本工作采用新设计的营养缺陷型淘汰筛选法,从本实验室筛选的芽孢杆菌Bacillus R2中分离到能够水解生淀粉的β-淀粉酶基因。该基因所在的DNA片段为5.25kb,在大肠杆菌(E.coli)中的表达产物具有与供体菌相同的淀粉酶特性,实验室条件下酶产量为500IU/ml以上,RDA值为57%,并且可以全部分泌到培养基中。 相似文献

构建可表达增强型绿色荧光蛋白 (Enhanced green fluorescent protein,EGFP) 的辅助病毒依赖型腺病毒载体 (Helper-dependent adenoviral vector,HDAd),并完成大量制备、纯化和体外表达鉴定。荧光显微镜证实HDAd/EGFP可表达,电镜下观察到经CsCl纯化后的腺病毒的典型形态。分光光度计法测定病毒的浓度为4.0×1012 颗粒数 (Virus particle,vp) /mL。与可表达EGFP的第一代腺病毒载体 (First generation adenoviral vector,FGAd) FGAd/EGFP进行了体外感染和转基因表达效率的比较研究,分别用约2 000 vp/细胞的HDAd/EGFP和FGAd/EGFP感染A549细胞,流式细胞仪检测EGFP的表达情况。通过相同时间点流式细胞仪分析EGFP的表达情况,可见HDAd/EGFP感染早期的A549细胞较FGAd/EGFP有更高的荧光表达率及更高的表达强度,显示HDAd载体具有转基因瞬时高表达的特性,是一种更有价值的疫苗载体。 相似文献

Cell fate commitment of pre-implantation blastocysts, to either the inner cell mass or trophoblast, is the first step in cell lineage segregation of the developing human embryo. However, the intercellular signals that control fate determination of these cells remain obscure. Human embryonic stem cells (hESCs) provide a unique model for studying human early embryonic development. We have previously shown that Activin/Nodal signaling contributes to maintaining pluripotency of hESCs, which are derivatives of the inner cell mass. Here we further demonstrate that the inhibition of Activin/Nodal signaling results in the loss of hESC pluripotency and trophoblast differentiation, similar to BMP4-induced trophoblast differentiation from hESCs. We also show that the trophoblast induction effect of BMP4 correlates with and depends on the inhibition of Activin/Nodal signaling. However, the activation of BMP signaling is still required for trophoblast differentiation when Activin/Nodal signaling is inhibited. These data reveal that the early lineage segregation of hESCs is determined by the combinatorial signals of Activin/Nodal and BMP. 相似文献

Experiments were conducted in artificial stream tanks with juvenile Chinese sturgeon, Acipenser sinensis, captured in the Yangtze Estuary to test two null hypotheses (1) juveniles have no preference for white or black substrates, and (2) white or black substrate color has no effect on juvenile feeding efficiency on the barcheek goby,Rhinogobius giurinus, a dark-colored prey of wild juveniles. Juveniles strongly preferred white substrate. However, in feeding tests, feeding efficiency by juveniles was similar on white or black substrates. Thus, substrate color had no effect on feeding efficiency. Results suggest the innate preference for white substrate is not a visual adaptation for capturing prey on a contrasting background, but is an adaptation predisposing juveniles to seek light-colored sandy substrate where forage abundance is greatest. 相似文献

The spontaneous excitation‐emission (ExEm) spectrum is introduced to the quantitative mExEm‐spFRET methodology we recently developed as a spectral unmixing component for quantitative fluorescence resonance energy transfer measurement, named as SPEES‐FRET method. The spectral fingerprints of both donor and acceptor were measured in HepG2 cells with low autofluorescence separately expressing donor and acceptor, and the spontaneous spectral fingerprint of HEK293 cells with strong autofluoresence was measured from blank cells. SPEES‐FRET was performed on improved spectrometer‐microscope system to measure the FRET efficiency (E) and concentration ratio (R C) of acceptor to donor vales of FRET tandem plasmids in HEK293 cells, and obtained stable and consistent results with the expected values. Moreover, SPEES‐FRET always obtained stable results for the bright and dim cells coexpressing Cerulean and Venus or Cyan Fluorescent Protein (CFP)‐Bax and Yellow fluorescent protein (YFP)‐Bax, and the E values between CFP‐Bax and YFP‐Bax were 0.02 for healthy cells and 0.14 for the staurosporine (STS)‐treated apoptotic cells. Collectively, SPEES‐FRET has very strong robustness against cellular autofluorescence, and thus is applicable to quantitative evaluation on the protein‐protein interaction in living cells with strong autofluoresence. 相似文献

S-RNase在配子体型自交不亲和性反应中起关键作用,HV区段被认为是雌蕊与花粉间特异识别的关键部位。应用生物信息学方法,对蔷薇科植物的S-RNase序列分析,并对HV区段一级结构邻近区和空间邻近区作物理化学性质分析,发现HV区C端的一段氨基酸序列符合蛋白质相互作用位点的特征;HVP区也是一个多态性区段,可能参与分子识别过程。因此,蔷薇科植物中,S-RNase与花粉S基因产物的作用方式可能为S-RNase的HVC区与花粉S基因产物先非特异性结合,再以HV区和HVP区进行分子间特异识别。 相似文献

【目的】在酿酒酵母中异源表达双孢蘑菇来源的酪氨酸酶基因PPO2,并研究酪氨酸酶在酿酒酵母胞内及胞外的酶学特性。【方法】提取双孢蘑菇总RNA,通过RT-PCR克隆酪氨酸酶基因PPO2,构建表达载体pSP-G1-PPO2,并转化至酿酒酵母进行表达,采用镍亲和层析纯化蛋白并研究其酶学性质。【结果】在酿酒酵母中正确表达了大小为65 kDa的酪氨酸酶蛋白。重组酶能催化底物酪氨酸产生黑色素。体外活性测定表明,酪氨酸酶催化最适温度为45°C,以酪氨酸和多巴为底物时最适pH分别为7.0和8.0。在酿酒酵母中测得底物酪氨酸浓度低于2.5 mg/mL时,黑色素的产量与底物浓度呈现正相关性。【结论】来源于双孢蘑菇的酪氨酸酶基因PPO2在酿酒酵母中成功表达,重组酶具有良好的酶学特性。利用酪氨酸酶产物黑色素的产量与底物浓度呈现正相关性这一特性,可将其作为细胞酪氨酸产量的传感器,为高通量筛选酪氨酸高产菌株提供了思路。 相似文献

在已有全长感染性克隆pLGFD3-8和pD70344的基础上,根据马传贫弱毒疫苗致弱过程中不同代次毒株LTR序列的分析,在LTR U3区选取特定的酶切位点对EIAV非编码区LTR基因进行了部分替换.将替换的全基因克隆转染驴胎皮肤细胞(FDD)并以驴白细胞(DL)传代,用逆转录酶活性检测、RT-PCR方法及Real-time RT-PCR验证其感染性.结果发现,其衍生病毒感染上述两种细胞均出现明显的细胞病变;细胞培养上清可检测到RT酶活性和RT-PCR阳性.电镜下可见大量典型的EIAV颗粒.pLGFD9-12嵌合克隆衍生病毒与其父本克隆衍生病毒pLGFD3-8具有相似的复制水平,pLGFD9-12嵌合克隆衍生病毒在DL细胞上复制水平略高于FDD细胞.此结果为进一步深入研究LTR对马传染性贫血病毒复制水平和毒力的影响奠定了基础. 相似文献

According to the principles and methods of ecology and system engineering, we set up an evaluation indicator system for multi-component
and multiple cropping systems, evaluated the comprehensive benefits of multi-component and multiple cropping systems using
grey relation clustering analysis and screened out the optimized model based on research done in the upland red soil in Jiangxi
Agricultural University from 1984 to 2004. The results show that the grey relation degree of “cabbage/potato/maize — sesame”
was the highest among 23 multi-component and multiple cropping systems and was clustered into the optimized system. This indicates
that “cabbage/potato/maize — sesame” can bring the best social, economic and ecological benefits, increase product yield and
farmers’ income and promote sustainable development of agricultural production. Therefore, it is suitable for promotion on
upland red soil. The grey relation degree of “canola/Chinese milk vetch/maize/mung bean/maize” was second, which is suitable
for implementation at the city outskirts. In conclusion, these two planting patterns are expected to play important roles
in the reconstruction of the planting structure and optimization of the planting patterns on upland red soil.
