RNA interference technology allows the systematic genetic analysis of the molecular alterations in cancer cells and how these alterations affect response to therapies. Here we used small interfering RNA (siRNA) screens to identify genes that enhance the cytotoxicity (enhancers) of established anticancer chemotherapeutics. Hits identified in drug enhancer screens of cisplatin, gemcitabine, and paclitaxel were largely unique to the drug being tested and could be linked to the drug's mechanism of action. Hits identified by screening of a genome-scale siRNA library for cisplatin enhancers in TP53-deficient HeLa cells were significantly enriched for genes with annotated functions in DNA damage repair as well as poorly characterized genes likely having novel functions in this process. We followed up on a subset of the hits from the cisplatin enhancer screen and validated a number of enhancers whose products interact with BRCA1 and/or BRCA2. TP53(+/-) matched-pair cell lines were used to determine if knockdown of BRCA1, BRCA2, or validated hits that associate with BRCA1 and BRCA2 selectively enhances cisplatin cytotoxicity in TP53-deficient cells. Silencing of BRCA1, BRCA2, or BRCA1/2-associated genes enhanced cisplatin cytotoxicity approximately 4- to 7-fold more in TP53-deficient cells than in matched TP53 wild-type cells. Thus, tumor cells having disruptions in BRCA1/2 network genes and TP53 together are more sensitive to cisplatin than cells with either disruption alone.  相似文献

BACKGROUND/AIMS: Turner's syndrome (TS) is associated with increased insulin resistance and adiposity, which might be associated with type 2 diabetes in later life. We aimed to determine whether the defect in insulin sensitivity is a primary intrinsic defect in TS or dependent on variation in body composition. METHODS: Sixteen women with TS not on growth hormone replacement but receiving oestrogen replacement therapy [age (mean +/- SD): 30.2 +/- 8.5 years; height-corrected fat-free mass: 26.1 +/- 3.1 kg/height] and a control group of 16 normal healthy women (age: 30.1 +/- 8.2 years; height-corrected fat-free mass: 25.9 +/- 2.4 kg/height) were studied. Fasting blood samples were obtained for measurement of glucose, insulin, IGF-I, IGFBP-1, IGFBP-3 and lipid levels. The hyperinsulinaemic euglycaemic clamp was performed to assess peripheral insulin sensitivity (M value), and the Homeostasis Model Assessment (HOMA-S) was used to estimate fasting insulin sensitivity. Body composition was assessed using a dual-energy X-ray absorptiometry scan. RESULTS: Fasting insulin sensitivity (HOMA-S 103.2 +/- 78.6 vs. 193.9 +/- 93.5, p = 0.006) was lower in TS subjects compared to controls as was whole-body insulin sensitivity (M value 2.9 +/- 1.9 vs. 5.5 +/- 2.6 mg/kg/min, p = 0.003). In a multiple regression analysis the Turner karyotype was significantly related to insulin sensitivity (p = 0.008) independent of any differences in fat-free mass and percent whole-body fat mass. CONCLUSION: The increased insulin resistance in women with TS is independent of measures of body composition and may represent an intrinsic defect related to their chromosomal abnormality.  相似文献

Non-viral vectors are promising vehicles for gene therapy but delivery of plasmid DNA to post-mitotic cells is challenging as nuclear entry is particularly inefficient. We have developed and evaluated a hybrid mRNA/DNA system designed to bypass the nuclear barrier to transfection and facilitate cytoplasmic gene expression. This system, based on co-delivery of mRNA(A64) encoding for T7 RNA polymerase (T7 RNAP) with a T7-driven plasmid, produced between 10- and 2200-fold higher gene expression in primary dorsal root ganglion neuronal (DRGN) cultures isolated from Sprague–Dawley rats compared to a cytomegalovirus (CMV)-driven plasmid, and 30-fold greater expression than the enhanced T7-based autogene plasmid pR011. Cell-free assays and in vitro transfections highlighted the versatility of this system with small quantities of T7 RNAP mRNA required to mediate expression at levels that were significantly greater than with the T7-driven plasmid alone or supplemented with T7 RNAP protein. We have also characterized a number of parameters, such as mRNA structure, intracellular stability and persistence of each nucleic acid component that represent important factors in determining the transfection efficiency of this hybrid expression system. The results from this study demonstrate that co-delivery of mRNA is a promising strategy to yield increased expression with plasmid DNA, and represents an important step towards improving the capability of non-viral vectors to mediate efficient gene transfer in cell types, such as in DRGN, where the nuclear membrane is a significant barrier to transfection.  相似文献

Exposing pepper ( Capsicum annuum ) plants to extremely high day temperatures (HDT) (day/night temperatures of 36 ± 2/10 ± 2°C), obtained by keeping the greenhouse closed during the day to exploit solar heating, prevented the development of low night temperature (LNT) symptoms. Plants of cultivars Fiesta and Selica grown under LNTs (10 ± 2°C) and moderate day temperatures (25 ± 2°C) during winter exhibited retarded growth, reduced leaf numbers, and deformed fruits with few or no seeds. LNT caused a reduction in the number and quality of pollen grains: the reduction in pollen quality was associated with reduced starch accumulation in pollen grains at 3 days before anthesis (DBA) and a decrease of more than two-fold in total soluble sugars in the mature pollen grains. This inhibitory effect was associated with more than 50% reduction in the enzymatic activities of the cell wall-bound and soluble acid invertases that catalyze the hydrolysis of incoming sucrose molecules. All these symptoms were prevented by HDT treatment which matched the vegetative and reproductive performance of the plants to those of plants grown under optimal night temperature (ONT) conditions (day/night temperatures of 23 ± 2/18 ± 2°C). HDT also prevented the inhibitory effect of LNT on enzymatic activities of both invertases in pollen at 5 DBA and brought about the accumulation of high levels of starch in pollen at 3 DBA. The results presented could support the development of a novel procedure for producing greenhouse crops with minimum or even with no fuel consumption for heating during the winter nights in regions with bright and sunny days.  相似文献

Mutants with defects in the cytochrome (cyt) b6/f complex were analyzed for their effect on the expression of a subgroup of nuclear genes encoding plastid-localized enzymes participating in chlorophyll biosynthesis. Their defects ranged from complete loss of the cytb6/f complex to point mutations affecting specifically the quinone-binding QO site. In these seven mutants, light induction of the tetrapyrrole biosynthetic genes was either abolished or strongly reduced. In contrast, a normal induction of chlorophyll biosynthesis genes was observed in mutants with defects in photosystem II, photosystem I, or plastocyanin, or in wild-type cells treated with 3-(3'4'-dichlorophenyl)-1,1-dimethylurea or 2,5-dibromo-3-methyl-6-isopropyl benzoquinone. We conclude that the redox state of the plastoquinone pool does not control light induction of these chlorophyll biosynthetic genes. The signal that affects expression of the nuclear genes appears to solely depend on the integrity of the cytb6/f complex QO site. Since light induction of these genes in Chlamydomonas has recently been shown to involve the blue light receptor phototropin, the results suggest that cytb6/f activity regulates a plastid-derived factor required for their expression. This signaling pathway differs from that which regulates state transitions, since mutant stt7, lacking a protein kinase involved in phosphorylation of the light-harvesting complex II, was not altered in the expression of the chlorophyll biosynthetic genes.  相似文献

To investigate the evolution and geographical origins of hexaploid wheat, we examined a 284 bp sequence from the promoter region of the GluDy locus, coding for the y subunit of high-molecular-weight glutenin. Fourteen different alleles were found in 100 accessions of Aegilops tauschii and 169 of Triticum aestivum. Two alleles were present in both species; the other 7 alleles from Ae. tauschii and 5 from T. aestivum were unique to their respective species. The two shared alleles differed at only one nucleotide position within the region sequenced, but their apparent association with the common haplotypes GluD1a and GluD1d, which have substantial differences within their GluDy coding regions, makes it unlikely that the alleles evolved independently in Ae. tauschii and T. aestivum. The results therefore support previous studies which suggest that there were at least two Ae. tauschii sources that contributed germplasm to the D genome of T. aestivum. The number of alleles present in T. aestivum, and the nucleotide diversity of these alleles, indicates that this region of the D genome has undergone relatively rapid change since polyploidisation. Ae. tauschii from Syria and Turkey had relatively high nucleotide diversity and possessed all the major GluDy alleles, indicating that these populations are probably ancient and not the result of adventive spread. The presence in the Turkish population of both of the shared alleles suggests that hexaploid wheat is likely to have originated in southeast Turkey or northern Syria, within the Fertile Crescent and near to the farming villages at which archaeological remains of hexaploid wheats are first found. A second, more recent, hexaploidisation probably occurred in Iran.  相似文献

While banding ferruginous pygmy-owls (Glaucidium brasilianum) and Eastern screech-owls (Megascops asio) in south Texas during 2004, we recorded Philornis mimicola (Diptera: Muscidae) and Ornithodoros concanensis (Acari: Argasidae) parasitizing nestlings. Inspection of nestlings revealed 54 P. mimicola and one O. concanensis. Inspection of nest material revealed 111 P. mimicola, including 57 puparia. The effect (e.g., blood loss, anemia) of these hematophagous parasites might have contributed to the demise of at least one Eastern screech-owl nestling. This is the first record of P. mimicola and O. concanensis parasitizing ferruginous pygmy-owls and Eastern screech-owls.  相似文献

The dynamics of the Hindmarsh-Rose (HR) model of bursting thalamic neurons is reduced to a system of two linear differential equations that retains the subthreshold resonance properties of the HR model. Introducing a reset mechanism after a threshold crossing, we turn this system into a resonant integrate-and-fire (RIF) model. Using Monte-Carlo simulations and mathematical analysis, we examine the effects of noise and the subthreshold dynamic properties of the RIF model on the occurrence of coherence resonance (CR). Synchronized burst firing occurs in a network of such model neurons with excitatory pulse-coupling. The coherence level of the network oscillations shows a stochastic resonance-like dependence on the noise level. Stochastic analysis of the equations shows that the slow recovery from the spike-induced inhibition is crucial in determining the frequencies of the CR and the subthreshold resonance in the original HR model. In this particular type of CR, the oscillation frequency strongly depends on the intrinsic time scales but changes little with the noise intensity. We give analytical quantities to describe this CR mechanism and illustrate its influence on the emerging network oscillations. We discuss the profound physiological roles this kind of CR may have in information processing in neurons possessing a subthreshold resonant frequency and in generating synchronized network oscillations with a frequency that is determined by intrinsic properties of the neurons. Research scientists and IACUC members are faced with the difficult task of balancing the necessity of using animals for experimental research and their mandate to protect the welfare of those animals used in that research. One way to reduce the number of research animals would be to reuse them, but the regulations do not specifically address this topic. To learn more about the reuse of research animals, the authors conducted an online survey of animal facilities involved in preclinical studies.
