本文从形态学和组织学方面研究了甘蔗幼叶胚性愈伤组织发生及体细胞胚胎的形成过程。甘蔗幼叶片切段培养于含2.4—D1.5mg/1的MS培养基上,4—6天后切段开始形成愈伤组织,约10天后愈伤组织表面出现白色颗粒状结构。将含有白色颗粒状结构的愈伤组织转移至不含激素的培养基中,7—10天后可见有小植株长出。组织学和形态学观察表明,甘蔗离体再生植株是通过体细胞胚胎发生途径。  相似文献

本文记述亚洲首次发现的猪尾鼠类化石。材料系1983年于云南禄丰古猿地点最晚中新世石灰坝组采集到的,计有代表两属三种——Platacanthomys dianensis sp.nov.,Typhlomysprimitivus sp.nov.和T.hipparionum sp.nov.的百余枚牙齿。文中除对新种的形态作了描述和对比外,还对猪尾鼠类的分类位置及系统发育作了探讨。  相似文献

The X-ray crystallographic structure of the human alpha-thrombin complex with hirulog 3 (a potent, noncleavable hirudin-based peptide of the "hirulog" class containing a beta-homoarginine at the scissile bond), which is isomorphous with that of the hirugen-thrombin crystal structure, was solved at 2.3-A resolution by starting with a model for thrombin derived from the hirugen-thrombin complex and was refined by restrained least squares methods (R = 0.132). Residues of hirulog 3 were well-defined in the electron density, which included most of the pentaglycine linker and the C-terminal helical turn that was disordered in a related structure of thrombin with hirulog 1. The interactions of D-Phe1'-Pro2'-beta-homoArg3' with the active site of thrombin were essentially identical to those of related structures of PPACK- (D-Phe-Pro-Arg chloromethyl ketone) and hirulog 1-thrombin, with the guanidinium function of the arginyl P1 residue forming a hydrogen-bonding ion pair with Asp189 of the S1 site. A noticeable shift in the CA atom of beta-homoArg3' due to the methylene insertion displaces the scissile bond from attack by Ser195, thus imparting proteolytic stability to the beta-homoArg hirulog derivative. Resolution of the pentaglycine spacer, linking N- and C-terminal functional domains into a single oligopeptide bivalent inhibitor, permitted delineation of corresponding S' subsites of thrombin. The position of Gly4' (P1') is stabilized by three hydrogen bonds with His57, Lys60F, and Ser195, while the conformational angles maintained in a strained, nonallowed configuration for non-glycyl amino acids.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

本文对云南滇西地区民间用于避孕和绝育秘方中的槌果藤,野花椒寄生、花椒寄生、螃蟹树寄生、鸡矢藤及野花椒的醇提取物进行了小鼠最大耐受量测定及小鼠抗生育实验,结果表明,花椒寄生、螃蟹树寄生的醇提取物具有明显抗生育活性,槌果藤也具有一定的抗生育作用。此外,还对槌果藤、野花椒寄生进行了化学成份预试。  相似文献

本文从构建杂种细胞14-7-1的基因组文库出发,用种特异的探针分离出含有人体基因组顺序的重组子,并进一步分析了其中13个克隆,得到8个单拷贝顺序。通过与已建立的杂种细胞克隆分布板杂交以及染色体的原位杂交方法,将1个单拷贝顺序FD11-1定位在11p11-q11上。由于已经报道在11号染色体上具有3个连锁群,它们分别位于11p15、11p13和11q13上,因此,FD11-1有可能为11号染色体连锁基因图的建立提供1个有意义的座位。  相似文献

黄皮(Clausena lansium (Lour.) Skeels)胚轴与完整种子的发育模式以及发育中电解质渗漏率变化有些不同. 种子生理成熟前、后的胚轴对脱水的反应也不同,前者经轻微脱水可提高萌发率和活力指数,后者不耐任何程度的脱水.活力指数的急剧下降伴随着电解质渗漏率的迅速上升.实验表明,黄皮种子在发育过程中没有形成耐脱水性. 细胞膜透性变化可反映脱水对种子的伤害程度  相似文献

用盆栽和遮阴试验研究了露水草(Cyanotis arachnoidea Clarke) 的光合作用特征,比较了不同遮光水平对光合速率、光合器官特性及光合产量的影响。结果如下:1.露水草是一种耐阴偏阳的C3 类草木植物,其光合作用的光饱和点约为650 μm ol·m - 2·s- 1,光补偿点约为17 μm ol·m - 2·s- 1,且具有高达130×10- 6的CO2 补偿点。2.露水草的最大净光合速率为12.45 μm ol·m - 2·s- 1。叶片净光合速率的日变化规律呈双峰曲线,主峰在11~12 时,次峰在15时左右。3.遮光20% ~50% 有利于露水草的生长。与对照相比,叶片中叶绿素b 的含量增加了47% ~83% ,并且由于净光合速率(相对光合速率)的提高,使光合生产量增加了12% ~18% 。  相似文献

Water relations, desiccation tolerance and longevity of Taxus brevifolia (Nutt.) seeds were studied to determine the optimal stage of development and storage conditions for seeds of this species. Seeds equilibrated to a range of relative humidities (RHs) had unusually low water contents which can be accounted for by the high lipid content of gametophyte tissues (71% of the dry mass). Water relations of embryonic tissue were more typical of those reported for other seed species. The water content below which freezing transitions were not observable in the embryo was ca 0.24 g H2O (g dry weight)−1 (g g−1) for all maturity classes studied. Embryos did not achieve significant levels of desiccation tolerance (survival to water contents less than 0.5 g g−1) until the latter stages of development when dry matter was maximal. Mature embryos could be dried to 0.025 g g−1 (seed water content of 0.010 g g−1) with no loss of viability. Thus, at the latter stages of development, embryo water content could be optimized to avoid both desiccation and freezing damage. Survival of mature seeds declined over a 2-year period when seeds were stored at temperatures between 5 and 35°C and RHs between 14 and 75%, corresponding to seed water contents between 0.015 and 0.07 g g−1. The deterioration rate was slowest for seeds stored at the lowest RH and temperature. Our data indicate that seeds of Taxus brevifolia show orthodox rather than recalcitrant storage characteristics, but that the optimum water content for storage was extremely low. The results suggest that even if stored at optimal water contents and low temperatures, T. brevifolia seeds will be relatively short lived. The high quantity of lipids or reducing sugars may be contributing factors in the poor storage characteristics.  相似文献

The Escherichia coli chaperonins, GroEL and GroES, as well as their complexes in the presence of a nonhydrolyzable nucleotide AMP-PNP, have been imaged with the atomic force microscope (AFM). We demonstrate that both GroEL and GroES that have been adsorbed to a mica surface can be resolved directly by the AFM in aqueous solution at room temperature. However, with glutaraldehyde fixation of already adsorbed molecules, the resolution of both GroEL and GroES was further improved, as all seven subunits were well resolved without any image processing. We also found that chemical fixation was necessary for the contact mode AFM to image GroEL/ES complexes, and in the AFM images. GroEL with GroES bound can be clearly distinguished from those without. The GroEL/ES complex was about 5 nm higher than GroEL alone, indicating a 2 nm upward movement of the apical domains of GroEL. Using a slightly larger probe force, unfixed GroEL could be dissected: the upper heptamer was removed to expose the contact surface of the two heptamers. These results clearly demonstrate the usefulness of cross-linking agents for the determination of molecular structures with the AFM. They also pave the way for using the AFM to study the structural basis for the function of GroE system and other molecular chaperones.  相似文献
