A series of cis-bis-(2-chloroethylamine)platinum(II) and platinum(IV) complexes were synthesized and characterized by elemental analysis, IR, and 1H and 195Pt NMR spectroscopic techniques. Complexes were tested in vitro against murine L1210 leukemia and human ovarian A2780 cell lines and in vivo against the L1210 leukemia model. Some of these complexes showed excellent antitumor activity in both systems. However, all were inactive against cisplatin-resistant A2780/CP cells.  相似文献

Synchronously dividing cultures of the unicellular green alga Scenedesmus obtusiusculus were cultivated for 24 or 70 h in medium high (1000 μM) or low (60 μM) in phosphorus. Aliquots of AlCl3 (0, 37, 74, 111, 148, 185, or 222 μmol) were added daily to 1 l cell suspension at the end of the cell division phase. Algae were also grown in media with different pH, adjusted with HCl, in the absence of AlCl3.
Effects of Al on cell metabolism vary with the intracellular Al concentration and with the concentration of Al available per cell. When the concentration of phosphorus is low, internal concentrations of Al are high and the chlorophyll content and the net dry matter production per cell increase, whereas the photosynthesis and the cell division are increased. Presence of Al in a low P medium decreases the pH of the medium down to 4.5. There are only small effects of Al in the presence of P, due to precipitation of most of the Al with P in the medium.
Despite the Al-induced decrease of the pH of the culture medium, effects caused by Al cannot be explained as a pH effect. Instead, the Al effect may, at least to some extent, be related to a decrease in availability of P in the metabolism, due to formation of aluminium phosphate inside the cell.  相似文献

Xiphinema americanum is listed as a quarantine nematode by The European Plant Protection Organisation because of its ability to transmit quarantine viruses. Detection of this nematode in soil samples, or in mixed nematode samples extracted from soil, is therefore of paramount importance. We recently described the use of magnetic beads (Dynabeads) in combination with probes such as lectins or antibodies for recovery of small endoparasitic nematodes including Meloidogyne spp. and Globodera rostochiensis from mixed nematode samples. Here we show that magnetic capture can be used with much larger nematodes such as X. americanum. We describe lectins and antisera that bind to the surface of this nematode and show that the antisera can be used in the Dynabeads system to recover and enrich X. americanum from mixed nematode samples.  相似文献

Era is a highly conserved GTPase essential for bacterial growth. The N-terminal part of Era contains a conserved GTPase domain, whereas the C-terminal part of the protein contains an RNA- and membrane-binding domain, the KH domain. To investigate whether the binding of Era to 16S rRNA and membrane requires its GTPase activity and whether the GTPase domain is essential for these activities, the N- and C-terminal parts of the Streptococcus pneumoniae Era - Era-N (amino acids 1-185) and Era-C (amino acids 141-299), respectively - were expressed and purified. Era-C, which had completely lost GTPase activity, bound to the cytoplasmic membrane and 16S rRNA. In contrast, Era-N, which retained GTPase activity, failed to bind to RNA or membrane. These results therefore indicate that the binding of Era to RNA and membrane does not require the GTPase activity of the protein and that the RNA-binding domain is an independent, functional domain. The physiological effects of the overexpression of Era-C were assessed. The Escherichia coli cells overexpressing Era and Era-N exhibited the same growth rate as wild-type E. coli cells. In contrast, the E. coli cells overexpressing Era-C exhibited a reduced growth rate, indicating that the overexpression of Era-C inhibits cell growth. Furthermore, overexpression of era-N and era-C resulted in morphological changes. Finally, purified Era and Era-C were able to bind to poly(U) RNA, and the binding of Era to poly(U) RNA was significantly inhibited by liposome, as the amount of Era bound to the RNA decreased proportionally with the increase of liposome in the assay. Therefore, this study provides the first biochemical evidence that both binding sites are overlapping. Together, these results indicate that the RNA- and membrane-binding domain of Era is a separate, functional entity and does not require the GTPase activity or the GTPase domain of the protein for activity.  相似文献

Liu, S. QAlterations in structure of elastic laminaeof rat pulmonary arteries in hypoxic hypertension. J. Appl. Physiol. 81(5): 2147-2155, 1996.The effectof hypoxic hypertension on the remodeling process of the elasticlaminae of the rat hilar pulmonary arteries (PAs) was studied byelectron microscopy. Rats were exposed to hypoxia (10%O2) for periods of 0.5, 2, 6, 12, 48, 96, 144, and 240 h. Changes in the structure of the PA elastic laminae were examined and analyzed with respect to changes in the PAwall tensile stress. The PA blood pressure increased rapidly within thefirst several hours of hypoxia and reached a stable level within 2 days, whereas the PA wall tensile stress increased initially due toelevated blood pressure and then decreased after 48 h due to vesselwall thickening and returned to the control level after 4 days. Inassociation with these changes, the elastic laminae, which appearedhomogeneous in normal control rats, changed into structures composed ofrandomly oriented filaments and edematous contents with an increase inthe volume during the early period of hypoxia and regained theirhomogeneous appearance and normal volume after 4 days. The changes inthe elastic laminae were correlated with changes in the tensile stress.These changes were associated with a transient decrease in thestiffness of the PAs. In hypoxic rats given nifedipine, no change wasfound in the blood pressure, the tensile stress, or the structure ofthe elastic laminae of the PAs despite continuous exposure to hypoxia.These results suggested that altered tensile stress in the PA wallplayed a critical role in the initiation and regulation of structuralchanges in the elastic laminae and that these changes might contribute to alterations in the mechanical properties of the PA in hypoxic hypertension.
