In this study, 20 laboratory reared Onychomys leucogaster from a parental population that is naturally exposed to plague were each fed a white mouse that had been inoculated with Yersinia pestis. Three of the 20 O. leucogaster died, four survived with antibody titers against Y. pestis and 13 survived with no titer against Y. pestis. In contrast, when 20 O. leucogaster from a plague naive parental population were fed infected prey, seven died and 13 survived with no antibody titer against Y. pestis. Our results suggest another means by which O. leucogaster from populations that are naturally exposed to plague may acquire the disease.  相似文献

The larch bark beetle (Ips subelongatus), which occurs in larch plantations over a vast area of eastern Asia, infects both dying and fallen trees. When its population reaches a high density, the beetle may also infect healthy trees, resulting in tree decline and, eventually, death. Leptographium spp., in both their sexual and asexual states, are mainly associated with conifer-infesting bark beetles; some species are important tree pathogens. The aims of this study were to identify the Leptographium spp. associated with I. subelongatus infestations of Larix spp. in northern China and to examine their pathogenicity towards the tree. Morphological studies and phylogenetic approaches based on multilocus DNA sequence data (ITS2- partial r28S, partial β-tubulin, and EF-1α gene regions) showed that three Leptographium species occur in association with I. subelongatus in the areas investigated: Leptographium taigense, which is recorded in China for the first time, and two new species, namely L. innermongolicum sp. nov. and L. zhangii sp. nov. Leptographium innermongolicum is closely related to L. taigense, whereas L. zhangii belongs to the Grosmannia piceaperda species complex. The pathogenicity of these Leptographium species towards mature Larix spp. was tested by stem inoculation in forests. All inoculations only resulted in small lesions on the inner bark; therefore, the three Leptographium species were not considered to be pathogenic.  相似文献

The protein elicitor from Alternaria tenuissima (PeaT1) presented excellent thermotolerance and potential application in agriculture as a pesticide. Previous synchrotron radiation circular dichroism study demonstrated that the secondary structures in PeaT1 protein are reversible with temperature change. To further clarify the mechanism of its thermotolerance, synchrotron radiation small angle x‐ray scattering (SAXS) technique was used to study the shape change of PeaT1 protein with temperature in this article. Ab initio structure restorations based on the SAXS data revealed that PeaT1 protein has a prolate shape with a P2 symmetry axis along the prolate anisometric direction. With temperature increase, a gooseneck vase‐like (25°C), to jug‐like (55°C), then to oval (85°C) shape change can be found, and these shape changes are also approximately reversible with temperature decrease. PeaT1 protein contains two homogenous molecules, and each of them consists of F, NAC, T, and UBA domains. The structures of the four domains were predicted. Simulated annealing algorithm was used to superimpose the domain structures onto the SAXS shapes. It was found that all the structural domains have position rotation and translation with temperature change, but the NAC domains are relatively stable, playing a role of frame. It has been proposed that bile acid suppression of CYP7A1 gene expression is mediated through a gut-liver signaling pathway fibroblast growth factor (FGF)15/19-fibroblast growth factor receptor 4 which is initiated by activation of farnesoid X receptor in the ileum but not in the liver. This study evaluated whether FGF15/19 protein levels in the portal blood reflected changes in FGF15/19 mRNA in the ileum. Studies were conducted in Sprague Dawley rats and New Zealand white rabbits fed regular chow (controls), supplemented with cholesterol (Ch) or cholic acid (CA). After feeding CA, ileal FGF15 mRNA increased 8.5-fold in rats and FGF19 rose 16-fold in rabbits associated with 62 and 75% reduction of CYP7A1 mRNA, respectively. Neither FGF15 nor FGF19 protein levels changed in the portal blood to correspond with the marked increase of FGF15/19 mRNA levels in the ileum or inhibited CYP7A1 expression in the liver. Further, in Ch-fed rats, CYP7A1 mRNA increased 1.9-fold (P < 0.001) although FGF15 mRNA levels in the ileum and portal blood FGF15 protein levels were not decreased. In Ch-fed rabbits, although FGF19 mRNA levels in the ileum and liver did not increase significantly, CYP7A1 mRNA declined 49% (P < 0.05). We were unable to find corresponding changes of FGF15/19 protein levels in the portal blood in rats and rabbits where the mRNA levels of FGF15/19 in the ileum and CYP7A1 in the liver change significantly.  相似文献

The effects of nitrogen (N) deposition and management practices on leaf litterfall and N and P return in Moso bamboo forest are not yet known. In this study, we investigated the effects of four levels of simulated N deposition, including low-N (N30, 30 kg ha?1 year?1), medium-N (N60, 60 kg ha?1 year?1), and high-N (N90, 90 kg ha?1 year?1), and a control with no N (N-free addition). The experiment was performed in a Moso bamboo forest under conventional management (CM) and intensive management (IM). The results showed that leaf litterfall and N and P return occurred mainly from March to June and accounted for 78.2–82.2, 78.5–82.1, and 85.6–94.6% of annual leaf litterfall, N return, and P return, respectively. Unlike CM, IM significantly increased leaf litterfall and N and P return. The positive effects were further amplified by low- and medium-N deposition, but not high-N deposition. The combination of low- and medium-N deposition and IM significantly increased N and P return, but not litterfall. Our results indicated that the interaction of anthropogenic management practices and N deposition need to be considered when estimating the effects of N deposition on the biogeochemical cycle of a forest ecosystem.  相似文献

High energy density Li‐ion hybrid flow capacitors are demonstrated by employing LiMn2O4 and activated carbon slurry electrodes. Compared to the existing aqueous flow electrochemical capacitors, the hybrid one exhibits much higher energy densities due to the introduction of high capacity Li‐insertion materials (e.g., LiMn2O4 in the present work) as the flowable electrode with asymmetrical cell configuration. A record energy density, i.e., 23.4 W h kg?1 at a power of 50.0 W kg?1 has been achieved for aqueous flow capacitors tested at static condition reported to date. A full operational Li‐ion flow capacitor tested in an intermittent‐flow mode has also been demonstrated. The Li‐ion hybrid flow capacitor shows great promise for high‐rate grid applications.  相似文献

A comprehensive approach is reported to construct stable and high volumetric energy density lithium–sulfur batteries, by coupling a multifunctional and hierarchically structured sulfur composite with an in‐situ cross‐linked binder. Through a combination of first‐principles calculations and experimental studies, it is demonstrated that a hybrid sulfur host composed by alternately stacking graphene and layered graphitic carbon nitride embraces high electronic conductivity as well as high polysulfide adsorptivity. It is further shown that the cross‐linked elastomeric binder empowers the hierarchical sulfur composites—multi‐microns in size—with the ability to form crack‐free and compact high‐loading electrodes using traditional slurry processing. Using this approach, electrodes with up to 14.9 mg cm?2 sulfur loading and an extremely low electrolyte/sulfur ratio as low as 3.5: 1 µL mg?1 are obtained. This study sheds light on the essential role of multifaceted cathode design and further on the challenges facing lithium metal anodes in building high volumetric energy density lithium–sulfur batteries.  相似文献

Cryopreservation of domestic animal sperm has been widely used for artificial insemination (AI), and egg yolk is one of the most commonly used cryoprotectants during the freezing-thawing process. The objectives of this study were to compare the effectiveness of egg yolk from five avian species (domestic chicken, domestic duck, domestic goose, Japanese quail or domestic pigeon) and to optimize the concentration of egg yolk on the cryopreservation of bull sperm in terms of frozen-thawed sperm progressive motility and viability. The results were two-fold. First, they showed that pigeon egg yolk provided the best cryoprotective effects on the cryopreservation of bull sperm, compared with egg yolk of chicken, quail, goose or duck. Second, the best concentration of pigeon egg yolk in extender was 20% during cryopreservation among five concentrations of 5, 10, 20, 30 or 40%. The results suggest that pigeon egg yolk could be used as an alternative to chicken egg yolk in extender but requires further testing in fertility trials.  相似文献

目的:研究KDR靶向RNA干扰对MCF-7细胞凋亡的影响,探讨其可能的机制。方法:采用阳离子脂质体Lipofecta.mine2000TM作为转染试剂将人KDR基因的siRNA转染人类乳腺细胞株MCF-7,诱RNAi,采用Hoechst33258染色和半定量RT—PCR检测Caspase-3、survivin的mRNA表达及细胞凋亡变化;比色法检测Caspase.3的活性;利用免疫组织化学方法检测survivin的表达,并用图像分析仪分析蛋白表达强度。结果:靶向KDR的siRNA转染MCF-7后,Caspase-3的mRNA表达上调,survivin基因mRNA及蛋白表达水平下调(P〈0.05)。结论:KDRsiRNA通过减少乳腺癌细胞survivin的表达,增加Caspase.3表达来促进肿瘤细胞凋亡,发挥其抗肿瘤作用。  相似文献
