Subgroup structure, spacing patterns, and the relationship between spatial and behavioral interations were compared for captive baboons and squirrel monkeys. Analysis of the structure, intensity, and permanence of subgroups revealed that the baboons formed low intensity, overlapping subgroups which were relatively flexible while the squirrel monkeys segregated into permanent, high intensity, mutually exclusive cliques. Clique associations and social proximity relationships were found to be better predictors than dominance rank of the nature and frequency of behavioral interactions in the two colonies.  相似文献

Antiviral resistance is currently monitored by a labelled enzymatic assay, which can give inconsistent results because of the short half‐life of the labelled product, and variations in assay conditions. In this paper, we describe a competitive surface plasmon resonance (SPR) inhibition assay for measuring the sensitivities of wild‐type neuraminidase (WT NA) and the H274Y (histidine 274 tyrosine) NA mutant to antiviral drugs. The two NA isoforms were expressed in High‐five™ (Trichoplusia ni) insect cells. A spacer molecule (1,6‐hexanediamine (HDA)) was conjugated to the 7‐hydroxyl group of zanamivir, and the construct (HDA‐zanamivir) was immobilized onto a SPR sensor chip to obtain a final immobilization response of 431 response units. The immobilized HDA‐zanamivir comprised a bio‐specific ligand for the WT and mutant proteins. The effects of the natural substrate (sialic acid) and two inhibitors (zanamivir and oseltamivir) on NA binding to the immobilized ligand were studied. The processed SPR data was analysed to determine 50% inhibitory concentrations (IC50‐spr), using a log dose–response curve fit. Although both NA isoforms had almost identical IC50‐spr values for sialic acid (WT = 5.5 nM; H274Y mutant = 3.25 nM) and zanamivir (WT = 2.16 nM; H274Y mutant = 2.42 nM), there were significant differences between the IC50‐spr values obtained for the WT (7.7 nM) and H274Y mutant (256 nM) NA in the presence of oseltamivir, indicating that oseltamivir has a reduced affinity for the H274Y mutant. The SPR inhibition assay strategy presented in this work could be applied for the rapid screening of newly emerging variants of NA for their sensitivity to antiviral drugs. American black bears (Ursus americanus) were extirpated from Oklahoma, USA, in the early twentieth century but have since recolonized eastern portions of the state after immigrating from Arkansas, where they were successfully translocated. Within the last 2 decades, a population of black bears was detected in the Oklahoma Ozark region, prompting studies to determine population size, growth rate, and genetic makeup. To understand how black bears were recolonizing the human-dominated landscape, we investigated resource selection at 2 scales. Between 2011 and 2016, we collected global positioning system collar spatial data for 10 males and 13 females. We calculated average kernel density home ranges on a seasonal scale for all collared bears. We used generalized linear mixed models to calculate resource selection functions at the study area, defined by locations of all radio-collared black bears (second order) and the scale of individual black bear home ranges (third order). Resource selection did not differ significantly by sex. Black bears across seasons and scales selected riparian forest and moist oak (Quercus spp.) forest land cover types and mostly selected against indicators of human activity (e.g., pasture-prairie, anthropogenic land cover types, roads, and areas of high human population density). Black bears also selected areas with rugged terrain at high elevations, although not consistently across seasons and scales. Black bear recolonization appeared to be negatively affected by areas and features characterized as human-altered. Further expansion of the range of black bears may be limited by anthropogenic disturbance in the region. © 2021 The Wildlife Society.  相似文献

Conventional MHC class Ia-restricted CD8(+) T cells play a dominant role in the host response to virus infections, but recent studies indicate that T cells with specificity for nonclassical MHC class Ib molecules may also participate in host defense. To investigate the potential role of class Ib molecules in anti-viral immune responses, K(b-/-)D(b-/-)CIITA(-/-) mice lacking expression of MHC class Ia and class II molecules were infected with lymphocytic choriomeningitis virus (LCMV). These animals have a large class Ib-selected CD8(+) T cell population and they were observed to mediate partial (but incomplete) virus clearance during acute LCMV infection as compared with K(b-/-)D(b-/-)β(2)-microglobulin(-/-) mice that lack expression of both MHC class Ia and class Ib molecules. Infection was associated with expansion of splenic CD8(+) T cells and induction of granzyme B and IFN-γ effector molecules in CD8(+) T cells. Partial virus clearance was dependent on CD8(+) cells. In vitro T cell restimulation assays demonstrated induction of a population of β(2)-microglobulin-dependent, MHC class Ib-restricted CD8(+) T cells with specificity for viral Ags and yet to be defined nonclassical MHC molecules. MHC class Ib-restricted CD8(+) T cell responses were also observed after infection of K(b-/-)D(b-/-)mice despite the low number of CD8(+) T cells in these animals. Long-term infection studies demonstrated chronic infection and gradual depletion of CD8(+) T cells in K(b-/-)D(b-/-)CIITA(-/-) mice, demonstrating that class Ia molecules are required for viral clearance. These findings demonstrate that class Ib-restricted CD8(+) T cells have the potential to participate in the host immune response to LCMV.  相似文献

Selective targeting of transfected mesenchymal stem cells (MSCs) carrying specific antioncogenes to the tumor was suggested as a treatment option. Bone morphogenetic protein-2 (BMP2) was shown to inhibit the proliferation and aggressiveness of osteosarcoma (OS) cells. Here, we aimed to assess the homing efficiency of intraperitoneally administered hMSCs transfected with BMP2 to the tumoral site and their effects on OS using an orthotopic xenograft murine model. Orthotopic xenograft murine model of OS in six-week-old female NOD/SCID mice using 143B cells was established. hMSCs transfected with BMP2 (BMP2+hMSC) were used. In vivo experiments performed on four groups of mice that received no treatment, or intraperitoneally administered BMP2, hMSCs, and BMP2+hMSCs. Histopathological and immunohistochemical studies were used to evaluate the pathological identification and to assess the dimensions and necrotic foci of the tumor, the features of lung metastases, and immunostaining against p27, Ki-67, and caspase-3 antibodies. The osteogenic differentiation markers BMP2, BMP4, COL1A1, OPN, OCN and PF4 evaluated using RT-PCR. The tumor dimensions in the hMSCs group were significantly higher than those of the remaining groups (p < 0.01). The number of metastatic foci in the BMP2+hMSCs group was significantly lower than those of the other groups (p < 0.01). The current results showed that the intraperitoneal route could be efficiently used for targeting hMSCs to the tumoral tissues for effective BMP2 delivery. In this study, the effects of BMP2 transfected hMSCs on human OS and metastasis were promising for achieving osteogenic differentiation and reduced metastatic process.  相似文献

ABSTRACT: Melanoma and renal cell carcinoma have a well-documented tendency to develop metastases to the brain. Treating these lesions has traditionally been problematic, because chemotherapy has difficulty crossing the blood brain barrier and whole brain radiation therapy (WBRT) is a relatively ineffective treatment against these radioresistant tumor histologies. In recent years, stereotactic radiosurgery (SRS) has emerged as an effective and minimally-invasive treatment modality for irradiating either single or multiple intracranial structures in one clinical treatment setting. For this reason, we conducted a review of modern literature analyzing the efficacy of SRS in the management of patients with melanoma and renal cell carcinoma brain metastases. In our analysis we found SRS to be a safe, effective and attractive treatment modality for managing radioresistant brain metastases and highlighted the need for randomized trials comparing WBRT alone vs. SRS alone vs. WBRT plus SRS in treating patients with radioresistant brain metastases.  相似文献

Opportunistic fungal infections increase morbidity and mortality in COVID-19 patients monitored in intensive care units (ICU). As patients’ hospitalization days in the ICU and intubation period increase, opportunistic infections also increase, which prolongs hospital stay days and elevates costs. The study aimed to describe the profile of fungal infections and identify the risk factors associated with mortality in COVID-19 intensive care patients. The records of 627 patients hospitalized in ICU with the diagnosis of COVID-19 were investigated from electronic health records and hospitalization files. The demographic characteristics (age, gender), the number of ICU hospitalization days and mortality rates, APACHE II scores, accompanying diseases, antibiotic-steroid treatments taken during hospitalization, and microbiological results (blood, urine, tracheal aspirate samples) of the patients were recorded. Opportunistic fungal infection was detected in 32 patients (5.10%) of 627 patients monitored in ICU with a COVID-19 diagnosis. The average APACHE II score of the patients was 28 ± 6. While 25 of the patients (78.12%) died, seven (21.87%) were discharged from the ICU. Candida parapsilosis (43.7%) was the opportunistic fungal agent isolated from most blood samples taken from COVID-19 positive patients. The mortality rate of COVID-19 positive patients with candidemia was 80%. While two out of the three patients (66.6%) for whom fungi were grown from their tracheal aspirate died, one patient (33.3%) was transferred to the ward. Opportunistic fungal infections increase the mortality rate of COVID-19-positive patients. In addition to the risk factors that we cannot change, invasive procedures should be avoided, constant blood sugar regulation should be applied, and unnecessary antibiotics use should be avoided.  相似文献

Quinoa (Chenopodium quinoa Willd.) is adapted to the harsh environments of the Andean Altiplano region. Its seeds have a well-balanced amino acid composition and exceptionally high protein content with respect to human nutrition. Quinoa grain is a staple in the diet of some of the most impoverished people in the world. The plant is an allotetraploid displaying disomic inheritance (2n=4x=36) with a di-haploid genome of 967 Mbp (megabase pair), or 2C=2.01 pg. We constructed two quinoa BAC libraries using BamHI (26,880 clones) and EcoRI (48,000 clones) restriction endonucleases. Cloned inserts in the BamHI library average 113 kb (kilobase) with approximately 2% of the clones lacking inserts, whereas cloned inserts in the EcoRI library average 130 kb and approximately 1% lack inserts. Three plastid genes used as probes of high-density arrayed blots of 73,728 BACs identified approximately 2.8% of the clones as containing plastid DNA inserts. We estimate that the combined quinoa libraries represent at least 9.0 di-haploid nuclear genome equivalents. An average of 12.2 positive clones per probe were identified with 13 quinoa single-copy ESTs as probes of the high-density arrayed blots, suggesting that the estimate of 9.0× coverage of the genome is conservative. Utility of the BAC libraries for gene identification was demonstrated by probing the library with a partial sequence of the 11S globulin seed storage protein gene and identifying multiple positive clones. The presence of the 11S globulin gene in four of the clones was verified by direct comparison with quinoa genomic DNA on a Southern blot. Besides serving as a useful tool for gene identification, the quinoa BAC libraries will be an important resource for physical mapping of the quinoa genome.  相似文献

Aim Applying water‐energy dynamics and heterogeneity theory to explain species richness via remote sensing could allow for the regional characterization and monitoring of vegetation community assemblages and their environment. We assess the relationship of multi‐temporal normalized difference vegetation index (NDVI) to plant species richness in vegetation communities. Location California, USA. Methods Sub‐regions containing species inventories for chaparral, coastal sage scrub, foothill woodland, and yellow pine forest communities were intersected with a vegetation community map and an AVHRR NDVI time series for 1990, 1991, 1992, 1995 and 1996. Principal components analysis reduced the AVHRR data to three variables representing the sum and temporal trajectories of NDVI within each community. A fourth variable representing heterogeneity was tested using the standard deviation of the first component. Quadratic forms of these variables were also tested. Species richness was analysed by stepwise regression. Results Chaparral, coastal sage scrub, and yellow pine forest had the best relationships between species richness and NDVI. Richness of chaparral was related to NDVI heterogeneity and spring greenness (r2 varied between 0.26 and 0.62 depending on year of NDVI data). Richness of coastal sage scrub was nonlinearly related to annual NDVI and heterogeneity (r2 0.63–0.81), with peak richness at intermediate values. Foothill woodland richness was related to heterogeneity in a monotonic curvilinear fashion (r2 0.28–0.35). Yellow pine forest richness was negatively related to spring greenness and positively related to heterogeneity (r2 0.40–0.46). Main Conclusions While NDVI's relationship to species richness varied, the selection of NDVI variables was generally consistent across years and indicated that spatial variability in NDVI may reflect important patterns in water‐energy use that affect plant species richness. The principal component axis that should correspond closely with annual mean NPP showed a less prominent role. We conclude that plant species richness for coarse vegetation associations can be characterized and monitored at a regional scale and over long periods of time using relatively coarse resolution NDVI data.  相似文献
