ser(485/491) phosphorylation was necessary but not sufficient for inhibition of AMPK activity in response to forskolin/isobutylmethylxanthine. We show that AMPK alpha-Ser(485/491) can be a site for autophosphorylation, which may play a role in limiting AMPK activation in response to energy depletion or other regulators. Thus, our findings not only demonstrate cross-talk between the cAMP/cAMP-dependent protein kinase and AMPK signaling modules, but also describe a novel mechanism by which multisite phosphorylation of AMPK contributes to regulation of its enzyme activity. 相似文献

This study determined the main mite species on rubber trees (clone RRIM-600) in Piracicaba, southeast of S?o Paulo State, from June 2002 to May 2003 and evaluated the possible relation between them. It was conducted in a plantation of 5 ha on 11 year old trees, 15 m high, surrounded with crops as pearl millet, cotton, bean or corn. Samples were taken monthly and consisted of five leaflets, five petioles (only from October 2002 on) and five terminal sections of twigs (10 cm) from 15 rubber trees. All mites of one leaflet, one petiole and one twig section of each plant were mounted for identification to genera/species to estimate the proportional occurrence of each species. A total of 84,850 mites belonging to 38 species of 34 genera and 16 families were found. Tydeidae was the family with the highest number of species (11), followed by Phytoseiidae and Stigmaeidae (4 each). The most abundant families were Eriophyidae, Tenuipalpidae and Tydeidae (totals of 43,023, 26,390 and 13,644 individuals, respectively). The highest population levels of the pest mites Calacarus heveae Feres and Tenuipalpus heveae Baker occurred at the end of the rainy season. The most abundant predators were Metaseiulus camelliae (Chant & Yoshida-Shaul), Amblyseius compositus Denmark & Muma and Euseius citrifolius Denmark & Muma. The predators could not prevent the increase of C. heveae and T. heveae from March on. However, their presence might have prevented an earlier increase and even higher levels of those mites. 相似文献
