炎症反应以炎症因子为代表,是骨关节炎(osteoarthritis,OA)中软骨下骨发生病变的重要机制。炎症因子半乳糖凝集素?3(galectin?3,Gal?3)会引起软骨下骨OA样变,但机制尚不清楚。利用因膝关节OA行全膝关节置换患者的胫骨平台标本,培养成骨细胞。分别在正常氧和缺氧条件下用Gal?3处理成骨细胞后,检测骨钙素、ERRαSirtuin 1的表达情况。同时,给予矿化液培养的成骨细胞以Gal?3处理,28 d后使用茜素红染色检测成骨细胞矿化程度并进行定量分析。结果显示,Gal?3抑制成骨细胞骨钙素的表达,在缺氧条件下诱导ERRαSirtuin1的表达,在正常氧条件下Gal?3促进OA成骨细胞的矿化。正常氧条件下,Gal?3可以诱导OA成骨细胞的异常矿化,缺氧条件下Gal?3促进成骨,表明Gal?3在OA成骨细胞的矿化中扮演重要角色。  相似文献

[目的]通过研究烟粉虱Bemisia tabaci取食传入植物体内的昆虫内共生菌种类,探明其在不同植物中的分布形态及时空动态.[方法]以B型烟粉虱、棉花、番茄、豇豆为实验材料,利用常规PCR检测烟粉虱取食后传入植物体内的共生菌种类;利用透射电镜(Transmission electron microscope,TEM)检测Rickettsia传入植物后的分布及形态;利用q-PCR技术检测豇豆叶片中Rickettsia含量的动态变化.[结果]B型烟粉虱体内含有原生共生菌P0rtiera、次生共生菌Ricfettsia,Hamiltonella和Hemipteriphilus,但只检测到Rickettsia可经烟粉虱传入棉花、番茄、豇豆植物体内,并可在植物体内存活、转移.在3种植物体内Rickettsia均分布于叶片韧皮部的筛管细胞中.烟粉虱、棉花、番茄组织内的Rickettsia形态基本一致,但豇豆中Rickettsia在形态上较小而钝圆.相同数量的烟粉虱取食,在豇豆体内最先检测到Rickettsia.随着烟粉虱取食时间的增加,豇豆体内的Rickettsia含量先增加后下降;而当无烟粉虱持续取食时,一定时间段内豇豆体内的Rickettsia先下降再小幅度上升,并可以在一定时间内保持不变.基于16S rDNA序列的系统发育分析表明,传入棉花、番茄、豇豆叶片中的Rickettsia与B型烟粉虱体内的Rickettsia高度同源.[结论]Rickettsia可经烟粉虱取食传入植物体内,分布并存活于韧皮部的筛管细胞中,并可在植物不同叶片之间转移;在不同植物宿主中,Rickettsia的形态会发生轻微变化;烟粉虱对Rickettsia的传播效率受到植物种类的影响.  相似文献

Nijmegen breakage syndrome (NBS) with NBS1 germ-line mutation is a human autosomal recessive disease characterized by genomic instability and enhanced cancer predisposition. The NBS1 gene codes for a protein, Nbs1(p95/Nibrin), involved in the processing/repair of DNA double-strand breaks. Hepatocellular carcinoma (HCC) is a complex and heterogeneous tumor with several genomic alterations. Recent studies have shown that heterozygous NBS1 mice exhibited a higher incidence of HCC than did wild-type mice. The objective of the present study is to assess whether NBS1 mutations play a role in the pathogenesis of human primary liver cancer, including HBV-associated HCC and intrahepatic cholangiocarcinoma (ICC). Eight missense NBS1 mutations were identified in six of 64 (9.4%) HCCs and two of 18 (11.1%) ICCs, whereas only one synonymous mutation was found in 89 control cases of cirrhosis and chronic hepatitis B. Analysis of the functional consequences of the identified NBS1 mutations in Mre11-binding domain showed loss of nuclear localization of Nbs1 partner Mre11, one of the hallmarks for Nbs1 deficiency, in one HCC and two ICCs with NBS1 mutations. Moreover, seven of the eight tumors with NBS1 mutations had at least one genetic alteration in the TP53 pathway, including TP53 mutation, MDM2 amplification, p14ARF homozygous deletion and promoter methylation, implying a synergistic effect of Nbs1 disruption and p53 inactivation. Our findings provide novel insight on the molecular pathogenesis of primary liver cancer characterized by mutation inactivation of NBS1, a DNA repair associated gene.  相似文献

蛋白质剪接技术为在蛋白质水平上直接对蛋白质进行修饰和加工提供了一种全新的解决方案,因而在蛋白质工程及相关领域具有非常广阔的应用前景。现阶段,大部分天然的蛋白质内含子在异源蛋白质中剪接活性非常低,极大限制了蛋白质内含子的开发和应用。为了开发一个可以同时对蛋白质内含子通用性和剪接活性进行筛选的系统,利用Bsa I限制性内切酶识别位点和切割不重合的特性,将Ter ThyX内含子(不含外显子序列)插入到卡那霉素抗性蛋白基因的多个位点。并且摒弃了以往需要结合天然外显子以实现剪接的方法,可以同时对蛋白质内含子的剪接活性和通用性进行筛选。Western blot结果和卡那霉素平板生长结果表明,通过卡那霉素筛选系统可以精确的将蛋白质内含子剪接反应与卡那霉素抗性结合起来,仅从卡那霉素平板上的菌落生长情况即可完成蛋白质内含子剪接活性阳性突变的筛选,是一个快速,稳定的定向进化筛选系统。  相似文献

DNA sequence variation within human leukocyte antigen (HLA) genes mediate susceptibility to a wide range of human diseases. The complex genetic structure of the major histocompatibility complex (MHC) makes it difficult, however, to collect genotyping data in large cohorts. Long-range linkage disequilibrium between HLA loci and SNP markers across the major histocompatibility complex (MHC) region offers an alternative approach through imputation to interrogate HLA variation in existing GWAS data sets. Here we describe a computational strategy, SNP2HLA, to impute classical alleles and amino acid polymorphisms at class I (HLA-A, -B, -C) and class II (-DPA1, -DPB1, -DQA1, -DQB1, and -DRB1) loci. To characterize performance of SNP2HLA, we constructed two European ancestry reference panels, one based on data collected in HapMap-CEPH pedigrees (90 individuals) and another based on data collected by the Type 1 Diabetes Genetics Consortium (T1DGC, 5,225 individuals). We imputed HLA alleles in an independent data set from the British 1958 Birth Cohort (N = 918) with gold standard four-digit HLA types and SNPs genotyped using the Affymetrix GeneChip 500 K and Illumina Immunochip microarrays. We demonstrate that the sample size of the reference panel, rather than SNP density of the genotyping platform, is critical to achieve high imputation accuracy. Using the larger T1DGC reference panel, the average accuracy at four-digit resolution is 94.7% using the low-density Affymetrix GeneChip 500 K, and 96.7% using the high-density Illumina Immunochip. For amino acid polymorphisms within HLA genes, we achieve 98.6% and 99.3% accuracy using the Affymetrix GeneChip 500 K and Illumina Immunochip, respectively. Finally, we demonstrate how imputation and association testing at amino acid resolution can facilitate fine-mapping of primary MHC association signals, giving a specific example from type 1 diabetes.  相似文献

采用硝普钠为NO供体,造成大鼠胰岛β细胞RIN-m的损伤,通过检测细胞活力、细胞内丙二醛(MAD)含量、总谷胱甘肽(GSH)含量和总超氧化物歧化酶(SOD)活性,探讨石参总黄酮的保护作用和机制。结果显示,石参总黄酮能明显提高损伤细胞的活力,降低细胞内MDA的含量、提高总GSH含量和SOD活力,表明石参总黄酮对NO所致RIN-m细胞的损伤具有保护作用,其机制可能与提高细胞内GSH含量与SOD活力有关。  相似文献

Background

As a serine/threonine protein kinase, p70S6K plays an important role in tumor cells. Evidence has revealed overexpression of p70S6K and phosphorylated p70S6K (p-p70S6K) in various tumor tissues, with these proteins identified as independent prognostic markers in non-small cell lung cancer (NSCLC). In this study, we explored the role of the p70S6K specific inhibitor PF-4708671 in NSCLC.

Methods

Three NSCLC cell lines (A549, SK-MES-1, and NCI-H460) were treated with PF-4708671 at five different concentrations, including 0.1μM, 0.3μM, 1μM, 3μM and 10μM, and protein levels were determined by Western-blot. Then, PF-4708671’s effects were assessed both in vitro (cell proliferation, apoptosis, cell cycle distribution, and invasion) and in vivo.

Results

The expression levels of p-p70S6K and the downstream effector S6 were significantly reduced by PF-4708671. Diametrically opposite, the downstream protein levels of BAD, Caspase3 and ERK had increased after treatment with PF-4708671. In addition, PF-4708671 drastically inhibited cell proliferation and invasion ability in A549, SK-MES-1 and NCI-H460 cells in vitro, causing cell cycle arrest in G0-G1 phase. Limited effects of PF-4708671 were observed on apoptosis in the three NSCLC cell lines assessed. Importantly, PF-4708671 could inhibit tumorigenesis in nude mice in vivo.

Conclusion

These findings demonstrated that the p70S6K specific inhibitor PF-4708671 has inhibitory effects on NSCLC tumorigenesis in vitro and in vivo. Therefore, P70S6K should be considered a new potential therapeutic target, and PF-470867 may be used as targeted drug for cancer treatment.  相似文献

在实地调查数据的基础上,本研究结合GIS空间分析技术和地理探测器模型,分析福建省入侵植物空间分布情况,以及地理与社会环境因子及其交互作用对入侵植物分布的影响.结果表明:福建省共记录入侵植物82种,其中,优势科为菊科,小蓬草、藿香蓟和空心莲子草出现频次最高.沿海区域的入侵植物物种数量多于内陆区域,福州和厦门为福建省外来入侵植物的两大热点地区.入侵植物在不同海拔均有分布,但入侵植物的种类随着海拔的升高总体呈下降趋势.地理探测器分析显示,自然环境因子中降水和社会经济因子中路网密度、人口密度是入侵植物空间分布的主要影响因子.各因子的空间交互作用会正向影响入侵植物的空间分布,这反映出入侵植物空间分布影响要素的复杂性.综上,将地理探测器应用到入侵植物研究领域是可行的,筛选出的环境指示因子可用于监测福建省入侵植物的适生区,从而为采取有效的防控措施提供科学依据.  相似文献
