Genes for seven putative serine proteases (ChpA–ChpG) belonging to the trypsin subfamily and homologous to the virulence factor pat-1 were identified on the chromosome of Clavibacter michiganensis subsp. michiganensis ( Cmm ) NCPPB382. All proteases have signal peptides indicating export of these proteins. Their putative function is suggested by two motifs and an aspartate residue typical for serine proteases. Furthermore, six cysteine residues are located at conserved positions. The genes are clustered in a chromosomal region of about 50 kb with a significantly lower G + C content than common for Cmm . The genes chpA , chpB and chpD are pseudogenes as they contain frame shifts and/or in-frame stop codons. The genes chpC and chpG were inactivated by the insertion of an antibiotic resistance cassette. The chpG mutant was not impaired in virulence. However, in planta the titre of the chpC mutant was drastically reduced and only weak disease symptoms were observed. Complementation of the chpC mutant by the wild-type allele restored full virulence. ChpC is the first chromosomal gene of Cmm identified so far that affects the interaction of the pathogen with the host plant.  相似文献

Sucrose Leakage from Isolated Parenchyma of Sugar Beet Roots   总被引:1,自引:0,他引:1  
The kinetics of sugar efflux from slices of sugar beet rootswas investigated using washing solutions of different osmoticpressure and calcium concentration. The leakage of sucrose isstrongly reduced in solutions of high osmotic pressure (>0·8MPa) or high calcium concentration (10 mM). Turgor-dependentnecrosis of parenchyma cells (plasmoptysis) is the main causeof sucrose efflux from the tissue in hypotonic media with lowcalcium activity. This was shown by good correlation betweenthe percentage of leaked sucrose and the percentage of tissuewater, which was in the free space after the washing procedure.The kinetics of sugar leakage from beet root parenchyma is nobasis for the estimation of the sugar contents of the free spaceor the cytoplasm in situ.  相似文献

AFTER infection of monkey kidney cells with simian virus 40 (SV40), several species of SV40 specific RNA are synthesized1. Most SV40 RNA have a molecular weight of about 6×105 and 8×105 as measured by polyacrylamide gel electrophoresis1. In addition to these classes of RNA, a large heterogeneous SV40 specific RNA species of up to three times the length of the monomeric SV40 DNA molecule has been observed1–4. Submission in the wolf and dog is defined on the basis ot itsmotivation: submission is the effort of the inferior to attainfriendly or harmonic social integration. Submission functions as an appeal or a contribution to socialintegration, but only if it meets a corresponding attitude inthe superior. The form of submissive behavior in wolf and dogis ritualized and symbolized cub-behavior. Two main forms ofsubmissive behavior occur in wolf and dog: active submission,derived from begging for milk or food, and passive submission,derived from the posture which the cub adopts when cleaned byits mother. The definition of submission is generally applicable to vertebratesliving in groups based on intimacy and a social hierarchicalorder. The concept of submission as the role of the defeatedin the terminal phase of fight with the function to inhibitautomatically aggression in the superior should be dismissed.In vertebrates at least three types of conflict with differentterminal phases occur: (1). Severe fight based on intolerance;ends with flight by the inferior or with his death. (2). Ritualizedfight over a privilege; ends with the "giving-up-the-claim ritual"of the inferior, which automatically blocks the aggression ofthe superior. (3). Minor conflict in closed groups; settledby submissive behavior of the inferior. In closed vertebrate groups, intermediate forms between (1)and (3) occur, depending on the proportion between activatedintimacy and intolerance.  相似文献

Evidence from vegetative anatomy, reproductive morphology, and palynology does not support a relationship of Oceanopapaver with Cistaceae, Cruciferae, Flacourtiaceae, Papaveraceae, and Tiliaceae, but suggests placement of the genus in Capparaceae. The apparent occurrence of myrosin cells, among other features, effectively excludes all of the aforenoted families except Capparaceae and Cruciferae. However, multicellular non-glandular trichomes, bracteate inflorescences, sepals and petals each occasionally other than four per flower, presence of an androgynophore, numerous stamens, tricolporate and binucleate pollen, the unilocular mature ovary, the stipitate fruit, and the exotegmic seed in Oceanopapaver favour Capparaceae over Cruciferae. Floral histology and vasculature provide no clues about the relationships of Oceanopapaver. A few features are anomalous, most notably the presence of secretory canals and secretory cells in the genus versus their absence in Capparaceae and their rarity in Cruciferae, the trichomic floral nectary in the genus versus the massive, non-trichomic nectaries in these two families, and the straight embryo in the genus versus the more or less curved or folded embryo in the two families. The fleshy endosperm in Oceanopapaver has counterparts in a few Capparaceae, contrary to previous claims that endosperm is absent or scanty in this family. The report of stamen fascicle traces for Oceanopapaver is the first for Capparaceae, but these should be sought elsewhere in the family. Within Capparaceae the genus fits best in Capparoideae compared to Cleomoideae or the nine other very restrictive subfamilies variously proposed for Capparaceae. There is no justification for the monotypic segregate Oceanopapaveraceae. The phylogenetic and functional anatomy of vegetative and reproductive structures is discussed.  相似文献

Interdiffusion coefficients of K and Na for tangential transport through cell walls were found to be independent of ion strength and only moderately (c. 50%) lower than values known for diffusion in water. Although at low salt concentrations alkali ion exchange diffusion on the cell wall pathway is facilitated by accumulation of diffusing species in the Donnan space, at all concentrations the resistance of the whole tissue for apoplastic cation exchange diffusion remained significantly higher than that of unstirred liquid of equal thickness, as the area fraction of the cell wall pathway in the parenchyma is small.  相似文献

Members of the megadiverse insect order Diptera (flies) have successfully colonized all continents and nearly all habitats. There are more than 154 000 described fly species, representing 10–12% of animal species. Elucidating the phylogenetic relationships of such a large component of global biodiversity is challenging, but significant advances have been made in the last few decades. Since Hennig first discussed the monophyly of major groupings, Diptera has attracted much study, but most researchers have used non‐numerical qualitative methods to assess morphological data. More recently, quantitative phylogenetic methods have been used on both morphological and molecular data. All previous quantitative morphological studies addressed narrower phylogenetic problems, often below the suborder or infraorder level. Here we present the first numerical analysis of phylogenetic relationships of the entire order using a comprehensive morphological character matrix. We scored 371 external and internal morphological characters from larvae, pupae and adults for 42 species, representing all infraorders selected from 42 families. Almost all characters were obtained from previous studies but required revision for this ordinal‐level study, with homology assessed beyond their original formulation and across all infraorders. We found significant support for many major clades (including the Diptera, Culicomorpha, Bibionomorpha, Brachycera, Eremoneura, Cyclorrhapha, Schizophora, Calyptratae and Oestroidea) and we summarize the character evidence for these groups. We found low levels of support for relationships between the infraorders of lower Diptera, lower Brachycera and major lineages of lower Cyclorrhapha, and this is consistent with findings from molecular studies. These poorly supported areas of the tree may be due to periods of rapid radiation that left few synapomorphies in surviving lineages.  相似文献

1. We aimed to demonstrate reproducible nutrition and growth of macrophytes in non‐axenic laboratory cultures preventing growth of phytoplankton and epiphytes. 2. Macrophyte shoot segments were planted in a mixture of commercial acid‐washed silica sand with crystalline tricalcium phosphate, and this artificial sediment was covered with a layer of pure silica sand. The liquid mineral media used did not contain phosphorus but were rich in all other nutrient elements. A CO2 reservoir provided sustainable CO2 supply to macrophyte cultures by gas diffusion through a polyethylene membrane. 3. Chara hispida, Chara tomentosa, Chara baltica, Elodea canadensis, Potamogeton pectinatus and Zanichellia palustris could be cultivated for long term without medium exchange and aeration. Microalgae growth was prevented by the absence of phosphate in the water column. Mobilisation of tricalcium phosphate and phosphate uptake by the rhizoids of C. hispida enabled sustainable rapid shoot growth and increased the concentration of inorganic phosphate in the shoot dry weight by five to six times in comparison with plants cultivated on pure silica sand. A significant growth support from tricalcium phosphate was also observed for E. canadensis, but the rate of phosphate uptake by the roots was not sufficient to maintain a storage pool of inorganic phosphate (Pi) in the growing shoots of this plant. 4. Membrane‐controlled CO2 supply from a reservoir and artificial sediments like the one described provide attractive options for the laboratory culture of macrophytes.  相似文献
