A computational intelligent system that models the human cognitive abilities may promise significant performance in problem learning because human is effective in learning and problem solving. Functionally modelling the human cognitive abilities not only avoids the details of the underlying neural mechanisms performing the tasks, but also reduces the complexity of the system. The complementary learning mechanism is responsible for human pattern recognition, i.e. human attends to positive and negative samples when making decision. Furthermore, human concept learning is organized in a hierarchical fashion. Such hierarchical organization allows the divide-and-conquer approach to the problem. Thus, integrating the functional models of hierarchical organization and complementary learning can potentially improve the performance in pattern recognition. Hierarchical complementary learning (HCL) exhibits many of the desirable features of pattern recognition. It is further supported by the experimental results that verify the rationale of the integration and that the HCL system is a promising pattern recognition tool.  相似文献

Quaternary aeolian deposits of the Canary Islands contain well‐preserved terrestrial gastropods, providing a suitable setting for assessing the taphonomy and compositional fidelity of their fossil record over ~13 kyr. Nine beds (12, 513 shells) have been analysed in terms of multivariate taphonomic and palaeoecological variables, taxonomic composition, and the stratigraphic and palaeontological context. Shells are affected by carbonate coatings, colour loss and fragmentation. Shell preservation is size‐specific: juveniles are less fragmented and show colour preservation more commonly than adults. In palaeosols, the adult shell density correlates negatively with the proportion of fragmented adults, negatively with the proportion of juveniles, and positively with the proportion of adults with coatings. High bioturbation intensity in palaeosols is associated with low shell fragmentation and high proportion of shells with coatings. These relationships imply that high adult density in palaeosols was driven by an increase in shell production rate (related to a decrease in predation rates on adults and a decrease in juvenile mortality) and a decrease in shell destruction rate (related to an increase in durability enhanced by carbonate precipitation). In dunes, the relationships between taphonomic alteration, shell density and bioturbation are insignificant. However, dune assemblages are characterized by a lower frequency of shells with coatings and higher rates of colour loss, indicating lower shell durability in dunes than in palaeosols. Additionally, non‐random differences in the coating proportion among palaeosols imply substantial temporal variation in the rate of carbonate crust formation, reflecting long‐term changes in bioturbation intensity that covaries positively with shell preservation. Dunes and palaeosols do not differ in species abundances despite differences in the degree of shell alteration, suggesting that both weakly and strongly altered assemblages offer data with a high compositional fidelity. Carbonate‐rich terrestrial deposits originating in arid conditions can enhance the preservation of gastropods and result in fossil assemblages that are suitable for palaeoecological and palaeoenvironmental studies of terrestrial ecosystems.  相似文献

Philippine vegetable farmers commonly use synthetic insecticides to control insect pests on yardlong beans ( Vigna unguiculata ssp. sesquipedalis ). An important pest on yardlong beans is the cowpea aphid, Aphis craccivora Koch. Overuse of chemical insecticides and the adverse consequences for farmer health and for the environment have been reported. The natural enemies of A. craccivora , such as the coccinelidae beetle, Menochilus sexmaculatus (Fabr.), do not provide economic control on their own. In the present study the efficacy of the biological insecticide neem (both commercial and homemade) alone, and in combination with diatomaceous earth against A. craccivora was evaluated. The same insecticides were also examined to investigate their deleterious effect on M. sexmaculatus . The efficacies of different treatments with biological insecticides were compared with the use of the synthetic insecticide Hostathion (triazophos). Experiments were conducted under Philippine lowland conditions during the dry season when the occurrence of pest problems in yardlong beans is very great. Commercial neem, NeemAzal-T/S (Trifolio-M GmbH, Lahnau, Germany), significantly reduced the number of A. craccivora . NeemAzal-T/S and diatomaceous earth in combination produced the best control of A. craccivora and were less toxic to M. sexmaculatus than treatment with Hostathion (triazophos). Aqueous homemade neem solution did not control the A. craccivora populations.  相似文献

The Src homology (SH)2 domain-containing protein-tyrosine phosphatase SHP-1 is tyrosine phosphorylated in platelets in response to the glycoprotein VI (GPVI)-selective agonist collagen-related peptide (CRP), collagen, and thrombin. Two major unidentified tyrosine-phosphorylated bands of 28 and 32 kDa and a minor band of 130 kDa coprecipitate with SHP-1 in response to all three agonists. Additionally, tyrosine-phosphorylated proteins of 50-55 and 70 kDa specifically associate with SHP-1 following stimulation by CRP and collagen. The tyrosine kinases Lyn, which exists as a 53 and 56-kDa doublet, and Syk were identified as major components of these bands, respectively. Kinase assays on SHP-1 immunoprecipitates performed in the presence of the Src family kinase inhibitor PP1 confirmed the presence of a Src kinase in CRP- but not thrombin-stimulated cells. Lyn, Syk, and SLP-76, along with tyrosine-phosphorylated 28-, 32-, and 130-kDa proteins, bound selectively to a glutathione S-transferase protein encoding the SH2 domains of SHP-1, suggesting that this is the major site of interaction. Platelets isolated from motheaten viable mice (mev/mev) revealed the presence of a heavily tyrosine-phosphorylated 26-kDa protein that was not found in wild-type platelets. CRP-stimulated mev/mev platelets manifested hypophosphorylation of Syk and Lyn and reduced P-selectin expression relative to controls. These observations provide evidence of a functional role for SHP-1 in platelet activation by GPVI.  相似文献

以黄曲霉(Aspergillus flavus.)为出发菌株,经3次紫外线、1次^60Co、3次亚硝基胍多重复合诱变处理,选育获得曲酸生产菌UCN7—17,配以最佳培养条件,发酵7d,曲酸产量由原来的0.926%,提高到6.3%。实验证明采用多因子复合诱变,能有效改变菌株对诱变因素敏感性,提高变异率,逐步提高突变株的产酸水平。  相似文献

Rubella, a viral disease characterized by a red skin rash, is well controlled because of an effective vaccine, but outbreaks are still occurring in the absence of available antiviral treatments. The Rubella virus (RUBV) papain-like protease (RubPro) is crucial for RUBV replication, cleaving the nonstructural polyprotein p200 into two multifunctional proteins, p150 and p90. This protease could represent a potential drug target, but structural and mechanistic details important for the inhibition of this enzyme are unclear. Here, we report a novel crystal structure of RubPro at a resolution of 1.64 Å. The RubPro adopts a unique papain-like protease fold, with a similar catalytic core to that of proteases from Severe acute respiratory syndrome coronavirus 2 and foot-and-mouth disease virus while having a distinctive N-terminal fingers domain. RubPro has well-conserved sequence motifs that are also found in its newly discovered Rubivirus relatives. In addition, we show that the RubPro construct has protease activity in trans against a construct of RUBV protease–helicase and fluorogenic peptides. A protease–helicase construct, exogenously expressed in Escherichia coli, was also cleaved at the p150–p90 cleavage junction, demonstrating protease activity of the protease–helicase protein. We also demonstrate that RubPro possesses deubiquitylation activity, suggesting a potential role of RubPro in modulating the host''s innate immune responses. We anticipate that these structural and functional insights of RubPro will advance our current understanding of its function and help facilitate more structure-based research into the RUBV replication machinery, in hopes of developing antiviral therapeutics against RUBV.  相似文献

A new series of coumarin-yl-chalcone derivatives (3a-m) had been designed and synthesized through different reactions such as aromatic addition, cyclization and Claisen-Schmidt reactions in good yields (54–78%). 5-acetyl-4-(2-hydroxyphenyl) -6-methyl-3, 4-dihydropyrimidin-2(1H) -one (1) has been synthesized by multi-component one pot reaction of salicylaldehyde, methyl acetoacetate and urea, which was further reacted with malonic acid employing ZnCl2 catalyst to yield 5-acetyl-4-(4-hydroxy-2-oxo-2H-chromen-8-yl) -6-methyl-3, 4-dihydropyrimidin-2(1H) -one (2). The title compounds (3a-m) were synthesised by reacting 5-acetyl-4-(4-hydroxy-2-oxo-2H-chromen-8-yl) -6-methyl-3, 4-dihydropyrimidin-2(1H)-one (2) with different aromatic aldehydes in the presence of potassium hydroxide. In silico studies, a preliminary screening method for predicting the anti-cancer activity was performed for the synthesized compounds (3a-m) against Src, Alb tyrosine kinase and homology model protein (PDB ID: 4csv). The derivatives 3h and 3m showed moderate binding energies. The in vitro cytotoxic activity was evaluated for the compounds 3h and 3m by using human cancer cell-line morphology and MTT assay against three human cell-lines A549 (Lung), Jurkat (Leukemia) and MCF-7 (Breast). The results indicate that the derivatives 3h and 3m display significant anti-cancer activity, however it was found to be less cytotoxic when compared to the standard used i.e. Imatinib.  相似文献

With almost 900 described species, Hydraena Kugelann (Hydraenidae) is one of the largest genera among Coleoptera. The subgeneric classification of Hydraena has been controversial, with 11 subgeneric names having so far been attributed to it. Some of these, Haenydra Rey and Spanglerina Perkins, have been treated as valid genera, as subgenera or as species groups. The most recent complete treatment of the genus, based on a cladistic analysis of morphological characters, recognized two major lineages, and only these were classified as subgenera: Hydraenopsis (mainly Gondwanan distribution), and Hydraena s.str. (mainly Laurasian). Here, we reconstruct the phylogeny of Hydraena using 212 species plus several outgroups and approximately 4 kb of sequence data from two nuclear (SSU and LSU) and four mitochondrial genes (cox1, rrnL, trnL and nad1). Data were aligned with two different strategies of multiple alignment (implemented in mafft and prank ), and the phylogenies reconstructed using maximum likelihood and Bayesian methods. We estimated approximate ages of the main nodes using a relaxed molecular clock with Bayesian methods, and an a priori evolutionary rate of 0.01 substitutions/site/million years (Ma) plus a calibration point based on a biogeographical split. We found strong support for the monophyly of Hydraena and many of the clades recognized with morphological data. The following clades are considered as subgenera: Phothydraena Kuwert, Spanglerina Perkins, Holcohydraena Kuwert, Hydraenopsis Janssens and Hydraena s.str. The placement of three species groups, two Neotropical (H. multispina group, H. paeminosa group) and one South African/Madagascan (H. monikae group), is uncertain, and they are considered incertae sedis within Hydraena. The origin of the genus was estimated to be in the Lower Eocene, with many species complexes diversifying in the Pleistocene. Dispersal events seem to have played a key role in order to determine the current distribution of the species groups in the southern hemisphere (mainly in Hydraenopsis).  相似文献

Genome-scale models are used for an ever-widening range of applications. Although there has been much focus on specifying the stoichiometric matrix, the predictive power of genome-scale models equally depends on reaction directions. Two-thirds of reactions in the two eukaryotic reconstructions Homo sapiens Recon 1 and Yeast 5 are specified as irreversible. However, these specifications are mainly based on biochemical textbooks or on their similarity to other organisms and are rarely underpinned by detailed thermodynamic analysis. In this study, a to our knowledge new workflow combining network-embedded thermodynamic and flux variability analysis was used to evaluate existing irreversibility constraints in Recon 1 and Yeast 5 and to identify new ones. A total of 27 and 16 new irreversible reactions were identified in Recon 1 and Yeast 5, respectively, whereas only four reactions were found with directions incorrectly specified against thermodynamics (three in Yeast 5 and one in Recon 1). The workflow further identified for both models several isolated internal loops that require further curation. The framework also highlighted the need for substrate channeling (in human) and ATP hydrolysis (in yeast) for the essential reaction catalyzed by phosphoribosylaminoimidazole carboxylase in purine metabolism. Finally, the framework highlighted differences in proline metabolism between yeast (cytosolic anabolism and mitochondrial catabolism) and humans (exclusively mitochondrial metabolism). We conclude that network-embedded thermodynamics facilitates the specification and validation of irreversibility constraints in compartmentalized metabolic models, at the same time providing further insight into network properties.  相似文献
