3-(Hetero)aryl substituted 7-azaindoles possessing multikinase inhibitor activity are readily accessed in a one-pot Masuda borylation-Suzuki coupling sequence. Several promising derivatives were identified as apoptosis inducers and, emphasizing the multikinase inhibition potential, as sphingosine kinase 2 inhibitors. Our measurements provide additional insights into the structure-activity relationship of meriolin derivatives, suggesting derivatives bearing a pyridine moiety with amino groups in 2-position as most active anticancer compounds and thus as highly promising candidates for future in vivo studies.  相似文献

Illumination of biological membranes with visible light in the presence of membrane-active sensitizers (e.g. rose bengal) is known to inactivate transport proteins such as ion channels and ion pumps. In some cases, however, illumination gives rise to an activation of transport. This is shown here for ion channels formed by alamethicin in lipid membranes, and for porin channels, which were isolated from the outer membrane of E. coli (OmpC) and from the outer membrane of mitochondria (VDAC) and were reconstituted in lipid membranes. An activation (in the form of an increased conductance) was also observed in the presence of the cation carriers valinomycin and nonactin. The activation phenomena were only present, if the membranes were made from lipids containing unsaturated double bonds. Activation was reduced in the presence of the antioxidant vitamin E.We suggest that the activation of the different transport systems has a common physical basis, namely an increase of the dielectric constant, εm, of the membrane interior by the presence of polar oxidation products of photodynamically induced lipid peroxidation. Experimental evidence for an enhanced dielectric constant was obtained from the finding of a light-induced increase of the membrane capacitance in the presence of rose bengal.  相似文献

The mRNA codon in the ribosomal A-site is recognized by aminoacyl-tRNA (aa-tRNA) in a ternary complex with elongation factor Tu (EF-Tu) and GTP. Here we report the 13 A resolution three-dimensional reconstruction determined by cryo-electron microscopy of the kirromycin-stalled codon-recognition complex. The structure of the ternary complex is distorted by binding of the tRNA anticodon arm in the decoding center. The aa-tRNA interacts with 16S rRNA, helix 69 of 23S rRNA and proteins S12 and L11, while the sarcin-ricin loop of 23S rRNA contacts domain 1 of EF-Tu near the nucleotide-binding pocket. These results provide a detailed snapshot view of an important functional state of the ribosome and suggest mechanisms of decoding and GTPase activation.  相似文献

Background

Rare mutations of the low-density lipoprotein receptor gene (LDLR) cause familial hypercholesterolemia, which increases the risk for coronary artery disease (CAD). Less is known about the implications of common genetic variation in the LDLR gene regarding the variability of cholesterol levels and risk of CAD.

Methods

Imputed genotype data at the LDLR locus on 1 644 individuals of a population-based sample were explored for association with LDL-C level. Replication of association with LDL-C level was sought for the most significant single nucleotide polymorphism (SNP) within the LDLR gene in three European samples comprising 6 642 adults and 533 children. Association of this SNP with CAD was examined in six case-control studies involving more than 15 000 individuals.

Findings

Each copy of the minor T allele of SNP rs2228671 within LDLR (frequency 11%) was related to a decrease of LDL-C levels by 0.19 mmol/L (95% confidence interval (CI) [0.13–0.24] mmol/L, p = 1.5×10−10). This association with LDL-C was uniformly found in children, men, and women of all samples studied. In parallel, the T allele of rs2228671 was associated with a significantly lower risk of CAD (Odds Ratio per copy of the T allele: 0.82, 95% CI [0.76–0.89], p = 2.1×10−7). Adjustment for LDL-C levels by logistic regression or Mendelian Randomisation models abolished the significant association between rs2228671 with CAD completely, indicating a functional link between the genetic variant at the LDLR gene locus, change in LDL-C and risk of CAD.

Conclusion

A common variant at the LDLR gene locus affects LDL-C levels and, thereby, the risk for CAD.  相似文献

The elongation factors (EF) Tu and G and initiation factor 2 (IF2) from bacteria are multidomain GTPases with essential functions in the elongation and initiation phases of translation. They bind to the same site on the ribosome where their low intrinsic GTPase activities are strongly stimulated. The factors differ fundamentally from each other, and from the majority of GTPases, in the mechanisms of GTPase control, the timing of Pi release, and the functional role of GTP hydrolysis. EF-Tu x GTP forms a ternary complex with aminoacyl-tRNA, which binds to the ribosome. Only when a matching codon is recognized, the GTPase of EF-Tu is stimulated, rapid GTP hydrolysis and Pi release take place, EF-Tu rearranges to the GDP form, and aminoacyl-tRNA is released into the peptidyltransferase center. In contrast, EF-G hydrolyzes GTP immediately upon binding to the ribosome, stimulated by ribosomal protein L7/12. Subsequent translocation is driven by the slow dissociation of Pi, suggesting a mechano-chemical function of EF-G. Accordingly, different conformations of EF-G on the ribosome are revealed by cryo-electron microscopy. GTP hydrolysis by IF2 is triggered upon formation of the 70S initiation complex, and the dissociation of Pi and/or IF2 follows a rearrangement of the ribosome into the elongation-competent state.  相似文献

Grab samples of shallow (5–35 m) marine benthic sediments and epi- and infaunal assemblages were taken from nearshore regions around Casey Station, Windmill Islands, East Antarctica. A hierarchical, spatially nested sampling design was used with locations (kilometres apart), sites (hundreds of metres apart) and plots (tens of metres). Two potentially impacted, polluted locations (adjacent to a sewage outfall and an old garbage tip) were compared with two control locations in an asymmetrical design. Significant differences in assemblages were found between locations and between sites within locations. Significant differences in the abundances of taxa at several taxonomic levels (species, family, order, phylum) were found at all three spatial scales. Significant differences were also detected between the polluted and control locations. Compared with other Antarctic locations, the assemblages were dominated by crustaceans (90–97% of individuals) and there was a paucity of polychaete fauna at the locations sampled. The moss Syntrichia caninervis is the dominant soil bryophyte in a blackbrush (Coleogyne ramosissima) community in the southern Nevada Mojave Desert, with a mean cover of 6.3%. A survey of the 10-ha study site revealed an expressed ramet sex ratio of 14♀ : 1♂ (N = 890), with 85% of ramets not expressing sex over their life span, and an expressed population sex ratio of 40♀ : 2♂ : 1♀♂ (female : male : mixed-sex, N = 89), with 52% of populations not expressing sex. A greater incidence of sex expression was associated with shaded microsites, higher soil moisture content, and taller ramets. Shaded microsites had higher surface soil moisture levels than exposed microsites. In the exposed microhabitat, surface soil moisture was positively correlated with ramet height but not with sex expression. Male ramets and populations were restricted to shaded microhabitats, whereas female ramets and populations were found in both shaded and exposed microhabitats, suggesting gender specialization. The rarity of mature sporophytes, found in 0% of the ramets sampled and in only 3% of the populations, is probably due to the rarity of mixed-sex populations. We hypothesize that mixed-sex populations are rare because of factors relating to male rarity and that the differential cost of sex expression reduces the clonal growth capacity of male individuals.  相似文献
