Combining measurements of electric potential and pH with such of chlorophyll fluorescence and leaf gas exchange showed heat stimulation to evoke an electrical signal (propagation speed: 3–5 mm s−1) that travelled through the leaf while reducing the net CO2 uptake rate and the photochemical quantum yield of both photosystems (PS). Two-dimensional imaging analysis of the chlorophyll fluorescence signal of PS II revealed that the yield reduction spread basipetally via the veins through the leaf at a speed of 1.6 ± 0.3 mm s−1 while the propagation speed in the intervein region was c. 50 times slower. Propagation of the signal through the veins was confirmed because PS I, which is present in the bundle sheath cells around the leaf vessels, was affected first. Hence, spreading of the signal along the veins represents a path with higher travelling speed than within the intervein region of the leaf lamina. Upon the electrical signal, cytoplasmic pH decreased transiently from 7.0 to 6.4, while apoplastic pH increased transiently from 4.5 to 5.2. Moreover, photochemical quantum yield of isolated chloroplasts was strongly affected by pH changes in the surrounding medium, indicating a putative direct influence of electrical signalling via changes of cytosolic pH on leaf photosynthesis.  相似文献

In plants, the enzymes for cysteine synthesis serine acetyltransferase (SAT) and O-acetylserine-(thiol)-lyase (OASTL) are present in the cytosol, plastids and mitochondria. However, it is still not clearly resolved to what extent the different compartments are involved in cysteine biosynthesis and how compartmentation influences the regulation of this biosynthetic pathway. To address these questions, we analysed Arabidopsis thaliana T-DNA insertion mutants for cytosolic and plastidic SAT isoforms. In addition, the subcellular distribution of enzyme activities and metabolite concentrations implicated in cysteine and glutathione biosynthesis were revealed by non-aqueous fractionation (NAF). We demonstrate that cytosolic SERAT1.1 and plastidic SERAT2.1 do not contribute to cysteine biosynthesis to a major extent, but may function to overcome transport limitations of O-acetylserine (OAS) from mitochondria. Substantiated by predominantly cytosolic cysteine pools, considerable amounts of sulphide and presence of OAS in the cytosol, our results suggest that the cytosol is the principal site for cysteine biosynthesis. Subcellular metabolite analysis further indicated efficient transport of cysteine, γ -glutamylcysteine and glutathione between the compartments. With respect to regulation of cysteine biosynthesis, estimation of subcellular OAS and sulphide concentrations established that OAS is limiting for cysteine biosynthesis and that SAT is mainly present bound in the cysteine–synthase complex.  相似文献

Schoch, R.R. & Witzmann, F. 2011: Cranial morphology of the plagiosaurid Gerrothorax pulcherrimus as an extreme example of evolutionary stasis. Lethaia, Vol. 45, pp. 371–385. The plagiosaurid Gerrothorax pulcherrimus from the Triassic of Greenland and Germany is represented by skulls ranging from 4 to 12 cm in length and sheds light on ontogeny, individual variation, and variation in time and space. Ontogeny was remarkably stable in G. pulcherrimus, with the smallest known specimens resembling the adults closely in most features. A true ontogenetic change is evident in the ornament of dermal bones, in that the smallest specimens have ridges whereas in the successively larger ones, pustules spread over increasingly larger areas. The skull becomes proportionally longer, and the adductor chambers relatively narrower. The positive allometry of both the orbits and the interpterygoid vacuities suggests that the eye supporting musculature – rather than the jaw adductors – increased proportionally during growth. Individual, not age‐related variation in the dermal skull roof affects partial fusion of parietals, presence and extent of the interfrontoparietal, and the morphological pattern of the posterior skull table. The ventral surface of the basal plate of the parasphenoid ranges from smooth over poorly to heavily ornamented or dentigerous. Considering the impressive longevity of more than 35 Myr, the morphological changes of G. pulcherrimus are minor. Our ecological interpretation for G. pulcherrimus is that it relied on the permanent presence of water, but was flexible with respect to the size and nature of the water body as well as to changes in salinity. The unparalleled extent of evolutionary stasis may therefore be based on the ecological flexibility of this morphologically so tightly constrained temnospondyl. □Ecological flexibility, ontogeny,Temnospondyli, Triassic, variation.  相似文献

RNA interference (RNAi) is a natural mechanism for regulating gene expression, which exists in plants, invertebrates, and mammals. We investigated whether non-viral infusion of short interfering RNA (siRNA) by the intracerebroventricular route would enable a sequence-specific gene knockdown in the mouse brain and whether the knockdown translates into disease-relevant behavioral changes. Initially, we targeted enhanced green fluorescent protein (EGFP) in mice overexpressing EGFP. A selective knockdown of both EGFP protein and mRNA was observed throughout the brain, with lesser down-regulation in regions distal to the infusion site. We then targeted endogenous genes, encoding the dopamine (DAT) and serotonin transporters (SERT). DAT-siRNA infusion in adult mice produced a significant down-regulation of DAT mRNA and protein and elicited hyperlocomotion similar, but delayed, to that produced on infusion of GBR-12909, a potent and selective DAT inhibitor. Similarly, SERT-siRNA infusion resulted in significant knockdown of SERT mRNA and protein and elicited reduced immobility in the forced swim test similar to that obtained on infusion of citalopram, a very selective and potent SSRI. Application of this non-viral RNAi approach may accelerate target validation for neuropsychiatric disorders that involve a complex interplay of gene(s) from various brain regions.  相似文献

Determining the geographical range of invasive species is an important component of formulating effective management strategies. In the absence of detailed distributional data, species distribution models can provide estimates of an invasion range and increase our understanding of the ecological processes acting at various spatial scales. We used two complementary approaches to evaluate the influence of historical and environmental factors in shaping the distribution of the Argentine ant ( Linepithema humile ), a widespread, highly invasive species native to South America. Occurrence data were combined with environmental data at incremental spatial scales (extent and resolution) to predict the suitable range of the ant invasion using ecological niche models. In addition, we also used a spread model that simulated the jump dispersal of the species to identify the most plausible scenarios of arrival of L. humile in the NE Iberian Peninsula at local scales. Based on the results of both modelling practices, we suggest that L. humile might have reached its maximum geographic range at regional scales in the NE Iberian Peninsula. However, the species does not appear in equilibrium with the environment at small spatial scales, and further expansions are expected along coastal and inland localities of the Costa Brava. Long-distance jumps are ultimately responsible for the spread of the Argentine ant in the area. Overall, our study shows the utility of combining niche based models with spread models to understand the dynamics of species' invasions.  相似文献

Until recently the North Atlantic Islands were believed to house only mammals introduced by humans. Recent work has demonstrated that at least the Canary Islands house(d) a native mammal fauna. New data including chromosome numbers, genetic distances and analysis of vocalizations are given for the two extant shrew species, Crocidura canariensis and C. osorio , and their possible sister taxa are evaluated. Evidence is presented for the hypothesis that the two island species originated from two different lineages of the Palaearctic branch of the genus Crocidura. The data support the present status of the Canary Island shrews as local endemics of high conservation priority.  相似文献

The cranial anatomy of the plagiosaurid temnospondyl Plagiosuchus pustuliferus, from the Middle Triassic of Germany, is described in detail on the basis of a newly discovered skull and mandibular material. The highly derived skull is characterized by huge orbitotemporal fenestrae, a reduction of the circumorbital bones – the prefrontal, postfrontal and (probably) postorbital are lost – and the expansion of the jugal to occupy most of the lateral skull margin. Ventrally the extremely long subtemporal vacuities correlate with the elongate adductor fossa of the mandible. The dentition is feebly developed on both skull and mandible. Ossified ?ceratobranchials and ‘branchial denticles’ indicate the presence of open gills clefts in life. The remarkably divergent cranial morphology of P. pustuliferus highlights the extraordinary cranial diversity within the Plagiosauridae, probably unsurpassed within the Temnospondyli. Specific structural aspects of the skull – including an extremely short marginal tooth row, feeble dentition and an elongated chamber for adductor musculature – together with evidence for a hyobranchial skeleton, suggests that P. pustuliferus utilized directed suction feeding for prey capture. © 2009 The Linnean Society of London, Zoological Journal of the Linnean Society, 2009, 155 , 348–373.  相似文献

The temnospondyl Sclerocephalus from the Permo‐Carboniferous of Germany is one of the most completely preserved and most abundant Palaeozoic tetrapods. Here, we review the complete osteology of the genus, based on a range of fully grown specimens housed in public collections. Among the four valid species, Sclerocephalus haeuseri and Sclerocephalus nobilis reached an adult size of well beyond 1 m in length, and had robust postcranial skeletons. In the skull, the exoccipital and sphenethmoid bones were ossified, completing the well‐known ossification sequence in S. haeuseri. Large adults had an elongate trunk and a laterally compressed tail, and some individuals also retained lateral line sulci: features that taken together suggest an aquatic life. The coracoid, pubis, carpals, tarsals, and the bony tail are fully ossified in the largest specimens. The genus Sclerocephalus forms a weakly supported clade nesting firmly at the base of the Stereospondylomorpha, and the close resemblance between Sclerocephalus, Onchiodon, and Eryops is found to be partially based on shared derived states, but is mostly based on symplesiomorphies. Cladistic analysis of 54 characters and 18 taxa finds more support for the Eryopoidea hypothesis (Eryopidae + Stereospondylomorpha) than for the Euskelia hypothesis (Eryopidae + Zatracheidae + Dissorophoidea). This indicates that the large temnospondyls of the Permian and Mesozoic probably formed a natural group, and that the terrestrial adaptations of Eryops and the dissorophoids probably evolved by convergence.  相似文献

Arabidopsis plants were exposed to high light or sulphur depletion alone or in combination for 6 d, and changes of photosynthetic parameters and metabolite abundances were quantified. Photosynthetic electron transport rates (ETRs) of plants exposed to sulphur depletion and high light decreased strongly at day 2 of the acclimation period. After 3 d of treatment, the photosynthetic capacity recovered in plants exposed to the combined stresses, indicating a short recovery time for re‐adjustment of photosynthesis. However, at metabolic level, the stress combination had a profound effect on central metabolic pathways such as the tricarboxylic acid (TCA) cycle, glycolysis, pentose phosphate cycle and large parts of amino acid metabolism. Under these conditions, central metabolites, such as sugars and their phosphates, increased, while sulphur‐containing compounds were decreased. Further differential responses were found for the stress indicator proline accumulating already at day 1 of the high‐light regime, but in combination with sulphur depletion first declined and after a recovery phase reached a delayed elevated level. Other metabolites such as raffinose and putrescine seem to replace proline during the early combinatorial stress response and may act as alternative protectants. Our findings support the notion that plants integrate the selectively sensed stress factors in central metabolism.  相似文献
