FTO is a nuclear protein belonging to the AlkB-related non-haem iron- and 2-oxoglutarate-dependent dioxygenase family. Although polymorphisms within the first intron of the FTO gene have been associated with obesity, the physiological role of FTO remains unknown. Here we show that a R316Q mutation, inactivating FTO enzymatic activity, is responsible for an autosomal-recessive lethal syndrome. Cultured skin fibroblasts from affected subjects showed impaired proliferation and accelerated senescence. These findings indicate that FTO is essential for normal development of the central nervous and cardiovascular systems in human and establish that a mutation in a human member of the AlkB-related dioxygenase family results in a severe polymalformation syndrome. 相似文献

This study investigated the relationship between spatial variations in predation risk and abundance of northern redbelly dace Phoxinus eos at both macroscale (littoral v. pelagic zones) and microscale (structured v. open water habitats in the littoral zone) of Canadian Shield lakes. Minnow traps were placed in both structured and open water habitats in the littoral zone of 13 Canadian Shield lakes, and estimates of the relative predation risk of P. eos in both the pelagic and the littoral zones were obtained from tethering experiments. Results showed that (1) the mean abundance of P. eos in the littoral zone was positively correlated with the relative predation risk in the pelagic zone, (2) P. eos preferentially used structured over open water habitats in the littoral zone and (3) this preference was not related to the relative predation risk in the littoral zone but decreased as the relative predation risk increased in the pelagic zone. At the lake level, these results support the hypothesis that P. eos enter the littoral zone to avoid pelagic piscivores. At the littoral zone level, the results do not necessarily contradict the widely accepted view that P. eos preferentially use structured over open habitats to reduce their predation risk, but suggest that flexibility in antipredator tactics ( e.g. shelter use v. shoaling) could explain the spatial distribution of P. eos between structured and open water habitats. 相似文献

Few methods for quantifying the dynamics of temporal processes are readily applicable to spatially extended systems when equations governing the motion are unknown. The objective of this paper is to illustrate how the MRP-RQA (multivariate recurrence plot-recurrence quantification analysis) approach may serve to characterize ecosystems driven by both deterministic and stochastic forces. The strength of the MRP-RQA approach resides in its independence from constraining assumptions regarding outliers, noise, stationarity and transients. Its utility is demonstrated by means of two spatiotemporal series (summer and spring datasets) of light intensity variations in an old growth forest ecosystem. Results revealed qualitative differences in homogeneity, transiency, and drift typologies between the MRPs derived from each dataset. RQA estimates of determinism and Kolmogorov entropy supported the idea that mixed chaotic–stochastic dynamics may be common in mesoscale forest habitats. Advantages and inconveniences of the MRP-RQA approach are also discussed in the more general context of monitoring ecosystems. 相似文献

Can objects or events ever capture one''s attention in a purely stimulus-driven manner? A recent review of the literature set out the criteria required to find stimulus-driven attentional capture independent of goal-directed influences, and concluded that no published study has satisfied that criteria. Here visual search experiments assessed whether an irrelevantly large object can capture attention. Capture of attention by this static visual feature was found. The results suggest that a large object can indeed capture attention in a stimulus-driven manner and independent of displaywide features of the task that might encourage a goal-directed bias for large items. It is concluded that these results are either consistent with the stimulus-driven criteria published previously or alternatively consistent with a flexible, goal-directed mechanism of saliency detection. 相似文献

Lymphatic vessels play an essential role in intestinal lipid uptake, and impairment of lymphatic vessel function leads to enhanced adipose tissue accumulation in patients with lymphedema and in genetic mouse models of lymphatic dysfunction. However, the effects of obesity on lymphatic function have been poorly studied. We investigated if and how adipose tissue accumulation influences lymphatic function. Using a lymphatic specific tracer, we performed in vivo near-infrared (NIR) imaging to assess the function of collecting lymphatic vessels in mice fed normal chow or high-fat diet (HFD). Histological and whole mount analyses were performed to investigate the morphological changes in initial and the collecting lymphatic vessels. HFD was associated with impaired collecting lymphatic vessel function, as evidenced by reduced frequency of contractions and diminished response to mechanostimulation. Moreover, we found a significant negative correlation between collecting lymphatic vessel function and body weight. Whole mount analyses showed an enlargement of contractile collecting lymphatic vessels of the hind limb. In K14-VEGF-C mice, HFD resulted in a reduced spreading of the tracer within dermal lymphatic vessels. These findings indicate that adipose tissue expansion due to HFD leads to a functional impairment of the lymphatic vasculature, predominantly in collecting lymphatic vessels. 相似文献

The first goal of this study was to determine whether morphological variation in the freshwater blenny Salaria fluviatilis results in spatially structured populations distributed around Corsica, France, which would suggest genetically differentiated populations through reproductive isolation by distance. The second goal was to determine whether some morphological traits are related to water velocity, one of the most contrasting habitat characteristics in these rivers, which would suggest an adaptation to local conditions. The results showed that the morphology of S. fluviatilis differed among the three main geographic areas studied in Corsica and that geographically distant populations of S. fluviatilis were less similar morphologically and genetically than close ones. The results also indicated that the morphological differences among populations conformed to functional expectations. Overall, the results suggest that the morphological variation of S. fluviatilis from Corsican rivers is an adaptive response to water velocity and that these populations are in a process of reproductive isolation by distance. 相似文献

Transcranial direct current stimulation (tDCS) is a neuromodulation technique that has been increasingly used over the past decade in the treatment of neurological and psychiatric disorders such as stroke and depression. Yet, the mechanisms underlying its ability to modulate brain excitability to improve clinical symptoms remains poorly understood 33. To help improve this understanding, proton magnetic resonance spectroscopy (1H-MRS) can be used as it allows the in vivo quantification of brain metabolites such as γ-aminobutyric acid (GABA) and glutamate in a region-specific manner 41. In fact, a recent study demonstrated that 1H-MRS is indeed a powerful means to better understand the effects of tDCS on neurotransmitter concentration 34. This article aims to describe the complete protocol for combining tDCS (NeuroConn MR compatible stimulator) with 1H-MRS at 3 T using a MEGA-PRESS sequence. We will describe the impact of a protocol that has shown great promise for the treatment of motor dysfunctions after stroke, which consists of bilateral stimulation of primary motor cortices 27,30,31. Methodological factors to consider and possible modifications to the protocol are also discussed. 相似文献

The cardiolipin phosphodiesterase of Escherichia coli was further characterized. This enzyme has a pH optimum of 7.0 and is Mg2+ dependent. Mn2+ and Co2+ could replace Mg2+ but other divalent cations were inhibitory or without effect. The enzyme is not periplasmic and does not appear to be associated with membrane fractions prepared by different methods. It is recovered as a soluble protein in the cytosol fraction but could not be readily purified because of its instability. With cell-free systems, a requirement for ATP or ADP could be shown under certain defined conditions. Other nucleotides were less effective or ineffective in stimulating the phosphodiesterase. The cells displayed the highest activity during the middle to late exponential stage but no marked requirement for ATP was apparent when the phosphodiesterase was obtained from such freshly grown cells. If, however, cells were starved for several hours in saline medium, the cardiolipin phosphodiesterase level fell and a requirement for added ATP could be shown. The cardiolipin phosphodiesterase is an enzyme distinct from cardiolipin synthase. The assay conditions are quite different from each of these enzymes as are their subcellular distributions. 相似文献

Microalgal cultures offer an interesting alternative for waste water treatment (urban, industrial or agricultural effluents) because they provide a tertiary biotreatment coupled with the production of potentially valuable biomass, which can be used for several purposes. We review the main abiotic, biotic and operative factors playing a role in the cultivation of microalgae. Various types of bioreactors are scrutinized keeping in view that the main limitation upon the type of usable bioreactors is the enormous volume of water to be treated. The choice of suitable microalgae and cyanobacteria is examined in terms of productivity and easiness of harvesting. The possible alternatives to harvesting are also reviewed with an emphasis on immobilized systems. Finally, the need for more research and development is discussed.This paper was presented at the Symposium on Applied Phycology at the Fourth International Phycological Congress, Duke University. 相似文献
