根据公布的黑麦特异重复序列pSc119.1设计特异引物, 分别对两套姊妹T1RS·1BL易位系川农12(CN12)、川农17(CN17)、川农18(CN18)和96Ⅰ176-1、96Ⅰ176-3的基因组DNA进行扩增。从CN12、CN17、CN18的基因组中都能扩增出目的片段, 在96Ⅰ176-1的基因组中, 除目的片段外, 还扩增出了一条非目的片段, 但在96Ⅰ176-3的基因组中没有扩增出产物。Southern blot分析表明, pSc119.1序列并未从96Ⅰ176-3的基因组中消失。将CN12、CN17、CN18的目的片段回收克隆后, 对每个片段各随机挑取10个克隆进行测序, pSc119.1序列在3个品种中都发生了变异, 且在CN18中的变异程度最大。所测的序列多数与原序列达94%和95%的相似性, 在这些序列中, 碱基的改变具有一定的规律, 即多数为转换, 少数为颠换, 且变异碱基和变异位置具有较高的一致性。远缘杂交后代的进化可能是一个持续过程, 姊妹系内的株系之间某些性状存在差异, 这些差异很可能与重复序列的变异有关, 这为后生遗传效应机制的研究提供了有用的材料。 相似文献

In Synechocystis sp. PCC 6803, the loop domain (aa 1–70) of the phycobilisome core-membrane linker, LCM, was found to interact with the glycosyl transferase homolog, Sll1466. Growth of a Sll1466 knock-out mutant was slightly faster in low light, but strongly inhibited in high light; the phenotype is discussed in relation to the regulation of light energy transfer to photosystem II. At the molecular level, the mutant shows the following changes compared to the wild type: (1) a smaller size and higher mobility of phycobilisomes on the thylakoid membrane, and (2) a changed lipid composition of the thylakoid membrane, especially decreased amounts of digalactosyl diacylglycerol. These results indicate a profound regulatory role for Sll1466 in regulating photosynthetic energy transfer. 相似文献

The development of artificial nucleases that hydrolyze DNA or RNA is of great interest in molecular biology, biotechnology, and medicine. We now report that a magnesium(II) complex of diethylenetriamine (Mg-dien) can effectively promote the double-stranded cleavage of plasmid DNA and the dideoxynucleotide dApdA under physiological conditions of pH and temperature. Experiments performed in the presence of hydrogen peroxide, radical scavengers, or under rigorously anaerobic conditions indicate that DNA cleavage mediated by Mg-dien occurs via a hydrolytic path. Mg-dien efficiently hydrolyzes supercoiled pBR322 DNA and the pseudo-first-order rate constant at 37 degrees C and pH 8.0 is estimated to be 1.60 h(-1). The dinucleotide dApdA hydrolysis, with Mg-dien at 170 microM, shows a rate enhancement factor of ca. 5 x 10(8). 1H and 31P(1H) NMR studies show that Mg-dien effectively hydrolyzes 5'-dAMP to give deoxyadenosine and inorganic phosphate. While Mg2+ has been found at the catalytic sites of many natural nucleases, Mg-dien appears to be the first synthetic Mg2+-containing system capable of hydrolyzing dideoxynucleotides and DNA and thus may provide a simple model system to assist mechanistic studies of naturally occurring nucleases. 相似文献

Increased amounts of intrathecally synthesized IgG and oligoclonal bands have long been recognized as a hallmark of multiple sclerosis (MS). B cells and plasma cells are components of the inflammatory infiltrates in both active and chronic MS lesions, and increased numbers of these cells are present in MS cerebrospinal fluid (CSF). Single-cell RT-PCR was used to analyze both the CD19+ B cell and CD138+ plasma cell populations in CSF of two patients with clinically definite MS and of one MS patient whose CSF was obtained after a clinically isolated syndrome, but before the second episode. Sequence analysis of amplified IgG V region sequences identified the rearranged germline segments, extent of somatic mutation, and clonal relationships within and between the two cell populations in the three MS patients. Expanded B cell and plasma cell clones were detected in each MS CSF and in all three patients the CD138+ IgG repertoire was more restricted. However, little if any significant sequence overlap was observed between the CD19+ and CD138+ repertoires of each donor. Detection of plasma cell clones by single-cell PCR will facilitate the in vitro production of recombinant Abs useful in identifying disease-relevant Ags. 相似文献

目的:对2012年广州市越秀区疑似麻疹风疹的血清学结果进行整理分析,以了解麻疹风疹病例的流行特点。方法:选取2012年1月-2012年12月的166例疑似麻疹风疹患者的血清作为研究对象,统计血清中IgM阳性抗体的检出情况,以及各年龄段、月份IgM抗体阳性的例数及构成比。结果:麻疹风疹疑似病例中有36.1%的患者检测出IgM抗体阳性,其中麻疹IgM阳性以0-1岁婴幼儿的检出率较高,可达44.4%。从月份来看,12月、8月和5月是麻疹的好发时期。风疹IgM阳性以20岁以上中老年患者检出率最高,为75.0%,7月、4月和5月是风疹的易感季节。结论:疑似麻疹风疹病例有较高的IgM抗体阳性检出率,0-1岁婴幼儿和20岁以上中老年人是麻疹风疹的易感人群,4-5、7-8和12月是发病的高峰时期,临床应注意加强防范。 相似文献

视觉感知的一系列研究都支持大范围拓扑感知的理论.拓扑性质作为整体性质,是视觉感知的基础.视觉对图形拓扑特征差异的感知要优先于对局部特征差异的感知.采用Y迷宫研究了小鼠对不同拓扑性质图形的识别.训练小鼠学习识别圆环和实心矩形这一对拓扑性质不同的图形,之后用拓扑特征相同或不同的其他图形测试小鼠,这些图形包括空心矩形、实心圆、缺口的圆环、缺口的空心矩形.实验结果表明,学会识别圆环(奖励)和实心矩形(无奖励)的小鼠无法区分实心圆和实心矩形以及圆环和空心矩形,但是能够分别从缺口圆环、缺口的空心矩形、实心圆与空心矩形组成的图形对中识别出空心矩形.因此证实了小鼠的视觉系统能够感知拓扑特征的差异并且具有对拓扑性质的概括能力.结果为拓扑知觉对视觉系统来说是基本的这一假设提供了证据. 相似文献

The study described the uncertainties of NPP estimation due to larch defoliation by caterpillars during 2001–2004 in Northeastern China, which is an order of magnitude larger in area and severe than all previous recorded outbreaks. Using “representative needles” collected in the field, we related spectral responses and photosynthetic contents of needles to ecophysiological parameters in NPP estimation for different degrees of caterpillar damage. It is concluded that locations of reflection peaks at or near green red near-infrared wavelength moving toward long-wave direction with aggravated damage degree, which is useful to earlier prediction of caterpillar damage. Contents of chlorophyll-a reduced by 40.26, 66.29 and 86.88% for light, moderate and severe damage samples respectively compared with healthy, chlorophyll-b reduced by 20.79, 52.67 and 82.39%, and carotenoid reduced by 9.80, 45.06 and 73.15%. Estimated NDVI values decreased by 9.89–31.08, 38.16–47.47 and 61.90–66.71%, respectively. It results to the decrease of LAI by 39.76–74.34, 85.85–93.64 and 94.61–98.51%, and reduction of FPAR and APAR by 10.16–37.47, 39.21–57.24 and 63.60–80.44%. In addition, the increased minimum visible reflectance during 580–680 nm leads to the decrease of estimated aboveground biomass by 14.62, 48.86 and 56.84%, and reduction of estimated autotrophic respiration by 10.32, 39.11 and 46.95%. Ultimately, the caterpillar outbreak results to the directly underestimation of NPP by 10.01–64.63, 39.32–75.37 and 80.26–100% for light, moderate and severe samples compared with healthy.
