Abstract. Fat and haematin levels of mature male Glossina morsitans morsitans Westwood were estimated at different times after feeding at temperatures between 15 and 30°C. Flies were kept (largely inactive) in 7.5 × 2.5 cm tubes, or in actograph cages, where flight activity increased with time after feeding. Haematin excretion was modelled as a series of three first order reactions, all with the same rate parameter. The model accounted for > 98% of the variance in mean haematin in each of seven experiments; the rate parameter increased linearly with temperature and activity level. A similar approach was adopted for modelling fat metabolism. The rate coefficients of lipogenesis increased with temperature, and that for lipolysis with temperature, activity level and their interaction. All experiments were analysed simultaneously to provide equations predicting haematin or fat levels for all times, for active or inactive flies, and for temperatures between 15 and 30°C. Haematin exhibited large variations between individuals, but for active flies the expected haematin content at a given time varied little between flies kept at 25 and at 30°C. In inactive flies kept at 25°C, lipogenesis peaked at ≈ 24 h and lipolysis at ≈ 48 h. For active flies the times were 12 and 24 h, respectively; both rates were about twice as high as in inactive flies. Active flies produced (up to 1 mg) more fat out of a given size of blood meal than inactive flies. Curves of fat content against logarithm of haematin content differed little with temperature, and can therefore be useful for comparative studies of field populations of tsetse.  相似文献

  • 1 The feeding cycle for male tsetse flies and the pregnancy cycle for females are used as frameworks for investigating the field behaviour of Glossina morsitans centralis Machado in Zambia, and hence the sampling biases of different capture devices.
  • 2 Flies were collected in the field using hand nets and an electric back pack on foot patrols, and hand nets, hand-catching and an electric screen on landrover patrols. They were analysed for wing-fray, vein-length, chloroform-extractable fat and haematin. In addition, samples of laboratory-bred females killed daily during their second pregnancy cycle were analysed for the last two parameters.
  • 3 The daily flying time of males is estimated from the fat—haematin curve to be about 32 min/day, which is identical to previous estimates for G. morsitans from Tanzania.
  • 4 From the varying numbers and fat content of male flies caught by the different sampling methods during the course of the feeding cycle it is concluded that both haematin levels and fat reserves influence fly behaviour.
  • 5 Comparison of the frequency distribution of wild-caught and laboratory-bred females along the corrected residual dry weight axis of the pregnancy cycle suggests that the results of combined electric-trap and hand-net sampling reflect the actual frequency distribution in the wild population at least for the first 7 days of the pregnancy cycle; but females in the last 2 days of their pregnancy cycle are apparently unavailable to sampling devices, perhaps because of their reduced flight activity.
  • 6 The very variable percentage female catch (10.9–43.4%) returned by the different sampling methods is discussed in the light of the detailed analysis of the differential availability of male and female flies to each capture device.

Purified chloroplast tRNAs were isolated fromPisum sativum leaves and radioactively labeled at their 3′ end using tRNA nucleotidyl transferase and α32P-labeled CTP. Pea ctDNA was fragmented using a number of restriction endonucleases and hybridized with thein vitro labeled chloroplast tRNAs by DNA transfer method. Genes for tRNAs have been found to be dispersed throughout the chloroplast genome. A closer analysis of the several hybrid regions using recombinant DNA plasmids have shown that tRNA genes are localized in the chloroplast genome in both single and multiple arrangements. Two dimensional gel electrophoresis of total ct tRNA have identified 36 spots. All of them have been found to hybridize withPisum sativum ctDNA. Using recombinant clones, 30 of the tRNA spots have been mapped inPisum sativum ctDNA.  相似文献

As the number of non-synonymous single nucleotide polymorphisms (nsSNPs) identified through whole-exome/whole-genome sequencing programs increases, researchers and clinicians are becoming increasingly reliant upon computational prediction algorithms designed to prioritize potential functional variants for further study. A large proportion of existing prediction algorithms are ‘disease agnostic’ but are nevertheless quite capable of predicting when a mutation is likely to be deleterious. However, most clinical and research applications of these algorithms relate to specific diseases and would therefore benefit from an approach that discriminates between functional variants specifically related to that disease from those which are not. In a whole-exome/whole-genome sequencing context, such an approach could substantially reduce the number of false positive candidate mutations. Here, we test this postulate by incorporating a disease-specific weighting scheme into the Functional Analysis through Hidden Markov Models (FATHMM) algorithm. When compared to traditional prediction algorithms, we observed an overall reduction in the number of false positives identified using a disease-specific approach to functional prediction across 17 distinct disease concepts/categories. Our results illustrate the potential benefits of making disease-specific predictions when prioritizing candidate variants in relation to specific diseases. We characterized β1 integrin subunit expression on three different cultures of benign human nevomelanocytes (NMC) and on four different cell cultures of human dysplastic nevus (DN) cells by flow cytometry analysis and examined their role in mediating cell spreading and migration on collagen type IV (CN IV) and laminin (LN) coated substrates by using a quantitative video image analysis system. The seven human NMC and DNC cultures expressed heterogeneous levels of β1, α2, α3 and α6 integrin subunits. Image analysis showed that a significant increase (P<0.001) in cell spreading and migration of the DN cells was induced on increasing coating concentrations of CN IV and LN. However, the NMC did not show an increase in cell spreading or migration on these substrates when compared to the substrates coated with denatured BSA only. The CN IV-induced cell spreading of the DN cells was significantly inhibited by anti-β1 mAb (AIIB2), anti-α2 mAb (P1E6), or anti-α3 mAb (P1B5), but not by mAb against α6 integrin subunit (GoH3). The DN cell spreading on LN was not significantly inhibited by these mAbs. In contrast, the migration of the DN on CN IV and LN was significantly inhibited by anti-β1 mAb, anti-α2 mAb, anti-α3 mAb and anti-α6 mAb. These data suggest that the α2 and α3 subunit are important for cell spreading of the DN on CN IV, although they are less important in cell spreading on the extracellular matrix component LN. The α2, α3 and α6 integrin subunits are important for the migration of DN cells on both CN IV and LN.  相似文献

Background  

In recent years, several new hypotheses on phylogenetic relations among arthropods have been proposed on the basis of DNA sequences. One of the challenged hypotheses is the monophyly of hexapods. This discussion originated from analyses based on mitochondrial DNA datasets that, due to an unusual positioning of Collembola, suggested that the hexapod body plan evolved at least twice.   • 1 Activity cycles of female tsetse (Glossina palpalis palpalis Robineau-Desvoidy) in the field are contrasted with those of males, using mark-release-recapture data from continuous biconical trap samples over a period of 80 days in two villages in Ivory Coast.
  • 2 Variability in recapture rates was examined using the techniques of autocorrelation and spectral analysis. In order to do this a two-dimensional diffusion model incorporating both mortality and emigration was first fitted to corrected recapture rate data to produce a trend line, deviations from which were subjected to the analysis.
  • 3 The autocorrelations for the data for males suggest considerable variability in activity, presumably associated with feeding, around a mean period of about 4 days in one village and seven in the other.
  • 4 Data for females show a strong regular periodicity of about 9–10 days, reflecting the pregnancy cycle, and shorter, more variable periodicities probably corresponding to feeding.

Evidence from the comparative biology of living birds and mammals is used to address the question ‘which came first, flight or endothermy?’. Birds and mammals have evolved different solutions to the problems of high energy flow demanded by endothermy. The heavy apparatus needed for processing food to allow the rapid assimilation of energy is housed in the head of mammals, but low down in the bird's body. The primitive inefficient tidal-flow system of ventilation is simply enlarged in mammals, but is replaced in birds by a lighter uni-flow system through air sacs and parabronchi. Birds avoid the weight problems associated with the mammalian systems of viviparity and lactation by nourishing their young with large quantities of yolk within the egg and an unprocessed diet after hatching. The apparent adaptedness for flight of the avian systems suggests that in the animals ancestral to birds the adaptations for high energy flow were constrained from the start by the need for aerodynamic stability, i.e. flight was initiated before endothermy. The implications of this conclusion for the origin of flight and feathers are discussed.  相似文献
