The expression of O- and N-glycan chains during the enterocytic differentiation of HT-29 cells was followed using L-[63H]-fucose and D-[6-3H]-glucosamine radiolabeling. Whatever the cell population, i.e., differentiated or undifferentiated, the incorporation of radiolabeled sugars into glycoproteins was similar. However, differences among these two cell populations were noted when the ratio [3H]-glucosamine/[3H]-galactosamine and the sensitivity of glycopeptides to mild alkaline treatment were followed. From these data, we could conclude that there is a shift in the high molecular weight glycopeptides during the differentiation of HT-29 cells meaning an increase of O-linked glycopeptides correlated with a decrease of N-linked forms.  相似文献

In vitro effects of thyroxine on erythrocyte deformability and mechanical fragility were observed. Deformability of erythrocytes was improved in a dose dependent manner by thyroxine. Mechanical hemolysis was found to be lower if thyroxine was included in erythrocyte suspensions at concentrations close to the physiological levels (10(-9)M). These changes might be related to the alterations of intracellular calcium concentration, as in the erythrocyte suspensions containing 10(-9)M thyroxine, intracellular calcium concentration was found to be 30 times lower than the control suspensions which did not contain thyroxine. Thyroxine also reduced the mechanical hemolysis ratio in calcium loaded cells. These observations suggest that thyroxine might play some role in the regulation of the mechanical properties of erythrocytes which might be mediated via the effects on calcium metabolism.  相似文献

在农业生产中长期使用化学农药已对环境和生态平衡造成一定破坏作用,同时有不少害虫也逐渐产生抗药性从而引起某些害虫的大流行,给农业生产带来巨大损失。应用苏云金杆菌杀虫蛋白基因(Bt基因)可构建具有抗虫作用的抗虫工程菌,这样通过拌种或植物叶面喷雾可达到快速、经济、有效的防治虫害的目的。国际上抗虫工程菌研究应用很快,如美国将BI基因转入到一种正常情况下定居在植物组织中的棒杆菌,将这种工程菌拌玉米种子,这样随植物生长该菌在植物体内大量繁殖,当玉米螟在茎和叶取食时,即因食用表达苏云金杆菌毒蛋白的工程菌而死亡。 田颖川等已克隆了苏云金芽孢杆菌内毒素基因CryIA(b)和CryIA(c)。本文将Bt基因CryIA(c)插入到大肠-枯草穿梭载体pBE-2中构建成Bt毒蛋白基因穿梭质粒pAMY,利用电穿孔法转人大肠杆菌DH5a,枯草芽孢杆菌B.subtilis BR151,IA511,野生型蜡状芽孢杆菌B.cereusa-47,短芽孢杆菌B.brevis A-5和枯草芽孢杆菌90-8,获得了具有较高杀虫活性的工程菌克隆。  相似文献

Fresh meat, poultry, and seafood purchased from Seattle area grocery stores were investigated for the presence of Shiga-like toxin-producing Escherichia coli by using DNA probes for Shiga-like toxin (SLT) genes I and II. High concentrations of boron (B) and selenium (Se) naturally found in the environment are detrimental to sustainable agriculture in the western USA. Greenhouse pot experiments were conducted to study B and Se uptake in three different plant species; Brassica juncea (L.) Czern (wild brown mustard), Festuca arundinacea Schreb. L. (tall fescue), and Brassica napus (canola) were grown in soil containing naturally occurring concentrations of 3.00 mg extractable B kg–1 and 1.17 mg total Se kg–1 soil. During the growing season, four intermediate harvests were performed on wild mustard and tall fescue. Final harvest I consisted of harvesting wild mustard, canola, and clipping tall fescue. Final harvest II consisted of harvesting wild mustard, which had been planted in soil in which wild mustard was previously grown, and harvesting previously clipped tall fescue. The greatest total amount of above ground biomass and below surface biomass was produced by tall fescue. Plants were separated into shoots and roots, weighted, and plant tissues were analyzed for total B and Se. The highest concentrations of tissue B were recovered in shoots of wild mustard and canola at final harvest I, while roots from tall fescue contained the highest concentrations of B irrespective of the harvest. Tissue Se concentrations were similar in all plants species. Soils were analyzed for residual B and Se. Extractable soil B concentrations at harvest times were lowered no less than 32% and total Se no less than 24% for all three species. The planting of wild mustard, canola, or tall fescue can reduce water-extractable B and total Se in the soil.  相似文献

In a world of accelerating changes in environmental conditions driving tree growth, tradeoffs between tree growth rate and longevity could curtail the abundance of large old trees (LOTs), with potentially dire consequences for biodiversity and carbon storage. However, the influence of tree-level tradeoffs on forest structure at landscape scales will also depend on disturbances, which shape tree size and age distribution, and on whether LOTs can benefit from improved growing conditions due to climate warming. We analyzed temporal and spatial variation in radial growth patterns from ~5000 Norway spruce (Picea abies [L.] H. Karst) live and dead trees from the Western Carpathian primary spruce forest stands. We applied mixed-linear modeling to quantify the importance of LOT growth histories and stand dynamics (i.e., competition and disturbance factors) on lifespan. Finally, we assessed regional synchronization in radial growth variability over the 20th century, and modeled the effects of stand dynamics and climate on LOTs recent growth trends. Tree age varied considerably among forest stands, implying an important role of disturbance as an age constraint. Slow juvenile growth and longer period of suppressed growth prolonged tree lifespan, while increasing disturbance severity and shorter time since last disturbance decreased it. The highest age was not achieved only by trees with continuous slow growth, but those with slow juvenile growth followed by subsequent growth releases. Growth trend analysis demonstrated an increase in absolute growth rates in response to climate warming, with late summer temperatures driving the recent growth trend. Contrary to our expectation that LOTs would eventually exhibit declining growth rates, the oldest LOTs (>400 years) continuously increase growth throughout their lives, indicating a high phenotypic plasticity of LOTs for increasing biomass, and a strong carbon sink role of primary spruce forests under rising temperatures, intensifying droughts, and increasing bark beetle outbreaks.  相似文献

This work firstly reported a new polycaprolactone based material functionalized with guanidinium ionic liquid (PCL-GIL) as the stationary phase with high resolution performance for capillary gas chromatography (GC). It is composed of polycaprolactone (PCL) and guanidinium ionic liquid (GIL) with amphiphilic conformation. The PCL-GIL capillary column coated by static method exhibited high column efficiency of 3942 plates/m and moderate polarity. As a result, the PCL-GIL column exhibited high-resolution capability. For a mixture of 27 analytes with a wide ranging polarity and outperformed the PCL-2OH and HP-35 columns, showing its advantageous separation capability for analytes of diverse types. Moreover, the PCL-GIL column showed high resolving capability for various positional isomers and cis-/trans-isomers, including alkylbenzenes, chlorobenzenes, naphthalenes, bromonitrobenzenes, chloronitrobenzenes, benzaldehydes, phenols, alcohols, respectively. In a word, PCL derivatized by GIL units as a new type of stationary phase has a promising future in GC separations.  相似文献

Obesity and low levels of physical and metabolic fitness are risk factors for cardiovascular disease and diabetes. The purpose of this investigation was to attenuate obesity and improve physical and metabolic fitness in elementary school children. Schools have the opportunity, mechanisms, and personnel in place to deliver nutrition education, fitness activities, and a school food service that is nutritious and healthy. Cohorts from grades 3 to 5 in two school districts in rural Nebraska (Intervention/Control) participated in a 2-year study of physical activity and modified school lunch program. Data collection for aerobic capacity, body composition, blood chemistry, nutrition knowledge, energy intake, and physical activity was at the beginning and end of each year. Int received enhanced physical activity, grade specific nutrition education, and a lower fat and sodium school lunch program. Con continued with a regular school lunch and team sports activity program. At year 2, Int lunches had significantly less energy (9%), fat (25%), sodium (21%), and more fiber (17%). However, measures of 24-hour energy intake for Int and Con showed significant differences for sodium only. Physical activity in the classroom was 6% greater for Int compared to Con (p < 0.05) but physical activity outside of school was ?16 % less for Int compared to Con (p < 0.05). Body weight and body fat were not different between schools for normal weight or obese children. No differences were found for cholesterol, insulin, and glucose; however, HDL cholesterol was significantly greater and cholesterol/HDL was significantly less for Int compared to Con (p < 0.05). It appears that compensation in both energy intake and physical activity outside of school may be responsible for the lack of differences between Int and Con.  相似文献
