Phosphorylation of ankyrin decreases its affinity for spectrin tetramer

The effects of phosphorylation on the interaction between spectrin and ankyrin were investigated. Spectrin and ankyrin were phosphorylated using purified human erythrocyte membrane and cytosolic (casein kinase A) kinases. These two kinases have similar properties as well as activities toward spectrin and ankyrin. Both kinases catalyzed the incorporation of about 2 mol of phosphate/mol of spectrin and about 7 mol of phosphate/mol of ankyrin. These phosphates were incorporated primarily into seryl and threonyl residues of the proteins. The phosphopeptide maps of ankyrin phosphorylated by the membrane kinase and casein kinase A were identical. Binding studies indicate that ankyrin exhibits different affinities for spectrin dimers (KD = 2.5 +/- 0.9 X 10(-6) M) and tetramers (KD = 2.7 +/- 0.8 X 10(-7) M). These dissociation constants were not appreciably affected by the phosphorylation of spectrin. On the other hand, phosphorylation of ankyrin was found to significantly reduce its affinity for either phosphorylated or unphosphorylated spectrin tetramers (KD = 1.2 +/- 0.1 X 10(-6) M) but not spectrin dimers (KD = 2.5 +/- 0.4 X 10(-6) M). The same results were obtained using either the membrane kinase or casein kinase A as the phosphorylating enzyme. The above observation suggests that ankyrin phosphorylation may provide an important mechanism for the regulation of the erythrocyte membrane cytoskeletal network.     

Epithelial differentiation at the edentulous alveolar ridge in man

Summary The epithelial lining of the mucosa of the edentulous, maxillary alveolar ridge was subjected to an ultrastructural and stereological analysis. Four biopsies collected from the non-inflamed crest, i.e., the center over former tooth sockets, in non-denture-wearing female patients 30 to 55 years of age were processed for light and electron microscopy. At the light-microscopic level, epithelial thickness was determined histometrically. Electron micrographs were sampled at two levels of magnification, from five strata in regions of epithelial ridges and from three strata over connective tissue papillae. Standardized stereological pointcounting techniques were employed to analyze a total of 990 electron micrographs. Observations and data revealed that at the alveolar ridge the oral epithelium is truly keratinizing and comprises four strata including a 40±5 m-thick stratum corneum, which displays the oral keratin pattern. The histoand cytodifferentiation were peculiar: (1) Compared to the neighbouring gingival and hard palate epithelium, that of the alveolar crest was markedly thicker, with elongated rete ridges indicating acanthosis. (2) The cytoarchitecture was identical neither to the gingival nor to the hard palate epithelium but revealed a mixture of features typical for either of these two epithelia. Reasons for this are explained on the basis of factors, possible genetic, inherent in epithelial cells that are possibly derived from both the gingival and the palatal environment.

A comparative study of the chromosomes in the incipient species of the Drosophila paulistorum complex

Summary Drosophila paulistorum Dobzhansky et Pavan is a complex of six races or incipient species. The races are mostly allopatric, but they are reproductively isolated sufficiently to permit them to exist also sympatrically in some places. The gene arrangements in the chromosomes of the races have been compared by means of examination of the giant chromosomes in the larval salivary glands; 28 strains of all races, and about an equal number of interracial hybrids have been studied.Chromosomal inversion polymorphism has been discovered in all races, even in the Guianan race of which only a single strain is available. Inversion heterozygotes are found in every one of the five chromosomal strands which the species has. Interracial hybrids tend to be heterozygous for more inversions than are present in the strains of the parental races. The Transitional race has however much the same gene arrangements as the widespread Andean — South Brazilian race.With the exception of the Transitional race, and of three other possible exceptions, each race has a collection of its own race-specific inversion polymorphs, not found in the other races. This very striking finding is discussed in connection with the hypothesis which envisages the origin of new species from marginal colonies at the periphery of the geographic distribution area of the ancestral species.The work reported in this article has been carried under Contract No. AT-(30-1)-1151, U.S. Atomic Energy Commission, mostly at the Department of Zoology, Columbia University, New York.     

Configurations resulting from iso-chromatid and iso-subchromatid unions after meiotic and mitotic prophase irradiation

Summary By making use of the chromosomes of Trillium erectum as a model, potential and actual configurations arising from presumed iso-chromatid and iso-subchromatid unions after irradiation of meiotic or mitotic prophase have been studied and analyzed. Diagrams and photographs of various recognizable types of chromatid or subchromatid rearrangements are presented. A minimum of two iso-chromatid unions within an arm of a single chromosome in meiotic prophase, if separated by a single chiasma, can give rise to a monocentric chromosome with a triplicated segment, the middle portion of which is an inversion. A minimum of two iso-subchromatid breaks within an arm at either meiotic or mitotic prophase also can result in the production of a monocentric chromosome containing a triplicated segment. The stage of appearance of dicentrics or bridges arising from chromatid or subchromatid unions in meiotic prophase is influenced by chiasma number and pattern and by the number of strands per chromosome or chromatid. Some of the rearrangements described may have genetic and evolutionary implication of considerable potential importance which has not been recognized previously.Research carried out at Brookhaven National Laboratory under the auspices of the U.S. Atomic Energy Commission.

Zusammenfassung Werden Keimlinge vonHelianthus annuus undVicia faba mittels einer Wasserstrahlpumpe mit Wasser infiltriert, so führt dies sofort in allen Organen der Pflanze zu einer sehr starken und mitunter völligen Hemmung des Wachstums. Wirkt der Unterdruck in Luft ein, so daß es hernach zu keiner Wasserfüllung der Interzellularen kommt, so unterbleibt jede Wachstumshemmung.Die Frage nach der Kausalbeziehung zwischen Infiltration und Wachstumshemmung konnte nicht geklärt werden, da die nächstiliegende Annahme, Infiltration führe zu einer Atmungshemmung, durch das Experiment nicht bestätigt werden konnte.Es wird darauf hingewiesen, daß die Zufuhr von Wirk- oder Nährstoffen durch Infiltration eine Methode ist, die in wachsenden Organen nur mit großem Vorbehalt angewendet werden darf, da eine im Wachstum weitgehend gehemmte Pflanze sich in einem anomalen Zustand befindet.Mit 8 Textabbildungen.
