模拟酸雨对主要酸性土壤中铝的溶出及形态的影响

本文研究了模拟酸雨对主要酸性土壤中铝的溶出及形态变化的影响。结果表明,模拟酸雨对土壤酸化的影响较小,但对土壤铝的溶出却影响明显,尤其在pH<4.0时;模拟酸雨对不同类型土壤的影响是不同的,其中以高度风化的酸性土壤较为敏感。模拟酸雨对土壤游离铝形态的影响是重要的,酸处理后,交换性铝略有增加,无定形活性铝增加较多,而有机络合态铝有减少的趋势。这表明在酸雨的长期作用下,铝终将转化为交换性铝和水溶性铝而进入环境并危害生态系统。     

The effect of the length of direct repeats and the presence of palindromes on deletion between directly repeated DNA sequences in bacteriophage T7.

The frequency of genetic deletion between directly repeated DNA sequences in bacteriophage T7 was measured as a function of the length of the direct repeat. The non-essential ligase gene (gene 1.3) of bacteriophage T7 was interrupted with pieces of synthetic DNA bracketed by direct repeats of various lengths. Deletion of these 76 bp long inserts was too low to be measured when the direct repeats were less than 6 bp long. However, the frequency of deletion of inserts with longer direct repeats increased exponentially as the length of the repeats increased from 8 to 20 bp. When inverted repeats (palindromes) were designed in the midst of the insert there was essentially no increase in deletion frequency between 10 bp direct repeats. But, the same palindromic sequences increased the deletion frequency between 5 bp direct repeats by at least two orders of magnitude. Thus, in this system homology at the endpoints is a more important determinant of deletion frequency than is the presence of palindromes between the direct repeats.     

Evidence for a new Escherichia coli protein resembling a lysyl-tRNA synthetase.

The amino acid sequence deduced from the nucleotide sequence of an open reading frame adjacent to the frdA gene of Escherichia coli shows 30.5% identity with the C terminus of Escherichia coli lysyl-tRNA synthetases. The three motifs characteristic of aminoacyl-tRNA synthetases of class 2 are recognizable within this sequence.     

The kinetics of the first wave of spermatogenesis in the newborn male mouse

A combination of autoradiography and air-dried techniques was used to calculate the duration of the major meiotic stages in the first wave of spermatogenesis in the newborn mouse. The data indicated that the entry into meiosis occurred asynchronously over 2 days, and the time required for each stage and the total cycle was constant. These time intervals were nearly identical with those estimated for adult animals in the present study and by other authors.     

Studies of myofibrillar ATPase in ragweed-sensitized canine pulmonary smooth muscle

The maximal shortening velocities of tracheal and pulmonary vascular smooth muscle from ragweed-sensitized dogs were significantly higher than those of muscles from their littermate controls. Myofibrils of tracheal and pulmonary vascular smooth muscle from ragweed-sensitized and control dogs were obtained with use of Triton X-100 homogenizing solution. The myofibrillar adenosinetriphosphatase (ATPase) activities of the sensitized tissues were significantly higher (P less than 0.05) than those of their respective controls.     

广西德峨苗族、彝族体质调查

对广西隆林县德峨乡男性22至60岁与女性20至60岁的576名苗族人(男395、女181)和178名彝族人(男88、女90)进行了活体观察与测量,计算出各项数据,总结德峨苗族、彝族的体质特征,并与国内一些民族相比较。     

Ti质粒T区tms基因的分离及其对高等植物分化的影响

根癌农杆菌Ti质粒的T区DNA带有致瘤基因,其基因1和基因2编码生长素吲哚乙酸生物合成途径中的两个酶。以pGV 354(pBR322质粒中插有Ti质粒C 58 T区DNA的HindⅢ15—HindⅢ22大片段)重组质粒出发,我们分离了基因1和基因2,并构建了带有卡那霉素抗性基因的重组质粒pBZ 692,通过基因载体pGV 3850,我们将基因1和基因2引入了高等植物。结果证明基因1和基因2能促使烟草、向日葵、土豆等转化组织分化长根,转化的根在MS_0培养基上能脱分化形成愈伤组织并自主生长,在转化的组织中有转化标记胭脂碱的存在。     

Resonance Raman spectra of the heme in leghemoglobin. Evidence for the absence of ruffling and the influence of the vinyl groups

Resonance Raman spectra of deoxy and carbonmonoxy leghemoglobin (Lb) are compared to the corresponding forms of human adult hemoglobin (HbA). It is found that the heme "core size" indicator line has nearly the same frequency for the two deoxyhemoglobins and the pi-electron density-sensitive line also falls at the same frequency. However, several other modes occur at very different frequencies in the spectra of the two proteins. From an examination of the spectrum of an HbA derivative in which the beta-carbon atoms of the heme vinyl groups were deuterated, it appears that the major differences between deoxy-HbA and -Lb may result from conformational changes in the vinyl groups. No evidence for the suggested ruffling (Irwin, M. J., Armstrong, R. S., and Wright, P. E. (1981) FEBS Lett. 133, 239-243) in deoxy-Lb was found. The spectra of carbonmonoxy-Lb and -HbA were also found to be very different. As in the deoxy case, some of these frequency differences could be attributed to vinyl group conformational differences. However, from the large difference in the pi-electron density-sensitive line, it appears that the vinyl pi-conjugation into the porphyrin in Lb(CO) may be different than it is in HbA(CO). The vinyl conformational differences may be a consequence of the looser heme pocket in Lb than in HbA. The difference in pi-conjugation could make a significant contribution to the difference in ligand binding affinity for these two globins.

Tyrosine-7 is an essential residue for the catalytic activity of human class PI glutathione S-transferase: chemical modification and site-directed mutagenesis studies.

The glutathione (GSH)-conjugating activity of human class Pi glutathione S-transferase (GST pi) toward 1-chloro-2,4-dinitrobenzene (CDNB) was significantly lowered by reaction with N-acetylimidazole, an O-acetylating reagent for tyrosine residues. Further, the replacement of Tyr7 in GST pi, which is conserved in all cytosolic GSTs, with phenylalanine by site-directed mutagenesis also lowered the activities toward CDNB and ethacrynic acid. The Km values of the mutant for both GSH and CDNB were almost equivalent to those of the wild type, while the Vmax of the former was about 55-fold smaller than that of the latter. Therefore, Tyr7 is considered to be an essential residue for the catalytic activity of GST pi.
