We isolated and characterized 12 microsatellite loci for the North American medicinal leech, Macrobdella decora. Macrobdella decora is abundant in central and northern North American freshwater systems. The resulting microsatellite library, the first for any species of leech or any clitellate annelid, demonstrates the efficacy of this type of analysis on unexploited leech populations and should serve as a baseline for comparison with other species, notably the commercially important and threatened European medicinal leech, Hirudo medicinalis.  相似文献

Abstract Biomass increase, C and N content, C2H2 reduction, percentage dry weight and chlorophyll a/b ratios were determined for clones of Azolla caroliniana Willd., A. filiculoides Lam., A. mexicana Presl., and A. pinnata R. Br. as a function of nutrient solution, pH, temperature, photoperiod, and light intensity in controlled environment studies. These studies were supplemented by a glasshouse study. Under a 16 h, 26°C day at a light intensity of 200 μmol m?2 s?1 and an 8 h, 19° C dark period, there was no significant difference in the growth rates of the individual species on the five nutrient solutions employed. Growth was comparable from pH 5 to pH 8, but decreased at pH 9. Using the same photoperiod and light intensity but constant growth temperatures of 15–40°C, at 5°C intervals, the individual species exhibited maximum growth, nitro-genase (N2ase) activity and N content at either 25° or 30°C. There was no difference in the temperature optima at pH 6 and pH 8. The tolerance of the individual species to elevated temperature was indicated to be A. mexicana> A. pinnata> A. caroliniana> A.filiculoides. At the optimum temperature, growth rates increased with increasing photoperiod at both pH 6 and pH 8 but N2ase activity was usually highest at a 16 h light period. At photon flux densities of 100, 200, 400 and 600 μmol m?2 s?1, during a 16 h light period and optimum growth temperature of the individual species, N2ase activity was saturated at less than 200 μmol m?2 s?1 and growth at 400 μmol m?2 s?1.No interacting effects of light and pH were noted for any species, nor were light intensities up to 1700 μmol m?2 s?1 detrimental to the growth rate or N content of any species in a 5 week glasshouse study with a natural 14.5 h light period and a constant temperature of 27.5°C. Using the optimum growth temperature, a 16 h light period, and a photon flux density of at least 400 μmol m?2 s?1, the Azolla species all doubled their biomass in 2 days or less and contained 5–6% N on a dry weight basis.  相似文献

A novel microsporidian parasite is described, which infects the crustacean host Gammarus duebeni. The parasite was transovarially transmitted and feminised host offspring. The life cycle was monomorphic with three stages. Meronts were found in host embryos, juveniles, and in the gonadal tissue of adults. Sporoblasts and spores were restricted to the gonad. Sporogony was disporoblastic giving rise to paired sporoblasts, which then differentiated to form spores. Spores were not found in regular groupings and there was no interfacial envelope. Spores were approximately 3.78 x 1.22 microns and had a thin exospore wall, a short polar filament, and an unusual granular polaroplast. All life cycle stages were diplokaryotic. A region from the parasite small subunit ribosomal RNA gene was amplified and sequenced. Phylogenetic analysis based on these data places the parasite within the genus Nosema. We have named the species Nosema granulosis based on the structure of the polaroplast.  相似文献

The order Thysanoptera (Paraneoptera), commonly known as thrips, displays a wide range of behaviours, and includes several pest species. The classification and suggested relationships among these insects remain morphologically based, and have never been evaluated formally with a comprehensive molecular phylogenetic analysis. We tested the monophyly of the suborders, included families and the recognized subfamilies, and investigated their relationships. Phylogenies were reconstructed based upon 5299 bp from five genetic loci: 18S ribosomal DNA, 28S ribosomal DNA, Histone 3, Tubulin‐alpha I and cytochrome oxidase c subunit I. Ninety‐nine thrips species from seven of the nine families, all six subfamilies and 70 genera were sequenced. Maximum parsimony, maximum likelihood and Bayesian analyses all strongly support a monophyletic Tubulifera and Terebrantia. The families Phlaeothripidae, Aeolothripidae, Melanthripidae and Thripidae are recovered as monophyletic. The relationship of Aeolothripidae and Merothripidae to the rest of Terebrantia is equivocal. Molecular data support previous suggestions that Aeolothripidae or Merothripidae could be a sister to the rest of Terebrantia. Four of the six subfamilies are recovered as monophyletic. The two largest subfamilies, Phlaeothripinae and Thripinae, are paraphyletic and require further study to understand their internal relationships.  相似文献

Anthropogenic soil erosion severely affects land ecosystems by reducing plant productivity and stimulating horizontal carbon and nitrogen movement at the surface. Climate warming may accelerate soil erosion by altering soil temperature, moisture, and vegetation coverage. However, no experiments have been carried out to quantify soil erosion with warming. In a long‐term field experiment, we explored how annual clipping for biofuel feedstock production and warming caused soil erosion and accompanying carbon and nitrogen losses in tallgrass prairie in Oklahoma, USA. We measured relative changes in soil surface elevation between clipped and unclipped plots with or without experimental warming. Our results show that average relative erosion depth caused by clipping was 1.65±0.09 and 0.54±0.08 mm yr?1, respectively, in warmed and control plots from November 21, 1999 to April 21, 2009. The soil erosion rate was 2148±121 g m?2 yr?1 in the warmed plots and 693±113 g m?2 yr?1 in the control plots. Soil organic carbon was lost at a rate of 69.6±5.6 g m?2 yr?1 in the warmed plots and 22.5±2.7 g m?2 yr?1 in the control plots. Total nitrogen was lost at a rate of 4.6±0.4 g m?2 yr?1 in the warmed plots and 1.4±0.1 g m?2 yr?2 in the control plots. The amount of carbon and nitrogen loss caused by clipping is equivalent to or even larger than changes caused by global change factors such as warming and rising atmospheric CO2 concentration. In addition, soil erosion rates were significantly correlated with clipping‐induced changes in soil moisture. Our results suggest that clipping for biofuel harvest results in significant soil erosion and accompanying losses of soil carbon and nitrogen, which is aggravated by warming.  相似文献

Magellanic penguin populations in the Falkland Islands may have decreased over the past decade. The post-breeding migration may be the period in which the birds are most vulnerable. To investigate this we equipped ten Magellanic Penguins after their moult at Seal Bay (51̀38'S, 58̀03'W), East Falkland, with satellite transmitters. The movements of the penguins were tracked for between 15 and 99 days until transmission ceased. All birds initially migrated to the northwest. However, four birds entered Argentinean coastal waters, then headed northeast following the coastline, The other penguins remained offshore, but also changed to a northeasterly course. Two birds migrated beyond 36̀S with a maximum distance to the colony of more than 1800 km, while the minimum distance travelled was up to 2700 km. Initially, the birds migrated quickly but their speed was reduced and became more variable as time progressed. A high concentration of positional fixes, associated with low travelling speeds, indicated at least four different areas where birds were assumed to forage efficiently. The relevance of these areas is discussed with respect to the diet of Magellanic Penguins and possible interactions with human activities.  相似文献
