ABSTRACT. Fed through a synthetic membrane, 80% of females of the human body louse, Pediculus humanus de Geer, engorged on whole human blood compared to 30% on platelet poor plasma. Both the plasma albumin and small molecular weight components of the cellular fraction seemed to stimulate engorgement by lice. Known haemo-phagostimulants such as adenosine triphosphate and diphosphoglyceric acid when added to plasma, did not replace the small molecular weight components of the cellular fraction as feeding stimuli. SYNOPSIS. On prolonged incubation of ethionine-sensitive (ES) cells of Ochromonas danica in L-ethionine-containing media, growth was resumed by an ethionine-resistant (ER) mutant. Such mutants arise at random and are selected by the ethionine-containing medium. Ethionine resistance is not lost on repeated transfers thru ethionine-less media. ES cells incubated with ethionine form a large posterior vacuole before they disintegrate. Inhibition of reserve substance utilization is suggested to underlie growth inhibition of O. danica by ethionine. In ES cells incubated with ethionine, 14 C uptake from labeled methionine, ethionine or serine is reduced by 65%. In ER cells the decrease in 14 C uptake is 90%. This decrease in uptake of ethionine seems to be how ER O. danica evades growth inhibition by ethionine. 相似文献

A novel, unbiased approach to plant viral disease diagnosis has been developed which requires no a priori knowledge of the host or pathogen. Next-generation sequencing coupled with metagenomic analysis was used to produce large quantities of cDNA sequence in a model system of tomato infected with Pepino mosaic virus . The method was then applied to a sample of Gomphrena globosa infected with an unknown pathogen originally isolated from the flowering plant Liatris spicata . This plant was found to contain a new cucumovirus, for which we suggest the name 'Gayfeather mild mottle virus'. In both cases, the full viral genome was sequenced. This method expedites the entire process of novel virus discovery, identification, viral genome sequencing and, subsequently, the development of more routine assays for new viral pathogens. 相似文献

Abstract. We have applied the recently developed Random Amplified Polymorphic DNA (RAPD) method to produce species-specific, DNA profiles for two sympatric, Venezuelan sandfly species, thought to be the vectors responsible for recent outbreaks of cutaneous and mucocutaneous leishmaniasis in the Andean State of Tachira. Moreover, within the profile, it was possible to identify a diagnostic DNA band for Lu.youngi of 0.32 kb. Abstract. Resistance to human body lice, Pediculus humanus humanus L, induced by feeding on rabbits immunized with an extract of louse gut was studied. The mortality of lice fed on immunized rabbits was 73%, significantly higher than that of lice fed on control rabbits (52%) ( P < 0.01). The proportion of dead nymphs and female lice with ruptured guts was significantly higher in lice fed on immunized rabbits (P<0.01). The size of the bloodmeal was 35% greater in female lice fed on control rabbits than on immunized rabbits. Lice fed on immunized rabbits laid 40% less eggs than those fed on the controls, they also demonstrated a significant decrease in the number of eggs per female over time ( P < 0.01). 86% of the eggs laid by lice fed on immunized animals hatched, compared with 92% hatching of eggs laid by the lice fed on control animals (P< 0.01). With the exception of the first bloodmeal the percentage of hatched eggs which were laid between any two bloodmeals was significantly smaller (p<0.01) in the lice fed on immunized rabbits than in the control group. The first nymphal stage of lice fed on immunized rabbits took an average of 5.2 days to moult to the second stage, compared with 4 days for those fed on control rabbits. 相似文献

ABSTRACT Aversive conditioning (AC) has the potential to temporarily reduce conflicts between humans and black bears (Ursus americanus). From 2002 to 2005, I evaluated the effectiveness of projectiles with varying impact intensities, pepper spray, and chasing on approximately 150 bears in Sequoia National Park. Aversive conditioning was successful in keeping bears that were not food-conditioned from becoming food-conditioned. For the bears that were already food-conditioned, 17 of 29 bears subjected to AC abandoned unwanted behaviors, 6 required continual treatments, and 6 were killed or relocated. Success with food-conditioned bears was highest when AC was applied soon after bears obtained human food. Aversive conditioning was less successful on yearlings than adults. Rubber slugs were slightly more effective than lower impact projectiles. 相似文献

Abstract. The human body louse, Pediculus humanus , showed eighteen midgut proteins ranging between 12 and 117 kDa, when analysed by SDS-PAGE electrophoresis. Seven of them (12 kDa, 17 kDa, 29 kDa, 35 kDa, 40 kDa, 55 kDa and 97 kDa) were major bands based on their intensity of staining. The immunization of rabbits with a midgut extract elicited the production of protective polyclonal antibodies. These antibodies reacted strongly with all major midgut proteins as well as with 63 kDa and 117 kDa proteins when tested by the Western blot technique. The analysis of the proteins revealed that the 12 kDa, 25 kDa, 29 kDa, 35 kDa, 45 kDa, 87 kDa and 97 kDa proteins are glycosylated and none of them contained a lipid moiety. By electroelution, the proteins of 35 kDa and 63 kDa were purified. On trypsinization, the proteins of 35 kDa and 63 kDa produced four major fragments (F, F2 , F3 , and F4 ) when resolved on a 18% SDS-PAGE.
