Capitol Reef National Park in central Utah, USA surrounds 22 managed fruit orchards started over a century ago by Mormon pioneers.
Honey bees are imported for pollination, although the area in which the Park is embedded has over 700 species of native bees,
many of which are potential orchard pollinators. We studied the visitation of native bees to apple, pear, apricot, and sweet
cherry over 2 years. Thirty species of bees visited the flowers but, except for pear flowers, most were uncommon compared
to honey bees. Evidence that honey bees prevented native bees from foraging on orchard crop flowers was equivocal: generally,
honey bee and native bee visitation rates to the flowers were not negatively correlated, nor were native bee visitation rates
positively correlated with distance of orchards from honey bee hives. Conversely, competition was tentatively suggested by
much larger numbers of honey bees than natives on the flowers of apples, apricots and cherry; and by the large increase of
native bees on pears, where honey bee numbers were low. At least one-third of the native bee species visiting the flowers
are potential pollinators, including cavity-nesting species such as Osmia lignaria propinqua, currently managed for small orchard pollination in the US, plus several fossorial species, including one rosaceous flower
specialist (Andrena milwaukiensis). We suggest that gradual withdrawal of honey bees from the Park would help conserve native bee populations without decreasing
orchard crop productivity, and would serve as a demonstration of the commercial value of native pollinators. 相似文献

Chemical shift assignment is the first step in all established protocols for structure determination of uniformly labeled
proteins by NMR. The explosive growth in recent years of magic-angle spinning (MAS) solid-state NMR (SSNMR) applications is
largely attributable to improved methods for backbone and side-chain chemical shift correlation spectroscopy. However, the
techniques developed so far have been applied primarily to proteins in the size range of 5–10 kDa, despite the fact that SSNMR
has no inherent molecular weight limits. Rather, the degeneracy inherent to many 2D and 3D SSNMR spectra of larger proteins
has prevented complete unambiguous chemical shift assignment. Here we demonstrate the implementation of 4D backbone chemical
shift correlation experiments for assignment of solid proteins. The experiments greatly reduce spectral degeneracy at a modest
cost in sensitivity, which is accurately described by theory. We consider several possible implementations and investigate
the CANCOCX pulse sequence in detail. This experiment involves three cross polarization steps, from H to CA[i], CA[i] to N[i],
and N[i] to C′[i−1], followed by a final homonuclear mixing period. With short homonuclear mixing times (<20 ms), backbone
correlations are observed with high sensitivity; with longer mixing times (>200 ms), long-range correlations are revealed.
For example, a single 4D experiment with 225 ms homonuclear mixing time reveals ∼200 uniquely resolved medium and long-range
correlations in the 56-residue protein GB1. In addition to experimental demonstrations in the 56-residue protein GB1, we present
a theoretical analysis of anticipated improvements in resolution for much larger proteins and compare these results in detail
with the experiments, finding good agreement between experiment and theory under conditions of stable instrumental performance. 相似文献

We studied the breeding system and flower visitors of the endangered plant, Penstemon haydenii, at several south-central Wyoming, USA occurrences. In agreement with earlier studies of the species 300 km to the east in
Nebraska, we found Wyoming plants to be self-incompatible and pollinator-dependent for sexual reproduction. Flower visitors
were several species of native bees in the families Apidae (particularly bumblebees), Halictidae (small sweat bees), and Megachilidae
(especially in the genus Osmia); and the masarid wasp Pseudomasaris vespoides. Especially important was Osmia brevis, an abundant megachilid bee, and one of only two species (the sweat bee Lasioglossum (Dialictus) pruinosum was the other) present at all five sites. As in Nebraska, fruit set did not differ between our experimental cross-pollination
treatment and an open-pollinated control. However, unlike Nebraska, open-pollinated treatments in Wyoming produced significantly
fewer seeds per fruit than the experimental out-crossing treatment. We discuss several possible explanations for seed limitation:
(1) a scarcity of pollinators early in the flowering season; (2) resource competition for developing ovules on open-pollinated
inflorescences but not on experimental inflorescences; (3) the deposition of self pollen through intra-inflorescence and intra-genet
pollinator movements; (4) few S-alleles and mating types in the Wyoming metapopulation compared to the Nebraska metapopulation,

The impacts of wind energy on bat populations is a growing concern because wind turbine blades can strike and kill bats, and wind turbine development is increasing. We tested the effectiveness of 2 management actions at 2 wind-energy facilities for reducing bat fatalities: curtailing turbine operation when wind speeds were <5.0 m/second and combining curtailment with an acoustic bat deterrent developed by NRG Systems. We measured the effectiveness of the management actions using differences in counts of bat carcasses quantified by daily and twice-per-week standardized carcass searches of cleared plots below turbines, and field trials that estimated searcher efficiency and carcass persistence. We studied turbines located at 2 adjacent wind-energy facilities in northeast Illinois, USA, during fall migration (1 Aug–15 Oct) in 2018. We estimated the effectiveness of each management action using a generalized linear mixed-effects model with several covariates. Curtailment alone reduced overall bat mortality by 42.5% but did not reduce silver-haired bat (Lasionycteris noctivagans) mortality. Overall bat fatality rates were 66.9% lower at curtailed turbines with acoustic deterrents compared to turbines that operated at manufacturer cut-in speed. Curtailment and the deterrent reduced bat mortality to varying degrees between species, ranging from 58.1% for eastern red bats (Lasiurus borealis) to 94.4 for big brown bats (Eptesicus fuscus). Hoary (Lasiurus cinereus) and silver-haired bat mortality was reduced by 71.4% and 71.6%, respectively. Our study lacked a deterrent-only treatment group because of the expense of acoustic deterrents. We estimated the additional reduction in mortality with concurrent deployment of the acoustic deterrent and curtailment under the assumption that curtailment and the acoustic deterrent would have reduced mortality by the same percentage at adjacent wind-energy facilities. Acoustic deterrents resulted in 31.6%, 17.4%, and 66.7% additional reductions of bat mortality compared to curtailment alone for eastern red bat, hoary bat, and silver-haired bat, respectively. The effectiveness of acoustic deterrents for reducing bat mortality at turbines with rotor-swept area diameters >110 m is unknown because high frequency sound attenuates quickly, which reduces coverage of rotor-swept areas. Management actions should consider species differences in the ability of curtailment and deterrents to reduce bat mortality and increase energy production. 相似文献

Urbanisation affects indigenous fauna in many ways; some species persist and even increase in urban areas, whereas others are lost. The causative mechanisms determining changes in distributions and community structure remain elusive. We investigated three hypothesized mechanisms, which influence success or failure of the insectivorous bat assemblage across the urban landscape of Sydney, Australia; landscape heterogeneity (diversity of land uses), productivity (as indexed by landscape geology) and trait diversity. We present data on species richness and activity (bat passes per night) collected systematically using ultrasonic bat detectors from randomly selected landscapes (each 25 km2). Landscapes were categorized into classes including ‘urban’, ‘suburban’ and ‘vegetated’, where suburban sites were additionally stratified based on geology, as a proxy for productivity. Four landscape elements were sampled within each landscape, including remnant bushland (>2 ha), riparian areas, open space/parkland and residential/built space. We found that there was significantly greater bat activity and more species of bat in areas on fertile shale geologies (p<0.05), supporting the productivity, rather than the heterogeneity hypothesis. Within landscapes, there was no significant effect of the landscape element sampled, although bushland and riparian sites recorded greater bat activity than open space or backyard sites. Using general linear mixed models we found bat activity and species richness were sensitive to landscape geology and increasing housing density at a landscape scale. Using an RLQ analysis a significant relationship was found between these variables and species traits in structuring the community present (p<0.01). Specifically, open‐adapted bats were associated with areas of greater housing density, while clutter‐adapted bats were uncommon in urban areas and more associated with greater amounts of bushland in the landscape. Overall we found greater support for the productivity and traits hypotheses, rather than the heterogeneity hypothesis. The degree of urbanisation and amount of bushland remaining, in combination with landscape geology, influenced bat activity and mediated the trait response. Our findings reflect global trends of species diversity and abundance in urban landscapes, suggesting that processes affecting bat species distribution in urban ecosystems may be predictable at a landscape scale. 相似文献

Modification of bacterial surface structures, such as the lipid A portion of lipopolysaccharide (LPS), is used by many pathogenic bacteria to help evade the host innate immune response. Helicobacter pylori, a gram-negative bacterium capable of chronic colonization of the human stomach, modifies its lipid A by removal of phosphate groups from the 1- and 4'-positions of the lipid A backbone. In this study, we identify the enzyme responsible for dephosphorylation of the lipid A 4'-phosphate group in H. pylori, Jhp1487 (LpxF). To ascertain the role these modifications play in the pathogenesis of H. pylori, we created mutants in lpxE (1-phosphatase), lpxF (4'-phosphatase) and a double lpxE/F mutant. Analysis of lipid A isolated from lpxE and lpxF mutants revealed lipid A species with a 1 or 4'-phosphate group, respectively while the double lpxE/F mutant revealed a bis-phosphorylated lipid A. Mutants lacking lpxE, lpxF, or lpxE/F show a 16, 360 and 1020 fold increase in sensitivity to the cationic antimicrobial peptide polymyxin B, respectively. Moreover, a similar loss of resistance is seen against a variety of CAMPs found in the human body including LL37, β-defensin 2, and P-113. Using a fluorescent derivative of polymyxin we demonstrate that, unlike wild type bacteria, polymyxin readily associates with the lpxE/F mutant. Presumably, the increase in the negative charge of H. pylori LPS allows for binding of the peptide to the bacterial surface. Interestingly, the action of LpxE and LpxF was shown to decrease recognition of Helicobacter LPS by the innate immune receptor, Toll-like Receptor 4. Furthermore, lpxE/F mutants were unable to colonize the gastric mucosa of C57BL/6J and C57BL/6J tlr4 -/- mice when compared to wild type H. pylori. Our results demonstrate that dephosphorylation of the lipid A domain of H. pylori LPS by LpxE and LpxF is key to its ability to colonize a mammalian host. 相似文献

PCR-based surveys have shown that guppies (Poecilia reticulata) have an unusually large visual-opsin gene repertoire. This has led to speculation that opsin duplication and divergence has enhanced the evolution of elaborate male coloration because it improves spectral sensitivity and/or discrimination in females. However, this conjecture on evolutionary connections between opsin repertoire, vision, mate choice, and male coloration was generated with little data on gene expression. Although highly conserved throughout evolution, the exact biological function of the prion protein is still unclear. In an effort to identify the potential biological functions of the prion protein we conducted a small-molecule screening assay using the Syrian hamster prion protein [shPrP(90-232)]. The screen was performed using a library of 149 water-soluble metabolites that are known to pass through the blood-brain barrier. Using a combination of 1D NMR, fluorescence quenching and surface plasmon resonance we identified thiamine (vitamin B1) as a specific prion ligand with a binding constant of ~60 μM. Subsequent studies showed that this interaction is evolutionarily conserved, with similar binding constants being seen for mouse, hamster and human prions. Various protein construct lengths, both with and without the unstructured N-terminal region in the presence and absence of copper, were examined. This indicates that the N-terminus has no influence on the protein's ability to interact with thiamine. In addition to thiamine, the more biologically abundant forms of vitamin B1 (thiamine monophosphate and thiamine diphosphate) were also found to bind the prion protein with similar affinity. Heteronuclear NMR experiments were used to determine thiamine's interaction site, which is located between helix 1 and the preceding loop. These data, in conjunction with computer-aided docking and molecular dynamics, were used to model the thiamine-binding pharmacophore and a comparison with other thiamine binding proteins was performed to reveal the common features of interaction. 相似文献
