目的探讨不同分娩方式对婴儿出生后1年内肠道菌群定植的影响。方法选取45例新生儿为研究对象,根据分娩方式分为自然分娩组(n=27)和剖宫产组(n=18)。收集婴儿出生后0(胎粪)、3、6和12个月的粪便标本,应用高通量测序技术分析肠道菌群多样性及组成。结果与自然分娩组比较,在0个月时剖宫产组婴儿粪便标本拟杆菌门的相对丰度显著降低(Z=-2.374 1,P=0.017 6)。2组研究对象中,除自然分娩组和剖宫产组0个月时婴儿粪便标本分别以埃希菌-志贺菌属和克雷伯菌属为优势菌属外,余下均以双歧杆菌属为优势菌属。相比于自然分娩组,在0个月时剖宫产组婴儿粪便标本埃希菌-志贺菌属和肠杆菌属所占比例显著降低(Z=-2.136 4,P=0.032 7;Z=-2.940 8,P=0.003 3),克雷伯菌属和罗氏菌属所占比例显著升高(Z=-2.642 4,P=0.008 2;Z=-2.299 4,P=0.021 5);6个月时罗氏菌属所占比例显著降低(Z=-2.045 0,P=0.040 9),肠球菌属所占比例显著升高(Z=-2.109 2,P=0.034 9)。结论不同分娩方式下的婴儿肠道菌群的构成存在显著差异。  相似文献

树木叶片的水力效率和安全性会对水分条件的改变做出一定的响应, 进而影响树木的生长和分布, 然而叶导水率(Kleaf)和叶水力脆弱性(P50)对不同水分条件的响应模式及其影响因素尚不清楚。该研究选取了晋西北关帝山和黑茶山两种水分条件下的8种树种, 测量其水力性状、叶片导管和形态性状, 比较两地不同树种的KleafP50的变化, 分析叶片水力效率和安全性之间的权衡关系, 并探讨叶片水力性状在不同树种及水分条件下的响应模式及其驱动因素。结果表明: 对同一树种而言, 湿润的关帝山叶最大导水率(Kmax)和P50均高于干旱的黑茶山; 对同一地区而言, 从在高水分条件下生长的树种到在易干旱环境生长的树种, KmaxP50均逐渐下降。KmaxP50、膨压丧失点水势(TLP)之间均存在显著相关关系。两地叶片P50与导管密度、导管塌陷预测值((t/b)3)、叶片厚度、比叶质量显著正相关, 与导管直径、叶面积显著负相关, 不同树种的KleafP50与叶导管性状的关系大于叶形态性状。同一树种的关帝山到黑茶山P50变化量(δP50)与比叶质量和叶干物质含量在两地的变化量显著正相关, 同一树种δP50与叶形态性状变化量的关系大于与叶导管性状的。以上结果表明: 随着水分条件变差, 叶片水力效率降低, 水力安全性提高, 不同树种叶片水力效率与安全性之间存在一定的权衡关系, 不同树种叶水力性状的差别受叶导管性状影响的程度大于受叶形态性状的影响, 同一树种叶水力安全性对水分条件变化的响应主要依靠叶形态性状的驱动, 树木在提高自身叶水力安全的同时增加了叶构建的碳投资。  相似文献

蒙古黄芪病程相关蛋白(Astragalus membranaceus pathogenesis-related protein-10, AmPR-10)具有核酸酶活性,对黄芪生长发育及抗病机制具有重要意义。但从天然黄芪中提取蛋白质的传统方法成本较高,蛋白得率较少。研究首次利用大肠杆菌表达体系对AmPR-10进行可溶性外源表达,构建了3种重组子:①以pET28a为载体构建pET28a-AmPR-10;②以pET30a为载体构建pET30a-AmPR-10;③以pET30a为载体、大肠杆菌分子伴侣skp修饰构建pET30a-skp-AmPR-10。通过SDS-PAGE分析,目的蛋白可溶性表达量比较结果是:pET30a-skp-AmPR-10 > pET30a-AmPR-10 > pET28a-AmPR-10。由蛋白质三级结构模拟分析发现,载体pET-30a上的标签S-tag通过影响AmPR-10局部α螺旋构象而提高目标蛋白的可溶性表达,分子伴侣skp通过提高融合蛋白整体α螺旋比例进一步提升目标蛋白可溶性表达量。研究解决了目前从天然黄芪中提取蛋白操作复杂、提取量小的问题。同时,经测定,目的蛋白具有核酸酶活性,为外源AmPR-10相关活性研究提供了依据。  相似文献

[目的]探讨片突菱纹叶蝉Hishimonus lamellatus Cai et Kuo不同种群中枣疯植原体与沃尔巴克氏体Wolbachia的感染情况和Wolbachia在不同器官组织分布,明确枣园菱纹叶蝉中Wolbachia的感染类型和分类地位,为研究Wolbachia感染对枣疯植原体潜在介体叶蝉生物学及生态学影响奠定基础.[方法]通过枣疯植原体和Wolbachia的基因特异性引物对片突菱纹叶蝉田间自然种群和实验室种群进行分子检测和鉴定.[结果]田间采集的片突菱纹叶蝉成虫植原体感染率在55%-61%之间,而Wolbachia感染率为3%-4%.田间采集的片突菱纹叶蝉自然种群经室内饲养,在1-4龄若虫中检测到Wolbachia,2-5龄若虫中检测到了植原体.片突菱纹叶蝉实验室饲养无植原体种群在其卵巢、卵和若虫中发现感染Wolbachia,在其唾液腺和消化道也检测到了Wolbachia,感染率在58%-100%之间.基于Wolbachia的wsp基因构建系统发育树,发现片突菱纹叶蝉体内的2个Wolbachia株系同属于B大组,但不同于B大组其他株系,属于新株系wLam1和wLam2.[结论]片突菱纹叶蝉成虫采自田间种群可以感染枣疯植原体和Wolbachia,无植原体叶蝉实验室饲养种群成虫感染Wolbachia显著高于田间种群,片突菱纹叶蝉体内2个Wolbachia株系属于B大组.这一研究结果为Wolbachia作为介体叶蝉生物防治剂进一步利用提供了基础信息.  相似文献

The marine mammalian Indo-Pacific humpback dolphin, once widely lived in waters of the Indian to western Pacific oceans, has become an endangered species. The individual number of this dolphin has significantly declined in recent decades, which raises the concern of extinction. Direct concentration on laboratorial conservation of the genetic and cell resources should be paid to this marine species. Here, we report the successful derivation of cell lines form the skin of Indo-Pacific humpback dolphin. The cell cultures displayed the characteristics of fibroblast in morphology and grew rapidly at early passages, but showed obvious growth arrest at higher passages. The karyotype of the cells consisted of 42 autosomes and sex chromosomes X and Y. The immortalized cell lines obtained by forced expression of the SV40 large T-antigen were capable of proliferation at high rate in long-term culture. Immortalization and long-term culture did not cause cytogenetically observable abnormality in the karyotype. The cell type of the primary cultures and immortalized cell lines were further characterized as fibroblasts by the specific expression of vimentin. Gene transfer experiments showed that exogenetic genes could be efficiently delivered into the cells by both plasmid transfection and lentivirus infection. The cells derived from the skin of the Indo-Pacific humpback dolphin may serve as a useful in vitro system for studies on the effects of environmental pollutants and pathogens in habitats on the dolphin animals. More importantly, because of their high proliferation rate and susceptibility to lentivirus, these cells are potential ideal materials for generation of induced pluripotent stem cells.  相似文献

Abstract

A new class of 5-halo (or methoxy)-6-alkoxy (azido or hydroxy)-5,6-dihydro-2′,3′-didehydro-3′-deoxythymidines (4–17) were investigated as potential anti-AIDS drugs. These 5,6-dihydro derivatives, which are also potential prodmgs of 2′,3′-didehydro-3′-deoxythymidine (D4T) were designed to have properties which would enhance their duration of action, lipophilicity and cephalic delivery to the central nervous system. The 5,6-dihydro derivatives of D4T (4–15), which differ in configuration at the C-5 and C-6 positions, were synthesized by the regiospecific addition of XR (X = Br, Cl, I; R = OMe, OEt, N3, OH) to the 5,6-olefinic bond of D4T. These 5,6-disubstituted-5,6-dihydro analogs of D4T are more lipophilic (P = 0.70 – 4.0 range) than D4T (P = 0.12) and are stable to E. coli thymidine phosphorylase. Regeneration of the 5,6-olefinic bond to give D4T, upon incubation of the 5-bromo- and 5-iodo-6-methoxy-5,6-dihydro derivatives (6, 7, 10, 11) with glutathione or a mouse liver soluble enzyme fraction, was extensive (50–95%). The most potent anti-HIV-1 agents, 5-iodo-6-methoxy (10, 11), 5-bromo-6-azido (14, 15) and 5-methoxy-6-hydroxy (16, 17) derivatives of D4T, exhibited anti-HIV activities comparable to D4T.

Lipopolysaccharide (LPS), also known as endotoxin, is the primary trigger of sepsis, which is associated with high mortality in patients. No therapeutic agents are currently efficacious enough to protect patients from sepsis characterized by LPS-mediated tissue damage and organ failure. Previously, a phosvitin-derived peptide, Pt5, which consists of the C-terminal 55 residues of zebrafish phosvitin, has been shown to function as an antibacterial agent. In this study, we have generated six mutants by site-directed mutagenesis based on the sequence of Pt5, and found that one of the six mutants, Pt5e, showed the strongest bactericidal activities against Escherichia coli and Staphylococcus aureus. We then demonstrated that Pt5e was able to bind to LPS and lipoteichoic acid (LTA). More importantly, we showed that Pt5e significantly inhibited LPS-induced tumor-necrosis factor (TNF)-α and interleukin (IL)-1β release from murine RAW264.7 cells and considerably reduced serum TNF-α and IL-1β levels in mice. Additionally, Pt5e protected the liver from damage by LPS, and remarkably promoted the survival rate of the endotoxemia mice. Furthermore, Pt5e displayed no cytotoxicity to murine RAW264.7 macrophages and no hemolytic activity toward human red blood cells. These data together indicate that Pt5e is an endotoxin-neutralizing agent with a therapeutic potential in clinical treatment of LPS-induced sepsis.  相似文献

Abstract

Background: Despite the in vitro and in vivo evidence, studies are limited in evaluating whether chemokines are potential inflammatory mediators in response to air pollution exposure in humans.

Methods: We conducted a panel study coinciding with the Beijing Olympics, when temporary air pollution controls were implemented. We measured a suite of serum chemokines among healthy adults before, during and after the Olympics, respectively. Linear mixed-effect models were used to evaluate changes in chemokine levels over the three time periods.

Results: In response to the 50% drop in air pollution levels during the games, levels of RANTES, MCP-2, and TARC decreased by 25.8%, 20.9% and 35.3%, respectively (p?<?0.001) from pre-Olympics, and then increased by 45.8%, 34.9% and 61.5%, respectively (p?<?0.001) after the games when air pollution levels went up again. Similar patterns were observed in subgroup analyses by sex, age, smoking and body mass index. GRO-α and IL-8 decreased significantly during the games (22.5% and 30.4%), and increased non-significantly after the games. Eotaxin-1 only increased significantly from during- to post-games.

Conclusions: The strongest associations with air pollution levels were observed among RANTES, TARC and MCP-2. Those chemokines may play important roles in the air pollution-induced inflammatory pathway.  相似文献

Shewanella oneidensis is able to respire on a variety of organic and inorganic substrates, including nitrate and nitrite. Conversion of nitrate to nitrite and nitrite to ammonium is catalysed by periplasmic nitrate and nitrite reductases (NAP and NRF) respectively. Global regulator Crp (c yclic AMP r eceptor p rotein) is essential for growth of S. oneidensis on both nitrate and nitrite. In this study, we discovered that crp mutants are not only severely deficient in nitrate or nitrite respiration, but are also hypersensitive to nitrite. This hypersusceptibility phenotype is independent of nitrite respiration. Using random transposon mutagenesis, we obtained 73 Δcrp suppressor strains resistant to nitrite. Transposon insertion sites in 24 suppressor strains were exclusively mapped in the region upstream of the cyd operon encoding a cytochrome bd oxidase, resulting in expression of the operon now driven by a Crp‐independent promoter. Further investigation indicated that the promoter in suppressor strains comes from the transposon. Mutational analysis of the cydB gene (encoding the essential subunit II of the bd oxidase) confirmed that the cytochrome bd oxidase confers nitrite resistance to S. oneidensis.  相似文献
