OBJECTIVES--To determine the prevalence of antibodies to the human T cell leukaemia/lymphoma viruses (HTLV-I and HTLV-II) in blood donors in north London in order to assess the economic impact and the logistic effects that routine screening would have on the blood supply. DESIGN--All donations collected by the north London blood transfusion centre between January 1991 and June 1991 were screened for antibodies to HTLV-I and HTLV-II by modified, improved Fujirebio gel particle agglutination test. Positive samples were titrated and retested as necessary. SUBJECTS--96,720 unpaid volunteers, who gave 105,730 consecutive donations of blood and plasma. SETTING--North London blood transfusion centre. MAIN OUTCOME MEASURE--Observed numbers of donors confirmed to be seropositive for HTLV by reference laboratories. RESULTS--Of 2622 (2.5%) initially reactive samples, 414 (0.4% of all samples) gave a titre of > or = 1 in 16 on the modified agglutination test. Thirty five of the 414 serum samples yielded positive results on one of two enzyme linked immunosorbent assays (ELISA (Cambridge Biotech and Abbot)), and none of these results were confirmed by either reference laboratory. Five samples yielded positive results on both ELISAs and all five of these were confirmed to contain antibodies to HTLV. One of the five contained antibodies to HTLV-II and the others antibodies to HTLV-I. Four seropositive donors were white women whose only risk factor for infection was sexual contact. The fifth (positive for antibodies to HTLV-II) was an Anglo-Caribbean man who admitted to previous misuse of intravenous drugs. CONCLUSION--The prevalence of antibodies to HTLV in blood donors in north London was one in 19,344 (0.005%). Up to 100 donors a year might be identified in the United Kingdom as being infected with HTLV, although prevalence in different regions may vary considerably.  相似文献

The effects of restoring serosal potassium to potassium-depleted toad urinary bladders have been re-examined using double-barrelled microelectrodes. The data confirm the existence of a time-lag phenomenon, a dissociation between potassium reaccumulation and restoration of short-circuit current. Returning serosal potassium stimulates an increase in intracellular potassium activity 21-26 min before any increase can be detected in short-circuit current. The reaccumulation of potassium has been further studied using split frog skin, a far more suitable preparation for electrophysiologic study than toad bladder. Under baseline short-circuited conditions, potassium is accumulated against an electrochemical gradient of 22 +/- 4 mV. Reaccumulation of potassium by potassium-depleted tissues can be blocked by inhibiting the Na,K-exchange pump with high concentrations of ouabain. On the other hand, blocking apical sodium entry by the addition of 10(-4) M amiloride to the outer bathing medium does not interfere with reaccumulation of potassium. The data support the concept that the time-lag phenomenon of toad bladder reflects stimulation of potassium reaccumulation by the sodium pump in exchange for the extrusion of excess cell sodium collected during the period of potassium depletion. This reaccumulation of potassium can proceed before the entry of significant added amounts of sodium across the apical plasma membrane.  相似文献

Five hundred thirty-four naturally bred Holstein dairy cows from two Colorado dairies were examined perrectum between 21 and 36 days after calving. Alternate cows in each herd received either 25 mg PGF-2 alpha or no treatment at the time of initial examination. No differences were found in days open or pregnancy rates between groups (P>0.25).Days open in 23 cows with pyometra were 98.7 ± 9.4, and 77.0 ± 2.2 in 493 herdmates without pyometra. The large inequalities in numbers precluded valid statistical comparisons between these groups. No overall difference (P>0.25) in reproductive performance was found in cows with pyometra treated at the initial examination compared to those not treated until two to four weeks after the initial examination. Contributing to the lack of significance were a number of cows with pyometra that had spontaneous cures before treatment was considered. Untreated cows that recovered spontaneously had better (P<0.05) reproductive performance than herdmates that required later treatments to initiate uterine evacuation. No overall advantage in reproductive performance was obtained by the routine treatment of postpartum dairy cows with PGF-2 alpha but treatment of cows with pyometra should not be delayed.  相似文献

Summary Computer simulations of tight epithelia under three experimental conditions have been carried out, using the rheogenic nonlinear model of Lew, Ferreira and Moura (Proc. Roy. Soc. London. B 206:53–83, 1979) based largely on the formulation of Koefoed-Johnsen and Ussing (Acta Physiol. Scand.42:298–308, 1958). First, analysis of the transition between the short-circuited and open-circuited states has indicated that (i) apical Cl permeability is a critical parameter requiring experimental definition in order to analyze cell volume regulation, and (ii) contrary to certain experimental reports, intracellular Na+ concentration (c Na c ) is expected to be a strong function of transepithelial clamping voltage. Second, analysis of the effects of lowering serosal K+ concentration (c K s ) indicates that the basic model cannot simulate several well-documented observations; these defects can be overcome, at least qualitatively, by modifying the model to take account of the negative feedback interaction likely to exist between the apical Na+ permeability andc Na c . Third, analysis of the effects induced by lowering mucosal Na+ concentration (c Na m ) strongly supports the concept that osmotically induced permeability changes in the apical intercellular junctions play a physiological role in conserving the body's stores of NaCl. The analyses also demonstrate that the importance of Na+ entry across the basolateral membrane is strongly dependent upon transepithelial potential,c Na m andc K s ; under certain conditions, net Na+ entry could be appreciably greater across the basolateral than across the apical membrane.  相似文献
