The human 8-oxoguanine DNA glycosylase (hOGG1) protein is responsible for initiating base excision DNA repair of the endogenous mutagen 8-oxoguanine. Like nearly all DNA glycosylases, hOGG1 extrudes its substrate from the DNA helix and inserts it into an extrahelical enzyme active site pocket lined with residues that participate in lesion recognition and catalysis. Structural analysis has been performed on mutant versions of hOGG1 having changes in catalytic residues but not on variants having altered 7,8-dihydro-8-oxoguanine (oxoG) contact residues. Here we report high resolution structural analysis of such recognition variants. We found that Ala substitution at residues that contact the phosphate 5' to the lesion (H270A mutation) and its Watson-Crick face (Q315A mutation) simply removed key functionality from the contact interface but otherwise had no effect on structure. Ala substitution at the only residue making an oxoG-specific contact (G42A mutation) introduced torsional stress into the DNA contact surface of hOGG1, but this was overcome by local interactions within the folded protein, indicating that this oxoG recognition motif is "hardwired." Introduction of a side chain intended to sterically obstruct the active site pocket (Q315F mutation) led to two different structures, one of which (Q315F(*149)) has the oxoG lesion in an exosite flanking the active site and the other of which (Q315F(*292)) has the oxoG inserted nearly completely into the lesion recognition pocket. The latter structure offers a view of the latest stage in the base extrusion pathway yet observed, and its lack of catalytic activity demonstrates that the transition state for displacement of the lesion base is geometrically demanding.  相似文献

Hormonal changes accompanying the cold stress (4°C) response that are related to the level of frost tolerance (FT; measured as LT50) and the content of the most abundant dehydrin, WCS120, were compared in the leaves and crowns of the winter wheat (Triticum aestivum L.) cv. Samanta and the spring wheat cv. Sandra. The characteristic feature of the alarm phase (1 day) response was a rapid elevation of abscisic acid (ABA) and an increase of protective proteins (dehydrin WCS120). This response was faster and stronger in winter wheat, where it coincided with the downregulation of bioactive cytokinins and auxin as well as enhanced deactivation of gibberellins, indicating rapid suppression of growth. Next, the ethylene precursor aminocyclopropane carboxylic acid was quickly upregulated. After 3-7 days of cold exposure, plant adaptation to the low temperature was correlated with a decrease in ABA and elevation of growth-promoting hormones (cytokinins, auxin and gibberellins). The content of other stress hormones, i.e., salicylic acid and jasmonic acid, also began to increase. After prolonged cold exposure (21 days), a resistance phase occurred. The winter cultivar exhibited substantially enhanced FT, which was associated with a decline in bioactive cytokinins and auxin. The inability of the spring cultivar to further increase its FT was correlated with maintenance of a relatively higher cytokinin and auxin content, which was achieved during the acclimation period.  相似文献

The influence of zinc(II) propionate on the efficacy of Lactobacillus fermentum against Salmonella enterica serovar Düsseldorf was tested in Japanese quails. Twenty one 3-day old Japanese quails were divided into 3 groups each consisting of 7 birds and inoculated orally: (i) group A (control) with Rogosa broth; (ii) group B with rifampicin-resistant L. fermentum (2.5 × 107 CFU/animal); and (iii) group C with rifampicin-resistant L. fermentum cultured in Rogosa broth supplemented with zinc(II) propionate (1 g Zn2+ /L of broth; i.e. 2.5 × 107 CFU/animal and 0.1 mg Zn2+ /animal). After 16 hours all birds were infected with a single dose of S. enterica serovar Düsseldorf (5.8 × 107 CFU /animal). During the next 6 days the chicks received the same inoculations as they had received earlier through the medium of drinking water. The viable counts of Salmonella, rifampicin-resistant lactobacilli and total aerobes in the feces and cecal content, zinc concentration in the cecal content and growth performance were evaluated. L. fermentum alone, and in combination with Zn(II) propionate significantly reduced shedding of Salmonella in the feces and also the amount of salmonellae present in cecal content as compared to control. The numbers of Salmonella in group C in all collections were lower than in group B, but a significant difference (p < 0.05) was noted only 24 h after infection. The viable counts of rifampicin-resistant lactobacilli were similar in groups B and C and numbers of total aerobes were reduced in these groups compared with control. L. fermentum and its combination with zinc(II) propionate increased daily weight gains in the chicks in comparison with control. Zinc concentration in the control and C group was 34.9 ± 6.2 mg · kg−1 and 676.3 ± 106.6 mg · kg−1 of cecal content, respectively. The flightless bug Pyrrhocoris apterus (L.) is polymorphic for both wing length and flight muscle development. The developed flight muscles of macropterous adults of both sexes first enlarge their volume during the first 5 days after adult emergence, but are then histolyzed in all males and females older than 10 and 14 days, respectively. The flight muscles of brachypterous adult males and females are underdeveloped due to their arrested growth. The total protein content of histolyzed dorsolongitudinal flight muscles from 21-day-old macropterous adults of both sexes is lower than that of developed dorsolongitudinal flight muscles in 5-10-days-old macropterous bugs, but substantially higher than the protein content of underdeveloped dorsolongitudinal flight muscles from adult brachypters. Histolyzed dorsolongitudinal flight muscles differ from the developed ones by decreased quantities of 18 electrophoretically separated proteins. Histolysis of developed dorsolongitudinal flight muscles is accompanied by significant decreases in citrate synthase, glyceraldehyde-3-phosphate dehydrogenase and β-hydroxyacyl-CoA dehydrogenase enzyme activities and an increase in alanine aminotransferase activity, and can be precociously induced by application of a juvenile hormone analogue. This is the first report of flight muscle polymorphism, histolysis of developed flight muscles and its endocrine control in insects displaying non-functional wing polymorphism.  相似文献

A synthesis of the alliance Eragrostion cilianensi-minoris in the Czech Republic is presented on the basis of 82 relevés including new unpublished data. A TWINSPAN classification and detrended correspondence analysis were used to identify the main vegetation types included in the alliance Eragrostion cilianensi-minoris. A syntaxonomic revision of the data set revealed five associations of the alliance: Digitario sanguinalis-Eragrostietum minoris, Portulacetum oleraceae, Eragrostio poaeoidis-Panicetum capillaris, Cynodontetum dactyli, and Hibisco trioni-Eragrostietum poaeoidis. The latter was recently found in several arable fields in Southern Moravia (Czech Republic) and was newly characterized.  相似文献

Cytokinins (CKs) are plant hormones affecting numerous developmental processes. Zeatin and its derivatives are the most important group of isoprenoid CKs. Zeatin occurs as two isomers: while trans-zeatin (transZ) was found to be a bioactive substance, cis-zeatin (cisZ) was reported to have a weak biological impact. Even though cisZ derivatives are abundant in various plant materials their biological role is still unknown. The comprehensive screen of land plants presented here suggests that cisZ-type CKs occur ubiquitously in the plant kingdom but their abundance might correlate with a strategy of life rather than with evolutionary complexity. Changing levels of transZ and cisZ during Arabidopsis ontogenesis show that levels of the two zeatin isomers can differ significantly during the life span of the plant, with cisZ-type CKs prevalent in the developmental stages associated with limited growth. A survey of the bioassays employed illustrates mild activity of cisZ and its derivatives. No cis?trans isomerization, which would account for the effects of cisZ, was observed in tobacco cells and oat leaves. Differences in uptake between the two isomers resulting in distinct bioactivity have not been detected. In contrast, cisZ and transZ have a different metabolic fate in oat and tobacco. Analysis of a CK-degrading enzyme, cytokinin oxidase/dehydrogenase (CKX), reveals that Arabidopsis possesses two isoforms, AtCKX1 expressed in stages of active growth, and AtCKX7, both of which have the highest affinity for the cisZ isomer. Based on the present results, the conceivable function of cisZ-type CKs as delicate regulators of CK responses in plants under growth-limiting conditions is hypothesized.  相似文献
