Targeting of ribosome-nascent chain complexes to the translocon in the endoplasmic reticulum is mediated by the concerted action of the signal recognition particle (SRP) and the SRP receptor (SR). Ribosome-stripped microsomes were digested with proteases to sever cytoplasmic domains of SRalpha, SRbeta, TRAM, and the Sec61 complex. We characterized protein translocation intermediates that accumulate when Sec61alpha or SRbeta is inactivated by proteolysis. In the absence of a functional Sec61 complex, dissociation of SRP54 from the signal sequence is blocked. Experiments using SR proteoliposomes confirmed the assembly of a membrane-bound posttargeting intermediate. These results strongly suggest that the Sec61 complex regulates the GTP hydrolysis cycle of the SRP-SR complex at the stage of signal sequence dissociation from SRP54.  相似文献

We have cloned and characterized the entire DNA polymerase gene and flanking regions from Kaposi's sarcoma-associated herpesvirus (KSHV) and two closely related macaque homologs of KSHV, retroperitoneal fibromatosis-associated herpesvirus-Macaca nemestrina (RFHVMn) and -Macaca mulatta (RFHVMm). We have also identified and partially characterized the corresponding genomic region of another KSHV-like herpesvirus, provisionally named "M. nemestrina rhadinovirus type 2 (MneRV-2)," with close similarity to rhesus rhadinovirus (RRV). A sequence comparison of these four macaque viruses and two KSHV-like gammaherpesviruses recently identified in African green monkeys, Chlorocebus rhadinovirus types 1 and 2 (ChRV-1 and ChRV-2) reveals the presence of two distinct lineages of KSHV-like rhadinoviruses in Old World primates. The first rhadinovirus lineage consists of KSHV and its closely related homologs RFHVMn, RFHVMm, and ChRV-1, while the second more distantly related lineage consists of RRV, MneRV-2, and ChRV-2. Our findings raise the possibility of the existence of another human KSHV-like herpesvirus belonging to the second rhadinovirus lineage.  相似文献

Signal transduction within the nucleus by mitogen-activated protein kinase.   总被引:30,自引:0,他引:30  
The nucleus is an important target of signal transduction by growth factor receptors that stimulate mitogen-activated protein (MAP) kinases. We tested the hypothesis that MAP kinases have a signaling role within the nucleus by examining the effect of the expression of a human MAP kinase isoform (p41mapk) in tissue culture cells. The expressed p41mapk was found to be localized in both the cytoplasmic and nuclear compartments of the cells. Significantly, the expression of p41mapk caused an increase in the phosphorylation of a nuclear substrate: Ser62 of c-Myc. Phosphorylation at Ser62 stimulated the activity of the NH2-terminal transactivation domain of c-Myc. Thus, p41mapk causes the phosphorylation and regulation of a physiologically significant nuclear target of signal transduction. These data establish that at least one MAP kinase isoform has a nuclear role during signal transduction.  相似文献

Background

Obtaining a draft genome sequence of the zebra finch (Taeniopygia guttata), the second bird genome to be sequenced, provides the necessary resource for whole-genome comparative analysis of gene sequence evolution in a non-mammalian vertebrate lineage. To analyze basic molecular evolutionary processes during avian evolution, and to contrast these with the situation in mammals, we aligned the protein-coding sequences of 8,384 1:1 orthologs of chicken, zebra finch, a lizard and three mammalian species.

Results

We found clear differences in the substitution rate at fourfold degenerate sites, being lowest in the ancestral bird lineage, intermediate in the chicken lineage and highest in the zebra finch lineage, possibly reflecting differences in generation time. We identified positively selected and/or rapidly evolving genes in avian lineages and found an over-representation of several functional classes, including anion transporter activity, calcium ion binding, cell adhesion and microtubule cytoskeleton.

Conclusions

Focusing specifically on genes of neurological interest and genes differentially expressed in the unique vocal control nuclei of the songbird brain, we find a number of positively selected genes, including synaptic receptors. We found no evidence that selection for beneficial alleles is more efficient in regions of high recombination; in fact, there was a weak yet significant negative correlation between ω and recombination rate, which is in the direction predicted by the Hill-Robertson effect if slightly deleterious mutations contribute to protein evolution. These findings set the stage for studies of functional genetics of avian genes.  相似文献

Zusammenfassung Die Mosaikhypothese (Papi et al.) und die Gradientenhypothese (Wallraff) der olfaktorischen Navigation berücksichtigen nicht die grundlegenden meteorologischen Bedingungen für die Ausbreitung atmosphärischer Spurenstoffe. Die Turbulenz der Atmosphäre und die Abbauprozesse der geeigneten Duftstoffe setzen der olfaktorischen Navigation enge Grenzen. Eine eindeutige Korrelation zwischen bestimmten Duftstoffen und den vorherrschenden Windrichtungen, wie sie der Mosaikhypothese zugrunde liegt, ist aufgrund der Abweichungen der realen Strömungsverhältnisse vom mittleren großräumigen Wind und der Instabilität der verschiedenen Duftquellen nicht möglich. Olfaktorisch wahrnehmbare Gradienten hängen von der Verweildauer geeigneter Spurenstoffe in der Atmosphäre ab. Langlebige Spurenstoffe besitzen im allgemeinen keinen deutlichen Gradienten, so daß im Bereich von einigen hundert Kilometern keine meßbaren Konzentrationsunterschiede festgestellt werden können. Kurzlebige atmosphärische Spurenstoffe besitzen zwar stärkere Gradienten, diese sind aber räumlich und zeitlich variabel und können für die olfaktorische Navigation nicht genutzt werden.
Meteorological conditions and hypotheses of olfactory navigation
Summary The mosaic hypothesis (Papi et al.) and the gradient hypothesis (Wallraff) of olfactory navigation don't consider the basic meteorological conditions of the dispersion of atmospheric substances. The turbulence of the atmosphere and the removal processes of suitable odorous substances limit strongly any kind of olfactory navigation. A significant correlation between certain odorous substances and the prevailing wind directions assumed by the mosaic hypothesis is not possible because of the differences between the actual air currents and the mean wind directions, and the instability of the different odorous sources. The realisation of olfactory gradients is depending on the residence times of the suitable substances. Long living substances generally don't have any significant gradients, and differences in concentration cannot be detected within distances of some hundred kilometers. Short living substances have strong gradients but varying in time and space, and cannot be used for olfactory navigation.

Due to its strong gradient in salinity and small temperature gradient the Mediterranean provides an ideal setting to study the impact of salinity on the incorporation of Mg into foraminiferal tests. We have investigated tests of Globorotalia inflata and Globigerina bulloides in plankton tow and core top samples from the Western Mediterranean using ICP-OES for bulk analyses and LA-ICP-MS for analyses of individual chambers in single specimens. Mg/Ca observed in G. inflata are consistent with existing calibrations, whereas G. bulloides had significantly higher Mg/Ca than predicted, particularly in core top samples from the easterly stations. Scanning Electron Microscopy and Laser Ablation ICP-MS revealed secondary overgrowths on some tests, which could explain the observed high core top Mg/Ca. We suggest that the Mediterranean intermediate and deep water supersaturated with respect to calcite cause these overgrowths and therefore increased bulk Mg/Ca. However, the different species are influenced by diagenesis to different degrees probably due to different test morphologies. Our results provide new perspectives on reported anomalously high Mg/Ca in sedimentary foraminifera and the applicability of the Mg/Ca paleothermometry in high salinity settings, by showing that (1) part of the signal is generated by precipitation of inorganic calcite on the foraminifer test due to increased calcite saturation state of the water and (2) species with high surface-to-volume shell surfaces are potentially more affected by secondary Mg-rich calcite encrustation.  相似文献
