The forward-mutation assay using the L-arabinose-sensitive strain SV3 of Salmonella typhimurium has been calibrated against a selected set of mutagens. Strain SV3 is sensitive to chemicals causing base-pair substitutions, frameshift mutations and deletions. New strains deficient for the excision-repair system or the lipopolysaccharide barrier or both have been selected from strain SV3. The additional mutations do not affect the independence of the assay from experimental artifacts due to physiological or lethal damage or differences in plating density. The new strains are more sensitive than SV3 to certain mutagens. Techniques for using this set of strains are presented and their relative advantages discussed. 相似文献

Comparison of the evolutionary dynamics of symbiotic and housekeeping loci: a case for the genetic coherence of rhizobial lineages 总被引:6,自引:1,他引:5
In prokaryotes, lateral gene transfer across chromosomal lineages may be
mediated by plasmids, phages, transposable elements, and other accessory
DNA elements. However, the importance of such transfer and the evolutionary
forces that may restrict gene exchange remain largely unexplored in native
settings. In this study, tests of phylogenetic congruence are employed to
explore the range of horizontal transfer of symbiotic (sym) loci among
distinct chromosomal lineages of native rhizobia, the nitrogen-fixing
symbiont of legumes. Rhizobial strains isolated from nodules of several
host plant genera were sequenced at three loci: symbiotic nodulation genes
(nodB and nodC), the chromosomal housekeeping locus glutamine synthetase II
(GSII), and a portion of the 16S rRNA gene. Molecular phylogenetic analysis
shows that each locus generally subdivides strains into the same major
groups, which correspond to the genera Rhizobium, Sinorhizobium, and
Mesorhizobium. This broad phylogenetic congruence indicates a lack of
lateral transfer across major chromosomal subdivisions, and it contrasts
with previous studies of agricultural populations showing broad transfer of
sym loci across divergent chromosomal lineages. A general correspondence of
the three rhizobial genera with major legume groups suggests that host
plant associations may be important in the differentiation of rhizobial nod
and chromosomal loci and may restrict lateral transfer among strains. The
second major result is a significant incongruence of nod and GSII
phylogenies within rhizobial subdivisions, which strongly suggests
horizontal transfer of nod genes among congenerics. This combined evidence
for lateral gene transfer within, but not between, genetic subdivisions
supports the view that rhizobial genera are "reproductively isolated" and
diverge independently. Differences across rhizobial genera in the
specificity of host associations imply that the evolutionary dynamics of
the symbiosis vary considerably across lineages in native settings.

A map of melon (Cucumis melo L.) with 411 markers (234 RFLPs, 94 AFLPs, 47 RAPDs, 29 SSRs, five inter-SSRs, and two isozymes) and one morphological trait (carpel number) was constructed using the F2 progeny of a cross between the Korean accession P1161375 and the Spanish melon type 'Pinyonet Piel de Sapo'. RFLPs were obtained using 212 probes from different genomic and cDNA melon libraries, including 16 Arabidopsis ESTs, 13 Cucumis known genes, and three resistant gene homologues. Most loci (391) mapped to 12 major linkage groups, spanning a total genetic distance of 1197 cM, with an average map interval of 3 cM/marker. The remaining 21 loci (six RAPDs and 15 AFLPs) were not linked. A majority (66%) of the markers were codominant (RFLPs, SSRs, and isozymes), making them easily transferable to other melon crosses. Such markers can be used as a reference, to merge other melon and cucumber maps already constructed. Indeed, some of them (23 SSRs, 14 RFLPs, one isozyme, and one morphological trait) could act as anchor points with other published cucurbit maps. 相似文献

Background
Increasingly lignocellulosic biomass hydrolysates are used as the feedstock for industrial fermentations. These biomass hydrolysates are complex mixtures of different fermentable sugars, but also inhibitors and salts that affect the performance of the microbial production host. The performance of six industrially relevant microorganisms, i.e. two bacteria (Escherichia coli and Corynebacterium glutamicum), two yeasts (Saccharomyces cerevisiae and Pichia stipitis) and two fungi (Aspergillus niger and Trichoderma reesei) were compared for their (i) ability to utilize monosaccharides present in lignocellulosic hydrolysates, (ii) resistance against inhibitors present in lignocellulosic hydrolysates, (iii) their ability to utilize and grow on different feedstock hydrolysates (corn stover, wheat straw, sugar cane bagasse and willow wood). The feedstock hydrolysates were generated in two manners: (i) thermal pretreatment under mild acid conditions followed by enzymatic hydrolysis and (ii) a non-enzymatic method in which the lignocellulosic biomass is pretreated and hydrolyzed by concentrated sulfuric acid. Plant–plant interactions are among the fundamental processes that shape structure and functioning of arid and semi‐arid plant communities. Despite the large amount of studies that have assessed the relationship between plant–plant interactions (i.e., facilitation and competition) and diversity, often researchers forget a third kind of interaction, known as allelopathy. We examined the effect of plant–plant interactions of three dominant species: the perennial grass Lygeum spartum, the allelopathic dwarf shrub Artemisia herba‐alba, and the nurse shrub Salsola vermiculata, on plant diversity and species composition in a semi‐arid ecosystem in NE Spain. Specifically, we quantified the interaction outcome (IO) based on species co‐occurrence, we analyzed diversity by calculation of the individual species–area relationship (ISAR), and compositional changes by calculation of the Chao‐Jaccard similarity index. We found that S. vermiculata had more positive IO values than L. spartum, and A. herba‐alba had values between them. Lygeum spartum and A. herba‐alba acted as diversity repellers, whereas S. vermiculata acted as a diversity accumulator. As aridity increased, A. herba‐alba transitioned from diversity repeller to neutral and S. vermiculata transitioned from neutral to diversity accumulator, while L. spartum remained as diversity repeller. Artemisia herba‐alba had more perennial grass species in its local neighborhood than expected by the null model, suggesting some tolerance of this group to its “chemical neighbor”. Consequently, species that coexist with A. herba‐alba were very similar among different A. herba‐alba individuals. Our findings highlight the role of the nurse shrub S. vermiculata as ecosystem engineer, creating and maintaining patches of diversity, as well as the complex mechanism that an allelopathic plant may have on diversity and species assemblage. Aim In their recent paper, Kissling & Carl (2008 ) recommended the spatial error simultaneous autoregressive model (SARerr ) over ordinary least squares (OLS) for modelling species distribution. We compared these models with the generalized least squares model (GLS) and a variant of SAR (SARvario ). GLS and SARvario are superior to standard implementations of SAR because the spatial covariance structure is described by a semivariogram model.
Innovation We used the complete datasets employed by Kissling & Carl (2008 ), with strong spatial autocorrelation, and two datasets in which the spatial structure was degraded by sample reduction and grid coarsening. GLS performed consistently better than OLS, SARerr and SARvario in all datasets, especially in terms of goodness of fit. SARvario was marginally better than SARerr in the degraded datasets.
