Eucommia ulmoides Oliver is a dioecious plant, which plays an important role in traditional Chinese medicine. However, there has not yet been any research on male and female E. ulmoides. The UPLC fingerprints and OPLS-DA approach were able to quickly and easily identify and quantify E. ulmoides and differentiate between the male and female fingerprints. In this study, we optimized the UPLC conditions and analyzed them to investigate fingerprints of twenty-four extracts of Eucommiae Cortex (EC) and twenty-four extracts of Eucommiae Folium (EF) under optimal conditions. It was demonstrated that thirteen and twelve substances were possible chemical markers for EC and EF male and female discrimination and that the level of these markers – chlorogenic acid and protocatechuic acid – was many times higher in male than in female. Circular RNAs play an important role in the development of various malignancies, including hepatocellular carcinoma (HCC). Nevertheless, the role of Hsa_circ_0093335 (circ0093335) in HCC has not yet been explored. To investigate the biological effects and molecular mechanisms of circ0093335 on HCC. Circ0093335 expression was detected in HCC cells and clinical specimens using qRT-PCR. The association between circ0093335 expression and HCC patients' clinical characteristics was determined using SPSS. The role of circ0093335 in HCC was estimated by overexpression and knockdown experiments in vitro and in vivo. qRT-PCR, nucleoplasma separation assay, FISH assay, RIP, dual luciferase reporter assay and rescue assay were used to validate the regulatory effect of circ0093335 on miR-338-5p. The study findings showed that circ0093335 was upregulated in HCC. High circ0093335 expression was linked with the tumour-node-metastasis stage and microvascular tumour invasion. circ0093335 is greatly involved in HCC cell proliferation, aggressive ability and mouse tumour growth, according to many in vitro and in vivo tests. Mechanistically, circ0093335 downregulated miR-338-5p expression by sponging, consequently promoting HCC progression. Our research indicated that circ0093335 might be a target for HCC therapy since it promotes tumour progression by acting as a miR-338-5p ‘sponge’. 相似文献

Arterial calcification is a common feature of cardiovascular disease. Sortilin is involved in the development of atherosclerosis, but the specific mechanism is unclear. In this study, we established calcification models in vivo and in vitro by using vitamin D3 and β-glycerophosphate, respectively. In vivo, the expression of SORT1 was up-regulated and the expression of miR-182 was down-regulated in calcified arterial tissues. Meanwhile there was a negative correlation between SORT1 expression and miR-182 levels. In vitro, downregulating SORT1 expression using shRNA inhibited β-glycerophosphoric induced vascular smooth muscle cells (VSMCs) calcification. Moreover, reduced sortilin levels followed transfection of miR-182 mimics, whereas there was a significant increase in sortilin levels after transfection of miR-182 inhibitors. A luciferase reporter assay confirmed that SORT1 is the direct target of miR-182. Our study suggests that SORT1 plays a vital role in the development of arterial calcification and is regulated by miR-182. 相似文献

Geometric morphometrics is a quick and reliable approach to differentiate fish stocks based on the variation of otolith shapes. In this study, morphometric analysis of otolith shapes was used to differentiate three species of Scomber. The sagittae morphology of S. scombrus otolith is totally different from that of S. japonicus and S. australasicus. Multivariate analysis consistently showed that S. japonicus was morphologically similar to S. australasicus, whereas a significant difference in otolith shapes was detected between S. scombrus and other two species of Scomber. The rostrum, antirostrum, excisural notch and dorsal-posterior margin of the otolith reflect the main variations between the three species of Scomber. Shape indices and Fourier coefficients were used to discriminate fish species using analysis of variance and Fisher discriminant analysis. The shape indices successfully differentiated 100%, 95.7% and 96.4% of otoliths in S. japonicus, S. australasicus and S. scombrus, respectively, while the Fourier coefficients only discriminated 70.0%, 61.9% and 91.3% of the sagittae in S. japonicus, S. australasicus and S. scombrus. This study indicates that the shape analysis on the sagittae morphometrics of otoliths is a better method to differentiate species of Scomber. 相似文献

目的探究中部和西部地区幼儿肠道菌群的结构差异与膳食的关系,为幼儿营养健康状况监测和营养改善工作提供有效的营养干预。方法选择“贫困地区儿童营养改善项目”河南汝阳和贵州福泉,随机抽取107名汉族健康幼儿为调查对象,食物摄入采用24 h消费调查方法。应用高通量测序技术对肠道菌群进行测序和生物信息分析,研究两县幼儿肠道菌群差异。结果两县幼儿肠道菌群组成结构较为一致,但Alpha多样性分析表明,贵州福泉县幼儿肠道物种丰富度和多样性高于河南汝阳县。细菌属水平分析中,两县幼儿肠道内均以拟杆菌属、普雷沃菌、柔嫩梭菌和双歧杆菌属为主导的菌群结构,但河南汝阳县幼儿肠道双歧杆菌属和乳杆菌属丰度显著高于贵州福泉县(12.36% vs. 7.44%;0.19% vs. 0.03%)。结论中西部地区幼儿肠道菌群结构差异不大,但有益菌含量存在显著差异,这为研究肠道菌群与膳食及营养健康状况之间的关系提供了依据。 相似文献
