Growth hormone release inhibiting hormone (GH-RIH) was infused at a rate of 1·3 μg/min for 28 hours into four patients with acromegaly, two of whom also had clinical diabetes mellitus. Growth hormone and glucagon were suppressed throughout the infusion though delayed secretion of insulin occurred in association with both meals and an oral glucose load. Glucose tolerance was improved in one diabetic patient who was taking chlorpropamide while the other required much less insulin than usual. Secretion of endogenous thyroid-stimulating hormone was lowered in one euthyroid patient on carbimazole. Luteinizing hormone, follicle-stimulating hormone, ACTH, and prolactin were not affected. Serum somatomedin levels were reduced in one patient. There was a rapid rebound of all the suppressed hormones when the infusions stopped. Longer-acting analogues of GH-RIH will be needed before long-term therapy of acromegaly or diabetes mellitus becomes possible, but such preparations should be available soon for clinical trial. 相似文献

Summary Sectoring was induced with x-rays or ultraviolet in a diploid yeast strain heterozygous for seven genes located on one chromosome arm. The frequencies of sectoring of different genes were approximately linearly related to their distance from the centromere. If two or more adjacent genes sectored, the event could be explained by mitotic crossing over. Sectoring of single genes, however, was mostly nonreciprocal and resembled a conversion-type event. Summary Mucosal hypertonicity, produced by the addition of NaCl, KCl, mannitol, urea, sucrose or raffinose, reduced the electrical resistance of toad urinary bladder and induced bullous deformations (blisters) of the most apical junctions of the mucosal epithelium: the smaller solutes were most effective in eliciting both phenomena. Study of the effect of addition and subsequent removal of mannitol from the mucosal medium indicated that both the electrical and morphologic changes are reversible and follow the same time course. Mucosal hypertonicity induced comparable changes in the tissue in the presence or absence of inhibition of active sodium transport by replacement of sodium by choline, or by addition of ouabain or amiloride. Dilution of the tonicity of the serosal medium similarly reduced the tissue resistance and induced blisters within the epithelium, demonstrating that the osmotic gradient, rather than the mucosal hypertonicity itself is the cause of the osmotically-induced resistance change. The data indicate, therefore, that the osmotic gradient reduces the electrical resistance of the tissue primarily by deforming the apical junctions.The simplest interpretation of the data is that the apical tight junctions are considerably more permeable to water and small solutes than had previously been thought. Addition of solute to the mucosal medium leads to the diffusion of solute into the junctions: the subsequent transfer of water from the lateral intercellular spaces and/or the adjacent cellular cytoplasm, deforms these structures and reduces the resistance to the passage of ions across the tissue. The results suggest that the apical junctions constitute the rate-limiting permeability barrier of the putative parallel shunt pathway across toad bladder. 相似文献

X-Ray-survival curves of haploid, diploid, triploid and tetraploid yeast strains homozygous for the X-ray-sensitive mutation rad52 (previously xs1 are presented. These curves suggest that strains carrying the rad52 mutation may be more susceptible than wild type to X-ray-induced dominant lethal damage. For the crosses (+ × +, rad52 × rad52, + × rad52, rad52 × +) in which only one parent was irradiated, the relationships between zygote survival and X-ray dose were similar except for rad52 × rad52. In this cross a considerably higher frequency of dominant lethal damage was observed. This observation indicates that the rad52 mutant lacks a repair system for X-ray damage and is consistent with the proposal that unrepaired chromosome damage is the event which leads to dominant lethality and reproductive cell death. 相似文献

Human erythrocytes were incubated in a Ringer's solution enriched with 10–18% H217O. The longitudinal relaxation time (T1) of the 17O was determined separately in samples of red cell suspesions, packed cells, and supernatant. The longitudinal relaxation of 17O in erythrocyte suspensions was non-exponential, reflecting water exchange across the cell membranes as well as relaxation processes inside and outside the cell.The T1 of intracellular 17O is 4–5 times shorter than in the supernatant, similar to the enhancement of proton relaxation by hemoglobin in erythrocytes and free solution at the frequency applied (8.13 MHz). This datum is consistent with the thesis that hemoglobin modifies the NMR relaxation behavior of water inside cells and in free solution in the same way.The rate constant for water exchange was calculated to be 60 and 107 s−1 at 25 and at 37° C, respectively. Na. Both the phorbol ester 12-O-tetradecanoylglycerol (DiC8) were used as PKC activators. Blocking Ca2+ entry into the cytosol (either from external or internal stores) reduced the subsequent natriferic effect of the PKC activators. This negative interaction did not simply reflect saturation of activation of the apical Na+ channels, since the stimulations produced by blocking Ca2+ entry and adding cyclic AMP were simply additive.The Ca2+ dependence of the natriferic effect could have reflected either a direct action of cytosolic Ca2+ on PKC or an indirect action on the final receptor site (the Na+ channel). To distinguish between these possibilities, the TPA- and phospholipid-dependent kinase activity of broken-cell preparations was assayed. The kinase activity was not stimulated by physiological levels of Ca2+, and in fact was inhibited at millimolar concentrations of Ca2+.We conclude that the effects of Ca2+ on the natriferic response to PKC activators are indirect. Reducing cytosolic uptake of Ca2+ may have stimulated Na+ transport by a chemical modification of the apical channels observed in other tight epithelia. The usual stimulation of Na+ transport produced by PKC activators in frog skin may reflect the operation of a nonconventional form of PKC. In rats, remnant particles derived from chylomicron-like emulsions containing 1,3-dioleoyl-2-stearoylglycerol (OSO) are removed from plasma more slowly than remnants derived from triolein emulsions. The effect associated with a saturated acyl chain at the glycerol 2-position could be reproduced by incorporating 2-stearoylglycerol (MS) in a triolein emulsion. When MS solubilized with rat albumin or in plasma was injected before the injection of a triolein emulsion, clearance of the triolein emulsion was unchanged. The metabolic fate of MS, monitored with 14C-labelled MS, was similar whether incorporated in triacylglycerol emulsion or injected independently. More than 95% of MS had disappeared from the circulation by 5 min after the injection and the radioactivity was found in liver, spleen, muscle and adipose tissue. Some MS label appeared in plasma triacylglycerol. Remnants made in vitro by incubating triolein or OSO emulsions with post-heparin plasma showed no differences in their disappearance from plasma. With OSO emulsion, the in vitro remnants were found to contain more MS than remnants made in vivo in hepatectomized rats. Simultaneous injections of mixtures containing OSO and triolein emulsions, or triolein emulsions with and without MS, each labelled with either [3H]cholesteryl oleate or [14C]cholesteryl oleate showed consistently slower remnant removal and decreased liver uptake of the emulsions containing OSO or MS. Affinity columns and immunodiffusion all indicated that there was no difference in the amounts of apolipoprotein E associated with OSO or triolein particles. The protein spectra of in vivo remnants derived from OSO and triolein emulsion were also similar when examined by SDS-PAGE and isoelectric focusing gels. Our results show that the effects due to OSO or MS are mediated by the presence of MS in the emulsion particle surface, while indirect effects expressed in plasma or liver are excluded. The precise mechanism of the effect remains to be established, but it does not correlate with measurable changes in the spectra of apolipoproteins associated with the emulsion remnants. 相似文献

Summary Although peptide neurotransmitters have been shown to modulate hormone secretion in many glands, there are very few studies of neurotransmitters in the parathyroid gland. Bovine parathyroid glands were collected at a local abattoir, fixed with paraformaldehyde, sectioned using a cryostat, and stained by indirect immunohistochemistry for calcitonin gene-related peptide and substance P. We were able to positively identify both neuropeptides. Nerve fibres containing calcitonin gene-related peptide and substance P were identified in contact with the tunica media of arteries and arterioles and dispersed throughout the stroma of the gland. While many of the fibres encircled parenchymal lobules, no intimate contact with the peripheral chief cells was observed. All immunoreactive fibres were found to contain both neuropeptides. Since calcitonin gene-related peptide and substance P are vasodilators, they may increase blood flow within the gland. In addition, the neuropeptides may diffuse from perilobular nerve fibres into the parenchyma, thereby modulating secretion of parathyroid hormone. 相似文献

Probit plots of sperm concentration for 1711 suspected infertile men (those with azoospermia being excluded) were compared for the untransformed and loge-, square root- and cube root-transformed values. For the distribution of sperm concentrations, which was highly skewed towards low values, the square-root transformation produced the most normal (Gaussian) distribution. Loge and cube-root transformations caused skewing towards high values. Such treatment of the data should always be considered before using parametric statistical tests to make comparisons between sperm concentrations of groups of men. 相似文献
