The relative biological availability (bioavailability) of three brands of ampicillin trihydrate capsules marketed in Canada was studied by means of a crossover experimental design and was evaluated by analysis of variance. After administration of a single oral dose (500 mg.) to volunteer subjects, the mean peak ampicillin serum concentration and the area under the time ampicillin serum concentration curve (AUC) were higher for one of the products than for the other two. A second study utilizing the same experimental design revealed no differences in these parameters when different production lots of the more available product were compared. Product bioavailability was not related to the in vitro dissolution time of the capsules. The experimental design and methods of statistical analysis are discussed. 相似文献

Rats were exposed to protons (155 MeV) or to helium (165 MeV/amu), neon (522 MeV/amu) or argon (670 MeV/amu) particles to evaluate the behavioral toxicity of these types of radiations. Behavioral toxicity was assessed using the conditioned taste aversion paradigm. Exposure to all types of radiation produced dose-dependent increases in the intensity of the acquired taste aversion. However, the intensity of the aversions, measured as the dose that produced a 50% decrease in the intake of the sucrose-conditioned stimulus, did not show significant variation as a function of the linear energy transfer (LET) of the radiation. Proliferation of normal (not immunized intentionally) murine spleen cells was elicited with concanavalin A, supernatant fluid from cultures of EL-4 cells, human recombinant interleukin 2 (IL-2), or a mixture of phorbol ester and calcium ionophore A23187. IL-2-induced proliferation was inhibited by membrane-permeable dibutyryl cyclic adenosine monophosphate (cAMP) or by the adenylate cyclase activator forskolin. Consistent with these observations was the finding that stimulation with IL-2 decreased and forskolin increased the intracellular content of cAMP. IL-2-induced proliferation, as well as that induced by concanavalin A or phorbol-ionophore mixture, was inhibited by monoclonal antibodies specific for L3T4 or Lyt-2 cell surface markers. This inhibition was observed even when antibodies were added several hours after exposure of cells to IL-2. Notably, antibodies did not alter the intracellular content of cAMP. Thus, the experimental data failed to establish a functional linkage between the inhibitory effect of antibodies and the regulatory effect of the adenylate cyclase system. However, our results provide a rational basis for the postulation that antibodies, upon binding to their corresponding ligands, generate a negative signal that interferes with IL-2-induced proliferation. Therefore, L3T4 and Lyt-2 molecules appear to play an important role in the regulation of lymphocyte proliferation. 相似文献

There is growing concern about mitigation-driven translocations that move animals from anthropogenic threats at donor sites because of their failure rate and lack of application of scientific principles and best practice. We reviewed all known lizard translocations in New Zealand between 1988 and 2013 and identified 85 translocations of 30 lizard taxa to 46 release sites. Most translocations (62%) were motivated by conservation goals for the species or the release site, and one-third were mitigation-driven translocations, typically motivated by habitat loss due to development. Mitigation-driven translocations began in 2003, and since that time have equalled the number of conservation-motivated translocations. Conservation-motivated translocations usually released lizards on islands without mammalian predators, whereas mitigation-driven translocations usually relocated lizards to mainland sites with introduced predators. Long-term monitoring has been sparse and often rudimentary. Eight lizard translocations have recorded population growth, including one mitigation-driven translocation that was into a fenced reserve. Research on commonly used management techniques to mitigate human-related impacts is recommended to establish whether these techniques benefit lizards in the long term. 相似文献

Seven captive-born lemurs (Lemur fulvus) at the Duke University Primate Center were presented with leaves of different maturity from five species of North Carolina trees. The animals demonstrated three distinct behaviors toward the novel plant material. They sniffed, tasted, and/or ingested it. New leaves were sniffed, but little tasting and ingestion was observed. Intermediate pine needles were sniffed and ingested but little tasted. Mature leaves were sniffed equally, but the mature leaves of tulip trees and honeysuckle were tasted significantly more than pine, sweetgum, and red maple. Pine, sweetgum, and red maple were ingested significantly more than tulip trees and honeysuckle. Male lemurs ate significantly more mature pine needles and new sweetgum leaves than did the females. Chemical analysis of these plant materials indicated that the new and mature leaves of tulip tree and honeysuckle contained alkaloids. Captive-born lemurs apparently use their sense of smell and taste in choosing what to eat and seem just as capable as free-ranging animals in finding food when faced with the chemical defenses that protect trees from insect predation. 相似文献

B cell stimulatory factor-1 (BSF-1) stimulates resting B cells to increase in volume and prepares these cells to enter the S phase in response to anti-IgM and other B cell mitogens. Interferon-gamma (IFN-gamma) blocks both the volume enlargement and preparation for DNA synthesis caused by BSF-1, although it has little effect on B cells already stimulated by BSF-1. The capacity of IFN-gamma to inhibit the action of BSF-1 on resting B cells suggests a mutual regulatory interaction between these two T cell-derived products. 相似文献
