In recent years, polyphenols have attracted considerable attention as agents that protect cells or molecules from oxidative myocardial injury. The aim of the study was to prove the cardioprotective benefits of the flavonoid morin in isoproterenol-induced myocardial infarcted rats. Male Wistar rats are treated orally with morin (10 and 20 mg/kg) daily for a period of 21 days. After 21 days of pretreatment, isoproterenol (100 mg/kg) was injected subcutaneously to rats at an interval of 24 h for 2 days to induce myocardial infarction. Electrocardiographical abnormalities and biomarkers were measured in normal and experimental rats. Isoproterenol-induced myocardial infarcted rats showed significant (p<0.05) increase in the levels of cardiac markers. Pretreatment with morin regulated the abnormalities in electrocardiograph and biomarkers. The lipid peroxidation products were increased and indicated the increased lipid peroxidation in isoproterenol-induced myocardial infarcted rats. The rats pretreated with morin significantly reduced lipid peroxidation. The altered lipid metabolism was observed in isoproterenol-induced myocardial infarcted rats and in pretreatment with morin-regulated lipid metabolism. Histopathological study evidenced that the pretreatment with morin inhibited myocardial damage. The results of this study proved the protective effect of morin as pretreatment and are rational to understand the beneficial effects of morin on cardioprotection against myocardial injury. Based on the results, the cardioprotective ability of morin on human beings can be studied in the future. 相似文献

The Drosophila melanogaster Gld gene has multiple and diverse developmental and physiological functions. We report herein that interactions among proximal promoter elements and a cluster of intronically located enhancers and silencers specify the complex regulation of Gld that underlies its diverse functions. Gld expression in nonreproductive tissues is largely determined by proximal promoter elements with the exception of the embryonic labium where Gld is activated by an enhancer within the first intron. A nuclear protein, GPAL, has been identified that binds the Gpal elements in the proximal promoter region. Regulation of Gld in the reproductive organs is particularly complex, involving interactions among the Gpal proximal promoter elements, a unique TATA box, three distinct enhancer types, and one or more silencer elements. The three somatic reproductive organ enhancers each activate expression in male and female pairs of reproductive organs. One of these pairs, the male ejaculatory duct and female oviduct, are known to be developmentally homologous. We report evidence that the other two pairs of organs are developmentally homologous as well. A comprehensive model to explain the full developmental regulation of Gld and its evolution is presented. 相似文献

In this study, S-allyl cysteine sulfoxide (SACS) was used to evaluate its preventive effect in isoproterenol (ISO)-induced myocardial ischemia in male Wistar rats. Rats were pretreated with SACS (40 and 80 mg kg(-1)) orally for 5 weeks. After the treatment period, ISO (150 mg kg(-1)) was administered subcutaneously to rats at an interval of 24 h for 2 days. The activities of beta-D-N-acetyl-glucosaminidase, beta-galactosidase, beta-glucosidase, and acid phosphatase increased in serum and heart in ISO-induced rats. In addition, these rats showed a significant (p < 0.05) increase in the activities of beta-glucuronidase and cathepsin-D in serum and heart and a significant (p < 0.05) decrease in their activities in lysosomal fraction of the heart. The activity of Na(+)K(+)-ATPase declined, while those of Ca(2+)- and Mg(2+)-ATPases significantly (p < 0.05) elevated in the heart of ISO-induced rats. Pretreatment with SACS (40 and 80 mg kg(-1)) showed a significant (p < 0.05) effect in all the biochemical parameters studied. The effect at a dose of 80 mg kg(-1) body weight was more effective than that at 40 mg kg(-1) body weight and brought back all the biochemical parameters to near normal levels. Hereby, our study shows the membrane-stabilizing as well as antioxidant effects of SACS in ISO-induced rats. 相似文献

Pitfall traps were used to monitor predatory arthropod numbers along two types of field boundary, a post and wire fence line and a Cupressus macrocarpa hedge, along the same paddock margin in Canterbury, New Zealand, over 24 months. The seven most abundant predator groups recorded were: Araneae > Phalangiidae > Staphylinidae > Coccinellidae > Chilopoda > Hemerobiidae > Carabidae. Araneae, Phalangiidae, Staphylinidae, Chilopoda and Hemerobiidae were found in larger numbers at the wire fence than at the hedge site, whereas the numbers of Carabidae and Coccinellidae adults exhibited no field margin preference. However, more species of Araneae and Staphylinidae were caught at the hedge site, whereas species richness of carabid beetles was greatest at the wire fence. Principal component analysis clearly separated the samples collected from the two habitats based on the assemblages of Araneae, Staphylinidae and Carabidae, and certain species in each of these taxonomic groups appeared to be particularly associated with one boundary type or the other. All the main taxonomic groups exhibited clear seasonal patterns, with distinct peaks in abundance occurring at certain times of the year. The results of the study reinforce the idea that management of field boundaries can be used to manipulate the type and abundance of particular groups of predatory arthropods, and that seasonal patterns should be taken into account in schemes of integrated pest management so that any adverse effects of biocide application on these beneficial species may be minimised. 相似文献

Prior research has indicated that the two sperm cells of Nicotiana tabacum are dimorphic, suggesting that they may participate in preferential fertilization during in vivo fusion with the egg and
central cells. To probe the mechanism of potential preferential fertilization in this plant, it will be necessary to use modern
sensitive molecular techniques. For this purpose, two individual populations of two sperm cells, constituting the Svn (associated
with the vegetative nucleus) and Sua (unassociated with the vegetative nucleus), were isolated in the thousands from tobacco
pollen tubes with a micromanipulator as a preliminary step toward research on gametic recognition using molecular techniques.
Microelectrophoresis of paired sperm cells from a single pollen tube was conducted at different developmental stages. Sperm
cells isolated from 1-, 2-, 3- and 4-cm stylar lengths migrated to the negative pole, with the Sua displaying significantly
greater electrophoretic mobility than the Svn, reflecting a more positively charged cell surface on the Sua. The sperm cells
isolated from 1-cm style are very sensitive to electron potential in an electrophoretic field, presumably reflecting that
they are still in a young state. Differences in cell surface charge between the Sua and Svn may be related with cell fate
during fertilization.

The stereoselective affinity of small-molecule binding to proteins is typically broadly explained in terms of the thermodynamics of the final bound complex. Using Brownian dynamics simulations, we show that the preferential binding of the MDM2 protein to the geometrical isomers of Nutlin-3, an effective anticancer lead that works by inhibiting the interaction between the proteins p53 and MDM2, can be explained by kinetic arguments related to the formation of the MDM2:Nutlin-3 encounter complex. This is a diffusively bound state that forms prior to the final bound complex. We find that the MDM2 protein stereoselectivity for the Nutlin-3a enantiomer stems largely from the destabilization of the encounter complex of its mirror image enantiomer Nutlin-3b, by the K70 residue that is located away from the binding site. On the other hand, the trans-Nutlin-3a diastereoisomer exhibits a shorter residence time in the vicinity of MDM2 compared with Nutlin-3a due to destabilization of its encounter complex by the collective interaction of pairs of charged residues on either side of the binding site: Glu25 and Lys51 on one side, and Lys94 and Arg97 on the other side. This destabilization is largely due to the electrostatic potential of the trans-Nutlin-3a isomer being largely positive over extended continuous regions around its structure, which are otherwise well-identified into positive and negative regions in the case of the Nutlin-3a isomer. Such rich insight into the binding processes underlying biological selectivity complements the static view derived from the traditional thermodynamic analysis of the final bound complex. This approach, based on an explicit consideration of the dynamics of molecular association, suggests new avenues for kinetics-based anticancer drug development and discovery. 相似文献

Wildlife management remains a matter of considerable controversy amongst many stakeholders, particularly where lethal control (culling) is used. Wild deer provide perhaps one of the best examples of such a management ‘problem’, especially where they are encountered in peri-urban environments. Faced with potential controversy, decision-makers in public land management organisations need information which clearly differentiates between generally acceptable management objectives and methods, and less mainstream preferences. We conducted a questionnaire survey aimed at assessing the preferences of community members in peri-urban Scotland for management methods in response to specific problems and analysed the results in relation to three social categories—age, gender and familiarity with wildlife. Active management was broadly accepted. Fencing dominated as the preferred first management response with little variation across social category, or between management problems. Changing human behaviour (e.g. introducing speed limits) was also a highly preferred response by the respondents in this study. In general, the order of preferred management responses stayed the same in relation to different management impacts. Culling (lethal control) had almost no support as a first management response to any problem, although considerably more respondents support it as a third preference. Our results suggest that culling is acceptable as a subsidiary management response in peri-urban areas, but only where other preferred management methods have been tried and failed. 相似文献

Losses of natural and semi-natural forests, mostly to agriculture, are a significant concern for biodiversity. Against this
trend, the area of intensively managed plantation forests increases, and there is much debate about the implications for biodiversity.
We provide a comprehensive review of the function of plantation forests as habitat compared with other land cover, examine
the effects on biodiversity at the landscape scale, and synthesise context-specific effects of plantation forestry on biodiversity.
Natural forests are usually more suitable as habitat for a wider range of native forest species than plantation forests but
there is abundant evidence that plantation forests can provide valuable habitat, even for some threatened and endangered species,
and may contribute to the conservation of biodiversity by various mechanisms. In landscapes where forest is the natural land
cover, plantation forests may represent a low-contrast matrix, and afforestation of agricultural land can assist conservation
by providing complementary forest habitat, buffering edge effects, and increasing connectivity. In contrast, conversion of
natural forests and afforestation of natural non-forest land is detrimental. However, regional deforestation pressure for
agricultural development may render plantation forestry a ‘lesser evil’ if forest managers protect indigenous vegetation remnants.
We provide numerous context-specific examples and case studies to assist impact assessments of plantation forestry, and we
offer a range of management recommendations. This paper also serves as an introduction and background paper to this special
issue on the effects of plantation forests on biodiversity.

Studies in other countries have identified social class as a risk factor for infant mortality. In Australia there is no systematic collection of population data by social class, partly due to the absence of a recognized measure. The use of occupational prestige as an indicator of social class is discussed and Australian prestige scales reviewed. In a population based study, logistic regression analysis of infant mortality in an Australian (NSW) population shows the effects of social class on infant mortality which remain when maternal age, marital status and parity are controlled. 相似文献
