Amaranth is an ancient crop with a high content of good quality proteins. Globulins are some of the most abundant storage proteins of amaranth grain. They contain two fractions distinguishable according to their different solubility: the salt-soluble 7S and 11S-globulins and the globulin-p soluble in mild-alkaline, low-ionic-strength solutions. As part of the amaranth proteins characterization, in this work we investigated the structural characteristics responsible for the different physicochemical properties of these globulins. We studied certain conformational parameters of the purified aggregates (AMGp) and individual molecules (IMGp) of globulin-p and of the partially purified globulin (ppGb) and compared the AMGp polypeptide sequences with the sequence of the 11S-globulin propolypeptide from Amaranthus (gi|122726601). The results indicated that the AMGp aggregates are responsible for the different solubility of globulin-p. Subtle conformational differences as determined by fluorescence spectroscopy and urea sensitivity were found between the molecules studied: The AMGp showed some surface differences from the IMGp and the ppGb; the AMGp also had a lower affinity for the hydrophobic fluorescent probe 1,8-aniline-naphthalene-sulfonate and a higher ionic charge than the ppGb and the IMGp, characteristics that might cause their lower solubility. In addition, we have demonstrated differences between the AMGp polypeptide sequences and that reported for amaranth 11S-globulin. These differences suggest that the globulin-p and 11S-globulin are two 11S-globulin isoforms comprised of polypeptides coming from different legumin-gene subfamilies. 相似文献

A double-blind study was carried out in 60 women with climacteric symptoms: 30 women were given Org OD 14 (2.5 mg) and 30 were given a placebo to be taken daily for six weeks. The effects of the medication on the climacteric symptoms, the subjective sensations, the plasma FSH levels and endometrial histology were studied. In the treated group compared with the control group the relief or improvement of the following climacteric symptoms were recorded: perspiration, palpitations, irritability and backache. A favourable effect on the subjective sensations was noted in both groups, although no significant difference for the group which received Org OD 14 was found. At the end of the treatment with Org OD 14, the FSH levels were found to be greatly reduced in comparison with the basal values; this, however, was not the case with the placebo group. With regard to endometrial histology, no sign of hyperplasia was found in any of the patients. No relevant side effects or symptoms of estrogenic or androgenic stimulation were recorded. For the climacteric patient needing estrogen therapy, it can be concluded that Org OD 14 is an effective and innocuous medication in the doses used. 相似文献

Catalase electrophoretic patterns and enzymatic activities were measured in four organs of two anuran species, Rana ridibunda perezii and Discoglossus pictus. The D. pictus enzyme appeared as two distinguishable bands, whereas R. ridibunda catalase was monomorphic. Electrophoretic mobility of the major D. pictus catalase band was greater than that of R. ridibunda. Enzymes from both species showed slower mobility than that from bovine liver. Catalase activities did not show significant differences according to sex in any of the organs tested in R. ridibunda. Enzyme activities were similar in liver, kidney and brain when both species were compared. Only the heart showed much higher activity in D. pictus than in R. ridibunda. The catalase activity levels followed the order: liver greater than kidney greater than heart in both species. The heart showed higher activity than the brain in D. pictus but not in R. ridibunda. 相似文献

The five major antioxidants enzymes, cytochrome oxidase (COX), GSH, and GSSG, and endogenous and in vitro stimulated lipid peroxidation (TBA-RS) were assayed in the lung of old (28 months) and young (9 months) adult rats due to the almost total absence of data of this kind in this tissue, which is normally exposed to relatively high pO2 throughout life. Catalase, selenium (Se)-dependent GSH peroxidase (GPx), GSH reductase, GSH, GSSG, GSSG/GSH, and in vivo and in vitro TBA-RS showed similar values in old and young animals. The decrease observed for non Se-dependent GPx disappeared when the values were expressed in relation to COX activity. Only superoxide dismutase showed a clear decrease when referred both to protein and COX activity. These results suggest that lung aging is not accelerated in old age due to a decrease in the antioxidant capacity of the tissue. Nevertheless, they are compatible with a continuous damage of the lung tissue by free radicals throughout the life span. 相似文献

The effects of melatonin and noradrenaline (NA) on bi‐directional melanosome transport were analysed in primary cultures of melanophores from the Atlantic cod. Both agents mediated rapid melanosome aggregation, and by using receptor antagonists, melatonin was found to bind to a melatonin receptor whereas NA binds to an α2‐adrenoceptor. It has previously been stated that melatonin‐mediated melanosome aggregation in Xenopus is coupled with tyrosine phosphorylation of a so far unidentified high molecular weight protein and we show that although acting through different receptors and through somewhat different downstream signalling events, tyrosine phosphorylation is of the utmost importance for melanosome aggregation mediated by both NA and melatonin in cod melanophores. Together with cyclic adenosine 3‐phosphate‐fluctuations, tyrosine phosphorylation functions as a switch signal for melanosome aggregation and dispersion in these cells. 相似文献
