【目的】检测 Q 型烟粉虱 Bemisia tabaci (Gennadius)体内 Rickettsia 的感染情况,研究分析Rickettsia 共生菌经烟粉虱传入豇豆植物后的分布、转移效率等。【方法】以 Q 型烟粉虱为实验材料,利用常规 PCR 及荧光原位杂交技术(FISH),检测了烟粉虱体内 Rickettsia 的感染率,以及 Rickettsia 传入豇豆植物体内后的存留情况。【结果】 Q 型烟粉虱可以通过取食将 Rickettsia 传至豇豆植株内;接虫数量与 Rickettsia传入效率及其在取食部位相邻的下部叶片中检测到的起始时间呈负相关;Rickettsia 经烟粉虱取食传入豇豆叶片后,集中分布在叶片的韧皮部筛管中;基于16S rRNA 的系统发育分析结果表明,Q 型烟粉虱体内的Rickettsia 与经取食传入豇豆叶片的 Rickettsia 高度同源。【结论】 Rickettsia 可以通过烟粉虱的取食传入植物体内,并且可以在相邻叶片之间转移传播,Rickettsia 在由寄主昆虫向植株传播过程中高度保守。  相似文献

Cyclic nucleotide phosphodiesterases (PDEs) comprise a superfamily of enzymes that serve as drug targets in many human diseases. There is a continuing need to identify high-specificity inhibitors that affect individual PDE families or even subtypes within a single family. The authors describe a fission yeast-based high-throughput screen to detect inhibitors of heterologously expressed adenosine 3',5'-cyclic monophosphate (cAMP) PDEs. The utility of this system is demonstrated by the construction and characterization of strains that express mammalian PDE2A, PDE4A, PDE4B, and PDE8A and respond appropriately to known PDE2A and PDE4 inhibitors. High-throughput screens of 2 bioactive compound libraries for PDE inhibitors using strains expressing PDE2A, PDE4A, PDE4B, and the yeast PDE Cgs2 identified known PDE inhibitors and members of compound classes associated with PDE inhibition. The authors verified that the furanocoumarin imperatorin is a PDE4 inhibitor based on its ability to produce a PDE4-specific elevation of cAMP levels. This platform can be used to identify PDE activators, as well as genes encoding PDE regulators, which could serve as targets for future drug screens.  相似文献

The Euler/Cardan angles are commonly used to define the motions of the upper arm with respect to the trunk. This definition, however, has a problem in that the angles of both the horizontal flexion/extension and the axial rotation of the shoulder joint become unstable at the gimbal-lock positions. In this paper, a new definition of the axial rotation angle was proposed. The proposed angle was stable over the entire range of the shoulder motion. With the new definition, the neutral position of the axial rotation agreed with that in the conventional anatomy. The advantage of the new definition was demonstrated by measuring actual complex motions of the shoulder with a three-dimensional motion capture system.  相似文献

To make identification unambiguous, the sequence names have been updated from “SA‐lsf‐27 and SA‐lsf‐37” to “GISAID accession ID: EPI_ISL_672581 and EPI_ISL_672589”.Lastly, the authors declare in the Materials and Methods section that all experiments employed SARS‐CoV‐2 pseudovirus in cultured cells. 目的:研究复合疲劳大鼠血液、肌肉、肝脏和脑中矿物元素代谢变化的影响。方法:将30只大鼠随机分为正常对照组、食物限制组和复合疲劳组。经过5天的实验干预后,提取动物的血液、腓肠肌、肝脏和脑,并利用原子吸收分光光度法测量各组织中的钾(K)、钙(Ca)、镁(Mg)、铁(Fe)、锌(Zn)和铜(Cu)。结果:相对正常对照组和食物限制组,复合疲劳大鼠的肌肉、肝脏和脑中的K(P<0.01)和肝脏中的Fe(P<005)明显升高,血液中的Cu(与正常对照组比较P<0.01,与食物限制组比较P<0.05)明显下降;与对照组相比,复合疲劳大鼠的血液中的K明显升高(P<0.05),血液中的Mg和Zn(P<0.05),肌肉中的Ca、Mg和Zn(PCa<0.05,PMg<0.05,PZn<0.01),肝脏中的Ca和Zn(PCa<0.01,PZn<0.05),以及脑中的Fe、Mg和Zn(PFe<0.05,PMg<0.05,PZn<0.01)明显降低。结论:在复合疲劳状态下,大鼠血液、肌肉、肝脏和脑中的K、Ca、Mg、Fe、Zn和Cu代谢发生变化,可能在疲劳的发生与缓解中发挥作用。  相似文献

张家界市部分农作物可食部位硒含量研究   总被引:1,自引:0,他引:1  
采用国家标准( GB/T5009.93-2003),用荧光分光光度计对张家界市22种农作物40个样本可食部位中的硒含量进行了分析.结果表明,测定的22个种类40个样本均属于富硒及高硒作物,以茶叶的硒平均含量最高为2.0102 mg/kg,其次是豆类平均含量为1.8518 mg/kg,薯类含量最低为1.0240 mg/kg.所测定的各种作物可食部位富硒能力依次为:茶叶2.0102 mg/kg>豆类(干)1.8518 mg/kg>水稻、玉米1.5326 mg/kg>蔬菜类1.3438 mg/kg>水果类1.0783 mg/kg>薯类1.0240 mg/kg.该研究确定了部分农作物硒含量的水平及富硒作物品种.  相似文献

Novel chitosan-supported cinchona alkaloids have been developed as heterogeneous catalysts for enantioselective Michael reaction. As-synthesized products as organocatalysts for asymmetric Michael reaction have a high efficiency, providing highly functionalized products (containing adjacent quaternary and tertiary stereocenters) with good stereoselectivity (up to 93% enantiomeric excess) in high yields and recyclability (up to five runs).  相似文献

Cdc42-interacting protein-4 (CIP4) is an F-BAR (Fer/CIP4 and Bin, amphiphysin, Rvs) family member that regulates membrane deformation and endocytosis, playing a key role in extracellular matrix (ECM) deposition and invasion of cancer cells. These processes are analogous to those observed during the initial epithelial-mesenchymal transition (EMT) of renal tubular epithelial cells. The role of CIP4 in renal tubular EMT and renal tubulointerstitial fibrosis was investigated over the course of the current study, demonstrating that the expression of CIP4 increased in the tubular epithelia of 5/6-nephrectomized rats and TGF-β1 treated HK-2 cells. Endogenous CIP4 evidenced punctate localization throughout the cytosol, with elevated levels observed in the perinuclear region of HK-2 cells. Subsequent to TGF-β1 treatment, CIP4 expression increased, forming clusters at the cell periphery that gradually redistributed into the cytoplasm. Simultaneously, EMT induction in cells was confirmed by the prevalence of morphological changes, loss of E-cadherin, increase in α-SMA expression, and secretion of fibronectin. Overexpression of CIP4 promoted characteristics similar to those commonly observed in EMT, and small interfering RNA (siRNA) molecules capable of CIP4 knockdown were used to demonstrate reversed EMT. Cumulatively, results of the current study suggest that CIP4 promotes TGF-β1-induced EMT in tubular epithelial cells. Through this mechanism, CIP4 is capable of inducing ECM deposition and exacerbating progressive fibrosis in chronic renal failure.  相似文献

2016年7月对抚仙湖进行采样调查,研究抚仙湖超微型浮游藻类(超微藻)的空间分布特征及关键影响因子。结果表明,抚仙湖超微藻平均丰度为(8.58±3.25)×103个/mL,其中超微蓝藻丰度显著高于超微真核藻。超微藻丰度在沿岸带较高,敞水区相对较低,北部最深点低于南部最深点;垂直方向上,超微藻丰度在水下10 m处达到最大值,随着深度的增加丰度逐渐下降。通过方差膨胀因子分析和建模得到超微藻丰度和环境因子的相关关系,水体的浊度、pH以及总磷对超微真核藻丰度有显著影响,而超微蓝藻的丰度主要是受到总磷的影响。结合流式细胞分选和高通量测序得到了抚仙湖超微真核藻的群落结构特征,主要是金藻纲、硅藻纲、甲藻纲等,其中金藻纲占绝对优势。在空间上,不同湖区和不同深度超微真核藻的群落组成也存在差异:表层水体以金藻纲、硅藻纲、甲藻纲为主;而在深层水体中超微真核藻的多样性降低,金藻纲为优势种。超微藻作为贫营养湖泊初级生产力的主要贡献者,对其组成和分布的研究有助于更全面的认识抚仙湖生态系统结构和功能。  相似文献

Many studies have verified that microRNAs contribute a lot to neuropathic pain progression. Furthermore, nerve-related inflammatory cytokines play vital roles in neuropathic pain progression. miR-183 has been identified to have a common relationship with multiple pathological diseases. However, the potential effects of miR-183 in the process of neuropathic pain remain undetermined. Therefore, we performed the current study with the purpose of finding the functions of miR-183 in neuropathic pain progression using a chronic sciatic nerve injury (CCI) rat model. We demonstrated that miR-183 expression levels were evidently reduced in CCI rats in contrast with the control group. Overexpression of miR-183 produced significant relief of mechanical hyperalgesia, as well as thermal hyperalgesia in CCI rats. Furthermore, neuropathic pain-correlated inflammatory cytokine expression levels containing interleukin-6 (IL-6) and interleukin-1β (IL-1β), cyclooxygenase-2 (COX-2) were obviously inhibited by upregulation of miR-183. Meanwhile, dual-luciferase reporter assays showed MAP3K4 was a direct downstream gene of miR-183. The expression levels of MAP3K4 were modulated by the increased miR-183 negatively, which lead to the downregulation of IL-6, IL-1β, and COX-2, and then reduced neuropathic pain progression, respectively. Overall, our study pointed out that miR-183 was a part of the negative regulator which could relieve neuropathic pain by targeting MAP3K4. Thus it may provide a new clinical treatment for neuropathic pain patients clinical therapy.  相似文献
