Field surveys over 2 years in contiguous beds of the seagrassesZostera capricorni andPosidonia australis showed that the green algaCodium duthieae was consistently more abundant inZ. capricorni than inP. australis. In 1 year, mature plants were also more abundant at the boundary between the seagrass beds than in either bed. Field experiments and programmes of sampling were used to investigate three potential explanations for the unusual distribution of this alga: (1) that the availability of substrata suitable for attachment of the alga differed between the two seagrass beds; (2) that mortality of matureC. duthieae differed between the seagrass beds; and/or (3) that the intensity of recruitment was different in the two seagrass beds.C. duthieae plants were exclusively epibionts of the bivalveAnadara trapezia. Detailed sampling showed that the abundance ofA. trapezia was similar in both seagrass beds and that the distribution of bivalves suitable as substrata forC. duthieae plants was not obviously related to proximity to the boundary between the beds. Two experiments investigated the survival ofC. duthieae plants in each bed. In the first, matureC. duthieae plants transplanted into theP. australis bed suffered similar rates of mortality to plants which were disturbed and moved within theZ. capricorni bed or which were left undisturbed in theZ. capricorni bed. Fewer of the host bivalves were recovered from theZ. capricorni bed, however, indicating that the mechanism of mortality differed between the beds, hosts being more frequently dislodged in theZ. capricorni bed. Removal of the leaves of the seagrasses had consistently greater effects on near-bottom current velocities in theZ. capricorni bed than in theP. australis bed and significantly increased mortality ofC. duthieae in theZ. capricorni bed. Survival of plants was greater in plots of artificial leaves ofP. australis placed in theZ. capricorni bed than in plots of the naturalZ. capricorni leaves or plots where the natural leaves were removed. Most mortality in theZ. capricorni bed was due to dislodgement of the alga and its bivalve substratum. Corresponding manipulations of leaves in theP. australis bed had consistently smaller effects on survivorship of both the alga and its host. Patterns in the recruitment of the alga most clearly reflected the distribution of adults.C. duthieae recruits were 5 times more abundant in theZ. capricorni bed and at the boundary between the two beds than in theP. australis bed. The results demonstrate how habitat structure, provided by the canopy of leaves of the two species of seagrass, can have contrasting effects on the recruitment and mortality of a macroalga. In the case ofC. duthieae, it appears that the differential pattern of recruitment is the primary determinant of the distribution of adult plants.  相似文献

A 4 kb fragment of chromosomal DNA was cloned from a clinical strain of methicillin-resistant Staphylococcus aureus. It comprises part of a section of the chromosome that was lost when the strain was cured of resistance to methicillin and to other antimicrobial agents. The fragment mediates an increased level of methicillin resistance when inserted into a shuttle vector and transformed back into the sensitive strain generated when the original DNA was deleted.  相似文献

Sections of a cloned 27 kb segment of chromosomal DNA, associated with resistance to four antimicrobial agents in a clinical isolate of methicillin-resistant Staphylococcus aureus (MRSA), were tested for their ability to determine resistance when transformed into a sensitive laboratory strain of S. aureus. This was achieved by inserting the sections into a newly constructed shuttle vector, amplifying the recombinant DNA in E. coli, and transforming protoplasts of the sensitive S. aureus strain. Two sections of the cloned DNA were found to determine resistance separately to mercuric ion and to tetracycline, in both S. aureus and Escherichia coli.  相似文献

Summary The responses to pure tone stimuli of individual auditory receptors in the locustValanga irregularis were quantified and the location of receptors within the auditory organ identified through the intracellular injection of Lucifer Yellow. Three groups of receptors were identified and classified according to their site of attachment to the tympanic membrane (Gray 1960). Group a cells were attached to the elevated process and were tuned to sound frequencies from 4 to 6 kHz (Fig. 2). Group c cells were attached to the folded body and were tuned to sound frequencies from 2 to 3.5 kHz (Fig. 3). Group d cells were attached to the pyriform vesicle and were tuned to sound frequencies from 14 to 25 kHz (Fig. 4). The receptor cell bodies belonging to these groups were separated into three distinct regions of the auditory organ (Fig. 5). There was no evidence of any tonotopic organisation within the three groups (Fig. 6). These data therefore support the conclusion that the receptors within the auditory organ of the locust are organized into specific physiological groups (Michelsen 1971a) and thus suggest that the mechanics of the auditory apparatus may indeed play a role in defining the tuning of the receptors.  相似文献

Two helix-enriched fragments were isolated from a partial tryptic digest of the microfibrillar proteins of wool. From the known sequences of two of the constituent microfibrillar polypeptides, components 7c and 8c-1, one of these fragments was identified as arising from the N-terminal half of the helix-rich region and the other from the C-terminal half of this region. On the basis of the molecular weight in benign and dissociating solvents, crosslinking with dimethylsuberimidate, and amino acid and sequence analyses the N-terminal fragment was shown to be a tetrameric structure, consisting of two segments from each of the components 7 and 8 families. The C-terminal fragment was shown to be a two-stranded coiled-coil containing one chain segment from each of components 7 and 8. The evidence is not compatible with anything but a two-stranded coiled-coil as the basic molecular structure for the wool microfibril and provides experimental proof of a parallel arrangement of the chains. The thermal stabilities of the two fragments and of the parent microfibrillar complex from which they are derived are very similar indicating that the non-helical end regions of the polypeptide chains do not greatly affect the stability of the α-helical rod domain.  相似文献

Sex-differences in the maximum rate of torque development (/dtmax) may be due to differences in maximum muscle strength, because higher torque values mathematically lead to higher values for the rate of change in torque. The rate of change in the isometric torque-time curve is often normalized to the isometric maximum voluntary contraction (MVC) to evaluate males and females on a relative scale. Normalization eliminates sex-differences in /dtmax in the lower limbs because males and females are more comparable (i.e., differences between the sexes are relatively small) with respect to both muscle size and strength. However, normalization fails to result in parody in /dtmax of the upper limb, leading to the idea that other factors may be involved. This study determined if sex-differences in /dtmax in the upper limb can be attributed to differences in isometric MVC and/or a neural variable related to rate of increase in muscle activation (Q30). Forty-six participants (23 males, 23 females) performed maximal isometric elbow flexion contractions, “as hard and as fast as possible”. Maximum torque (τmax), /dtmax, and the rate of increase in surface electromyographic (sEMG) activity (Q30) were assessed. Muscle plus bone cross-sectional area (M + B CSA) of the upper arm was calculated to estimate differences in muscle size, only for comparative purposes. Maximum strength (55.5%) and muscle size (41.9%) of the elbow flexors in males were much greater than that of females (p < 0.05). There was a large difference (61.2%) between males and females with respect to /dtmax that was reduced by statistical correction using an analysis of covariance (ANCOVA). The percent differences were reduced to 36.7% (p < 0.05) for τmax and 54.4% (p < 0.05) for Q30, but was nearly eliminated to 13.8% (p > 0.05) when both variables were used simultaneously as covariates. Since sex-differences in the upper limb /dtmax persist, additional neural or biomechanical factors may be involved.  相似文献
