Cerebellum is an important brain region involved in motor, cognition, learning and memory functions. Proteome mapping of the 21 days old rat cerebellum identified total 285 proteins, out of which 76 proteins were not reported earlier from rat brain. This includes 49 neuronal activity-specific proteins, 7 of which are reported for the first time from the cerebellum in this study. The protein sequence data for 31 proteins reported here have been integrated in the UniProt Knowledgebase.  相似文献

One of the mechanisms for establishment of infection employed by intra-macrophage pathogen-like Leishmania is inhibition of oxidative burst-mediated macrophage apoptosis to protect their niche for survival and replication. We tried to elucidate the underlying mechanism for this by using H2O2 for induction of apoptosis. Leishmania donovani-infected macrophages were much more resistant to H2O2-mediated apoptosis compared with control. Although infected cells were capable of comparable reactive oxygen species production, there was less activation of the downstream cascade consisting of caspase-3 and -7 and cleaved poly(ADP)-ribose polymerase. Suppressors of cytokine signaling (SOCS) 1 and 3 proteins and reactive oxygen species scavenging enzyme thioredoxin, known to be involved in stabilization of protein-tyrosine phosphatases, were found to be induced during infection. Induction of SOCS proteins may be mediated by Egr1, and silencing of Socs1 and -3 either alone or in combination resulted in reduced thioredoxin levels, enhanced activation of caspases, and increased apoptosis of infected macrophages. The induction of protein-tyrosine phosphatases, thioredoxin, SOCS, and Egr1 in L. donovani-infected macrophages was found to be unaffected by H2O2 treatment. SOCS knocked down cells also displayed decreased parasite survival thus marking reduction in disease progression. Taken together, these results suggest that L. donovani may exploit SOCS for subverting macrophage apoptotic machinery toward establishing its replicative niche inside the host.  相似文献

Chytridiomycosis is an emerging infectious disease of amphibians caused by the waterborne pathogen Batrachochytrium dendrobatidis (Bd) and is responsible for the recent decline of species worldwide. Elucidating patterns in disease prevalence has proved challenging, as small‐scale studies to date have provided conflicting results. We present data on the prevalence of Bd (the proportion of individuals with Bd) collected from amphibians sampled throughout British Columbia. We had two different datasets: our original dataset includes 1129 individuals sampled in 103 sites representing 14 species. A second dataset has 839 individuals sampled in 19 sites representing 10 species. We use a Bayesian state‐space occupancy modelling approach to estimate the probability that an individual is Bd+ as a function of individual and site characteristics. Cross‐validation techniques and the original dataset were used to find the best model: this model includes species, life stage, and geographic location. Our results suggest that Bd prevalence is not strongly related to seasonality, latitude or site type. Within a species, Bd prevalence depended on life stage; the watershed in which a site occurs may also usefully predict prevalence. Overall observed infection prevalence was ~16%. Our best model accurately assigns Bd status to an individual ~ 42% of the time. Taking advantage of the Bayesian framework, we ran an analysis with the second dataset using estimates from the original model as prior values. We present posterior density distributions for those sites and species with narrow credible intervals, and show that sites tend to be either highly likely or highly unlikely to have individuals with Bd, while individuals of some stages of some species have an intermediate likelihood of being Bd‐positive. The Bayesian model using informed priors had increased accuracy rates in assigning Bd status to both individuals and groups of individuals.  相似文献

The genetic fidelity of in vitro-raised plants of three successive regenerations of Nepenthes khasiana Hook. f. was assessed using three different single primer amplification reaction (SPAR) methods, viz., random amplified polymorphic DNA (RAPD), inter-simple sequence repeat (ISSR) and direct amplification of minisatellite DNA region (DAMD) markers. Out of 80 RAPD primers screened, 14 primers reflected a genetic variation of 4.1% in the first regeneration which was increased to 9.4% in the third regeneration. In the case of ISSR, out of 36 primers screened for assessment of genetic homogeneity of the regenerated plantlets, 12 primers showed an increase of genetic variation from 4.3% to 10% from the first to the third regenerations. In DAMD profiling, 15 primers were used for the evaluation of genetic fidelity where 8.47% of polymorphism was observed in the first regeneration which was increased to 13.33% in the third regeneration. The cumulative analysis reflected a genetic variation of 5.65% in the first regeneration which increased subsequently to 7.77% in the second regeneration and 10.87% in the third regeneration. The present study demonstrates SPAR technique to be an efficient tool for the assessment of clonal fidelity of in vitro-raised plants.  相似文献

Cryoconite holes have biogeochemical, ecological and biotechnological importance. This communication presents results on culturable psychrophilic bacterial diversity from cryoconite holes at Midre Lovénbreen (ML), Austre Brøggerbreen (AB), and Vestre Brøggerbreen (VB) glaciers. The culturable bacterial count ranged from 2.7 × 103 to 8.8 × 104 CFUs/g while the total bacterial numbers ranged from 5.07 × 105 to 1.50 × 106 cells at the three glaciers. A total of 35 morphologically distinct bacterial isolates were isolated. Based on 16S rRNA gene sequence data, the identified species belonged to eight genera namely Pseudomonas, Polaromonas, Micrococcus, Subtercola, Agreia, Leifsonia, Cryobacterium and Flavobacterium. The isolates varied in their growth temperature, NaCl tolerance, growth pH, enzyme activities, carbon utilization and antibiotic sensitivity tests. Fatty acid profiles indicate the predominance of branched fatty acids in the isolates. To the best of our knowledge, this is the first record of culturable bacterial communities and their characterization from glacier cryoconites from High Arctic. High amylase and protease activities expressed by Micrococcus sp. MLB-41 and amylase, protease and lipase activities expressed by Cryobacterium sp. MLB-32 provide a clue to the potential applications of these organisms. These cold-adapted enzymes may provide an opportunity for the prospect of biotechnology in Arctic.  相似文献

According to new radiometric determinations, the Chorhat Sandstone (Kheinjua Formation, Semri Group) is much older (1628 and 1632 Ma) than previously thought. This makes the carbonaceous fossils here described not only the oldest, but also the only reliably dated fossil record of the Vindhyan Supergroup, India. The new fossils represent a Chuaria-Tawuia assemblage with other carbonaceous remains of uncertain biologic affinities. Their size range (0.02-3.5 mm) indicates a transitional phase in the evolution from micro- to mega scopic organisms. Alternatively, low oxygen levels may have induced small size.  相似文献

The optical properties of colloidal ZnO nanoparticle (NP) solutions, with size ranging from several nm to around 200 nm, have been tailored to have high optical nonlinearity for bioimaging with no auto‐fluorescence above 750 nm and minimal auto‐fluorescence below 750 nm. The high second harmonic conversion efficiency enables selective tissue imaging and cell tracking using tunable near‐infrared femtosecond laser source ranging from 750‐980 nm. For laser energies exceeding the two‐photon energy of the bandgap of ZnO (half of 3.34 eV), the SHG signal greatly decreases and the two‐photon emission becomes the dominant signal. The heat generated due to two‐photon absorption within the ZnO NPs enable selective cell or localized tissue destruction using excitation wavelength ranging from 710–750 nm.
