Interactions with nucleoporins containing FxFG-repeat cores are crucial for the nuclear import of RanGDP mediated by nuclear transport factor 2 (NTF2). We describe here the 1.9 A resolution crystal structure of yeast NTF2-N77Y bound to a FxFG-nucleoporin core, which provides a basis for understanding this interaction and its role in nuclear trafficking. The two identical FxFG binding sites on the dimeric molecule are formed by residues from each chain of NTF2. Engineered mutants at the interaction interface reduce the binding of NTF2 to nuclear pores and cause reduced growth rates and Ran mislocalization when substituted for the wild-type protein in yeast. Comparison with the crystal structure of FG-nucleoporin cores bound to importin-beta and TAP/p15 identified a number of common features of their binding sites. The structure of the binding interfaces on these transport factors provides a rationale for the specificity of their interactions with nucleoporins that, combined with their weak binding constants, facilitates rapid translocation through NPCs during nuclear trafficking.  相似文献

Sense organs in the legs that detect body weight are an important component in the regulation of posture and locomotion. We tested the abilities of tibial campaniform sensilla, receptors that can monitor forces in the cockroach leg, to encode variations in body load in freely standing animals. Small magnets were attached to the thorax and currents were applied to a coil below the substrate. Sensory and motor activities were monitored neurographically. The tibial sensilla could show vigorous discharges to changing forces when animals stood upon their legs and actively supported the body weight. Firing of individual afferents depended upon the orientation of the receptors cuticular cap: proximal sensilla (oriented perpendicular to the leg axis) discharged to force increases while distal receptors (parallel to the leg) fired to decreasing forces. Proximal sensillum discharges were prolonged and could encode the level of load when increases were sustained. Firing of the trochanteral extensor motoneuron was also strongly modulated by changing load. In some postures, sensillum discharges paralleled changes in motor frequency consistent with a known interjoint reflex. These findings demonstrate that tibial campaniform sensilla can monitor the effects of body weight upon the legs and may aid in generating support of body load.  相似文献

Nuclear transport factor 2 (NTF2) is a small homodimeric protein that interacts simultaneously with both RanGDP and FxFG nucleoporins. The interaction between NTF2 and Ran is essential for the import of Ran into the nucleus. Here we use mutational analysis to dissect the in vivo role of the interaction between NTF2 and nucleoporins. We identify a series of surface residues that form a hydrophobic patch on NTF2, which when mutated disrupt the NTF2-nucleoporin interaction. Analysis of these mutants in vivo demonstrates that the strength of this interaction can be significantly reduced without affecting cell viability. However, cells cease to be viable if the interaction between NTF2 and nucleoporins is abolished completely, indicating that this interaction is essential for the function of NTF2 in vivo. In addition, we have isolated a dominant negative mutant of NTF2, N77Y, which has increased affinity for nucleoporins. Overexpression of the N77Y protein blocks nuclear protein import and concentrates Ran at the nuclear rim. These data support a mechanism in which NTF2 interacts transiently with FxFG nucleoporins to translocate through the pore and import RanGDP into the nucleus.  相似文献

Thamnurgus euphorbiae overwinters as adults in dry stems of Euphorbia characias until the end of March before exiting through circular holes cut with their mandibles. Females and males appeared in the field at the same time, and the first pairs were observed on 28 March on new stems of E. characias having the open‐flower at the beginning of its development. Throughout the rest of April it was possible to find and collect the fairly abundant adults. Mating occurs on new stems of the host plant and females walk up towards the top of the stem and start to mine the centre of it in order to penetrate. Oviposition occurs in new stems and eggs are laid singly along the interior of the stem; females laid 35–85 eggs. Upon hatching, larvae started feeding on vascular bundles and the inner cortex. There are three larval instars, and pupation occurred in the stem of the host plant; this insect is univoltine. Thamnurgus euphorbiae has been accepted by the Technical Advisory Group for Biological Control Agents of Weeds for release as a biological control agent of leafy spurge in the US. Prior to this study the biology of T. euphorbiae was unknown.  相似文献

A new agent, provisionally designated Herpesvirus salmonis, was isolated from post-spawning rainbow trout (Salmo gairdneri) and studied primarily in the RTG-2 rainbow trout cell line. Infection of RTG-2 cells resulted in the formation of syncytia and Cowdry type A intranuclear inclusions. Replication occurred regularly at 5 and 10°C, but was inconsistent at 15°C, largely inhibited at 0°C, and completely inhibited at 20°C or higher. The virus was acid, heat, ether, and chloroform labile, but stable to freezing and thawing. It did not hemagglutinate. Viral DNA had a buoyant density of 1.709 g/cm3 and a guanine-cytosine value of 50%. Hexagonal nucleocapsids had a diameter of 90 nm and were first seen in nuclei at 36 h. Enveloped forms measured about 150 nm and occurred both cytoplasmically and extracellularly. At 10°C, a one-step growth culture required about 96 h; cell-associated virus peaked at about 105 PFU/ml and exceeded released virus by a factor of about 10.  相似文献

Acepromazine-ketamine anesthesia in the rhesus monkey (Macaca mulatta).   总被引:2,自引:0,他引:2  
Six adult male rhesus monkeys (Macaca mulatta) were anesthetized using a combination of acepromazine maleate and ketamine hydrochloride administered by intramuscular injection. This combination produced a smooth induction to anesthesia requiring less than 5 minutes. The average duration of anesthesia was slightly less than 1 hour and was safely prolonged with additional doses of ketamine. The depth of anesthesia was sufficient for minor surgical procedures and for precise radiological studies. No deleterious side effects were noted, and animals recovered completely in a short time.  相似文献

Cotton (Gossypium hirsutum L. var. `Stoneville 213'), velvetleaf (Abutilon theophrasti Medic.), redroot pigweed (Amaranthus retroflexus L.), and hemp sesbania (Sesbania exaltata [Raf.] Cory) were grown in a controlled environment room at 31/25 C day/night temperature and three irradiances: 90, 320, and 750 μeinsteins meter−2 second−1. From total dry weights and leaf areas determined at intervals during the first exponential phase of growth, we used mathematical growth analysis techniques to calculate net assimilation rates (NAR), relative growth rates (Rw), relative leaf area expansion rates (Ra), leaf area partition coefficients (LAP), and leaf area ratios (LAR). In all four species, Rw, Ra, and NAR decreased with decreasing growth irradiance, while LAP and LAR increased. Within each species, Rw was positively correlated with NAR but negatively correlated with LAP and LAR. In comparisons among the four species within each growth irradiance, Rw was positively correlated with LAP. We discuss the relationship between LAP and LAR and show that LAP = (Ra/Rw) (LAR).  相似文献

The medical literature contains many examples of cases in which serendipitous observations have led to important findings. In the example described in this article, laboratory and field observations conducted at the Mohawk Nation Community of Akwesasne led to the important and unexpected finding that frogs once plentiful in the area were no longer observed. Laboratory tests comparing river sediments from Akwesasne to pristine sediment from Ithaca, NewYork, indicated multiple adverse health effects on developing frogs. Some of the behavioral changes observed in the laboratory were similar to those described by residents of Akwesasne before the onset of amphibian decline. The magnitude of changes paralleled frog body burden of polychlorinated biphenyls acquired from the Akwesasne sediment. The impact of these findings on the identification of a hazardous waste site and global amphibian decline are discussed.  相似文献
