The tumor suppressor p53 is important in the decision to either arrest cell cycle progression or induce apoptosis in response to a variety of stimuli. p53 posttranslational modifications and association with other proteins have been implicated in the regulation of its stability and transactivation activity. Here we show that p53 is phosphorylated by the mitotic kinase Aurora-A at serine 215. Unlike most identified phosphorylation sites of p53 that positively associate with p53 function (Brooks, C. L., and Gu, W. (2003) Curr. Opin. Cell Biol. 15, 164-171), the phosphorylation of p53 by Aurora-A at Ser-215 abrogates p53 DNA binding and transactivation activity. Downstream target genes of p53, such as p21Cip/WAF1 and PTEN, were inhibited by Aurora-A in a Ser-215 phosphorylation-dependent manner (i.e. phosphomimic p53-S215D lost and non-phosphorylatable p53-S215A retained normal p53 function). As a result, Aurora-A overrides the apoptosis and cell cycle arrest induced by cisplatin and gamma-irradiation, respectively. However, the effect of Aurora-A on p53 DNA binding and transactivation activity was not affected by phosphorylation of Ser-315, a recently identified Aurora-A phosphorylation site of p53 (Katayama, H., Sasai, K., Kawai, H., Yuan, Z. M., Bondaruk, J., Suzuki, F., Fujii, S., Arlinghaus, R. B., Czerniak, B. A., and Sen, S. (2004) Nat. Genet. 36, 55-62). Our data indicate that phosphorylation of p53 at Ser-215 by Aurora-A is a major mechanism to inactivate p53 and can provide a molecular insight for Aurora-A function. 相似文献

海洋野生鱼与养殖鱼比较, 其鱼油中含更多的二十碳五烯酸(EPA)、二十二碳六烯酸(DHA)、脂溶性维生素等活性成分。为提高海洋野生鱼的利用价值, 以野生小带鱼为原料进行酶法提油工艺研究。分析了不同的温度, 时间, pH值等影响因素下的提取、萃取以及离心效果, 以响应面法确定了最佳的酶解工艺条件: 液固比为6、pH7.3、酶量1000 u/g原料、搅拌速度200 r/min、45oC酶解90 min; 最优萃取条件: 萃取剂100 mL(每20 g鱼糜原料)、pH4.0、40oC萃取25 min; 离心条件: 离心速度3000 r/min (1865 g)、离心时间10 min。上述工艺条件下提油率为79.90%。改进了传统的鱼油提取工艺, 在活性成分保护上有较大改善。 相似文献

We have previously demonstrated that the two Exonuclease III (Xth) family members present within the obligate human pathogen Neisseria meningitidis, NApe and NExo, are important for survival under conditions of oxidative stress. Of these, only NApe possesses AP endonuclease activity, while the primary function of NExo remained unclear. We now reveal further functional specialization at the level of 3'-PO(4) processing for NExo. We demonstrate that the bi-functional meningococcal glycosylases Nth and MutM can perform strand incisions at abasic sites in addition to NApe. However, no such functional redundancy exists for the 3'-phosphatase activity of NExo, and the cytotoxicity of 3'-blocking lesions is reflected in the marked sensitivity of a mutant lacking NExo to oxidative stress, compared to strains deficient in other base excision repair enzymes. A histidine residue within NExo that is responsible for its lack of AP endonuclease activity is also important for its 3'-phosphatase activity, demonstrating an evolutionary trade off in enzyme function at the single amino acid level. This specialization of two Xth enzymes for the 3'-end processing and strand-incision reactions has not previously been observed and provides a new paradigm within the prokaryotic world for separation of these critical functions during base excision repair. 相似文献

Unexposed control subjects (n = 106), power distributing and office workers (n = 122), and manganese (Mn)-exposed ferroalloy smelter workers (n = 95) were recruited to the control, low and high groups, respectively. Mn concentrations in saliva, plasma, erythrocytes, urine and hair were significantly higher in both exposure groups than in the controls. The Fe concentration in plasma and erythrocytes, however, was significantly lower in Mn-exposed workers than in controls. The airborne Mn levels were significantly associated with Mn/Fe ratio (MIR) of erythrocytes (eMIR) (r = 0.77, p < 0.01) and plasma (pMIR) (r = 0.70, p < 0.01). The results suggest that the MIR may serve as a useful biomarker to distinguish Mn-exposed workers from the unexposed, control population. 相似文献

Rationale: Coronavirus disease 2019 (COVID-19) has caused a global pandemic. A classifier combining chest X-ray (CXR) with clinical features may serve as a rapid screening approach.Methods: The study included 512 patients with COVID-19 and 106 with influenza A/B pneumonia. A deep neural network (DNN) was applied, and deep features derived from CXR and clinical findings formed fused features for diagnosis prediction.Results: The clinical features of COVID-19 and influenza showed different patterns. Patients with COVID-19 experienced less fever, more diarrhea, and more salient hypercoagulability. Classifiers constructed using the clinical features or CXR had an area under the receiver operating curve (AUC) of 0.909 and 0.919, respectively. The diagnostic efficacy of the classifier combining the clinical features and CXR was dramatically improved and the AUC was 0.952 with 91.5% sensitivity and 81.2% specificity. Moreover, combined classifier was functional in both severe and non-serve COVID-19, with an AUC of 0.971 with 96.9% sensitivity in non-severe cases, which was on par with the computed tomography (CT)-based classifier, but had relatively inferior efficacy in severe cases compared to CT. In extension, we performed a reader study involving three experienced pulmonary physicians, artificial intelligence (AI) system demonstrated superiority in turn-around time and diagnostic accuracy compared with experienced pulmonary physicians.Conclusions: The classifier constructed using clinical and CXR features is efficient, economical, and radiation safe for distinguishing COVID-19 from influenza A/B pneumonia, serving as an ideal rapid screening tool during the COVID-19 pandemic. 相似文献

The excitation energy transfer processes in trimeric PEC have been studied by using steady-state and time-resolved fluorescence spectra techniques in detail. The results indicate that the energy transfer processes should take place between α84-PVB and β84-or β155-PCB chromophores with the time constants 34.7 ps and 175–200 ps individually; in contrast with monomeric PEC, from time-resolved fluorescence anisotropic spectrum technique, the decay constant of 45 ps which was assigned to the energy transfer time among three β84-PCB chromophores was observed and the energy levels of β84-and/or β155-PCB chromophores were confirmed to turn over in trimeric PEC. 相似文献

The carcass weight of Chinese Shanxi Black Pigs is relatively lower in comparison with that of the counterparts, i.e., Large White Pigs, although the former are resistant to harsh conditions. Since gut microflora has been recognized to play a key role in pork production, it is of interest to explore the microbial communities in different intestinal segments of pigs and its potential relatedness with host features. In this study, 16S rRNA gene amplicon sequencing, accompanied by the inter- and intra-group comparisons, was implemented to investigate the structure composition and potential functions of microbial communities of four distinct intestinal segments [duodenum (D), jejunum (J), ileum (I), and cecum (C)] between adult Chinese Shanxi Black Pigs and Large White Pigs. Comparative survey revealed that the dominant phylum in both breeds was Firmicutes, followed by Proteobacteria, and Bacteroidetes. At the genus level, Lactobacillus was predominant in all samples, and Prevotella was specifically prevalent in the cecum. Further inspection showed the differences of dominant species in the same segments between these two groups. Notably, unique taxa in C and D segments were more than that in I and J segments. Additionally, each segment was characterized by specifically enriched genera, and distinctive pathways were predicted in certain intestinal segments. In short, the findings presented a coherent picture of structure composition and predicted functionalities of gut microbiota in diverse intestinal segments of adult Chinese Shanxi Black Pigs and Large White Pigs, and extend the understanding of potential link between intestinal microbiota and their hosts. 相似文献
