Extracellular glycosaminoglycans (GAG) released by chick embryonic fibroblasts

Summary Administration of Concanavalin A (Con A) to cultured skin fibroblasts derived from chick embryos at two developmental stages produce variations in the relative concentration of individual glycosaminoglycan (GAG) secreted by the cells. This effect is different: at 7 days (increase of hyaluronic acid and dermatan sulphate and decrease of chondroitin sulphate) and at 14 days (dermatan sulphate is not detectable).All the cells bind the Con A specifically, but a different pattern of agglutination is present in fibroblasts of the two embryonic ages. Since Con A is well known to bind carbohydrate-containing surface proteins, the result suggests that the release of GAG by chick embryonic fibroblasts can be modulated by cell surface receptors.     

Ultrastructure of rat pituitary LH gonadotrophs in relation to serum and pituitary LH levels following repeated LH-RH stimulation

The effects of single and repeated LH-RH injections at 120 min intervals on female rat LH gonadotrophs and on pituitary and serum LH levels were investigated using electronmicroscopy and radioimmunoassay. A temporary stimulation of granule release, of protein and new granule synthesis and of the accumulation of lysosomal structures was found in LH cells after the first LH-RH injection. The temporary stimulations were massively enhanced after the second injection. These consecutive yet in their time-sequence overlapping processes account for the initial depletion of secretory granule content (3--15 min after LH-RH injection), for the subsequent regranulation and accumulation of granules above control levels (60--120 min after injection) and also for the reduction in the number of granules to control levels (150 min after LH-RH injection and thereafter). Increased polymorphic lysosomal structures are believed to be responsible for this reduction of excess granules. The amount of assayable pituitary and serum LH generally corresponds with the morphological changes observed in LH-gonadotrophs, thus further substantiating the above observations. A schema which summarizes the observed morphological and hormonal changes in their time-sequence in response to LH-RH stimulation depicts the short-term regulation of secretory processes in female gonadotrophs.     

A Golgi study of the hypothalamus of Actinopterygii

The posterior hypothalami of the polypteriform, Calamoichthys, and of the teleost, Anguilla, were studied by means of the Golgi technique. In Calamoichthys, the lateral lobes are not developed and the median lobe is simple. In Anguilla, the median (tuberal) lobe shows lophodendritic, CSF-contacting cells and horizontal cells in the periventricular grey and some reticular elements directed toward the cell-poor lateral areas. In the lateral lobes the periventricular grey is formed by multipolar neurons and a diffuse population of multipolar cells of uncertain identity. The nucleus diffusus lobi lateralis is formed by scarce multipolar neurons, often placed next to the external surface of the brain. The organization of the lateral lobes in Actinopterygii is reminiscent of highly developed integrative regions.     

Factor analysis in clusterbean (Cyamopsis tetragonoloba (L.) Taub)

Summary The pattern of diversity in forty genotypes of clusterbean (Cyamopsis tetragonoloba (L.) Taub) was analysed using the centroid method of factor analysis based on nine characters. Three factors were found to adequately account for most of the intercorrelations in the four environments. Branches, clusters, pods and seed yield/plant could be grouped as productivity factors; days to flowering, maturity and plant height as growth factors; seeds/pod and gum content as economic factor. Interestingly, the constitution of the factors were the same in all the environments. The loadings on the economic factors were relatively unstable, whereas the loadings on productivity and growth factors were stable. It is concluded that productivity and growth factors play a pivotal role towards diversity in clusterbean. The results of factor analysis provides confirmatory evidence of diversity in clusterbean, which was earlier studied by using generalized distance, and thus proves the adequacy of the centroid method in biological investigations.     

Morphological and functional peculiarities of mesenchymal cells in the pars tuberalis of the pituitary gland

Summary Following injection of high doses of horseradish peroxidase (HRP), mesenchymal cells distributed in the perisinusoidal space of the pars tuberalis of the hypophysis in cats, rabbits and Japanese quails, sequester the exogenously administrated peroxidase intensively. These cells are designated by the authors as horseradish peroxidase-uptake cells (HRP-uptake cells or HUC). HRP-uptake cells constitute a system of macrophages in the pars tuberalis of mammals and birds, and are located around the hypophysial portal veins. HRP-uptake cells differ in morphological and functional characteristics from similar cells in other parts of hypophysis. Fate of spermatozoa that do not participate in fertilization in the domestic fowl

Summary The fate of spermatozoa that do not participate in fertilization was investigated by electron microscopy. After artificial insemination, we observed several spermatozoa between the fibers of the outer layer of the vitelline membrane of the ovum. One or more spermatozoa were also found in a phagocytic vesicle of macrophages located in the intercellular space of the mucosal epithelium of the infundibulum or in the outer layer of the vitelline membrane.From these observations, we assume that the superfluous spermatozoa in the lumen of the anterior part of the oviduct might be removed by inclusion into the outer layer of the vitelline membrane and by phagocytosis by macrophages.The authors are greatly indebted to Assoc. The structure and distribution of satellite cells of cardiac muscles in decapod crustaceans

Summary The structure and distribution of satellite cells of cardiac muscles were examined in twenty-one species of animals chosen from each tribe within the order Decapoda (Arthropoda, Crustacea). The satellite cells were found in all animals observed. Most of them are morphologically identical with those described in different striated muscles of other species, but some cells have unusual features. The decapod satellite cell occasionally lies right over the region corresponding to the intercalated disc between the apposed cardiac muscle cells. The cell sends cytoplasmic processes into the adjacent muscle cells, enabling the plasma membrane to make close contact with the cleft opening of the intercalated disc, and with the myofibril at the level of the Z-line. Another characteristic feature is the presence of paired cells. Such cells are clearly separated from each other over most of the contact area by the respective plasma membranes, which are smooth in appearance and devoid of specialized regions. The significance of the presence of satellite cells in decapod cardiac muscle and its possible role are discussed and compared with those described for other species.     

The effect of a liquid elemental diet on cell proliferation in the colon of rats

Summary A liquid elemental diet (Vivonex) was given to rats for 6 days while control animals received a normal diet. At the end of the experiment each animal received one intraperitoneal injection of tritiated thvmidine at 8a.m. Animals from each group were killed hourly during the first 24h after the injection and the proliferative activity was studied by autoradiography of the mucosa of the colon using the labeled mitoses-wave method.The epithelial cell proliferation was significantly decreased in the colon of the Vivonex-fed animals.     

On the yolk sac of the cat

Summary The phase of primitive erythropoiesis in the feline yolk sac lasts from the 14th to the 20th day after mating. The globular nucleated primitive erythroblasts are formed extravascularly to some extent, but they can be clearly distinguished from the endoderm. They do not undergo a denucleation and are still present in the circulating blood on the 45th day. Aging primitive erythroblasts are characterized by a loss of polysomes, by the appearance of long intracytoplasmic electron-lucent channels, and by a nuclear pyknosis which can turn into a karyolysis. Definitive erythropoiesis begins around the 17th day but, even by the 19th day, it is not particularly prominent. It ends around the 45th day. It is almost exclusively intravascular. The distinction of immature primitive erythroblasts from erythroblasts of the definitive series is difficult, because it is based upon only slight differences in the heterochromatinization, in the nuclear-cytoplasmic ratio, and in the organelle content of the cells. In the definitive series, the nuclear divisions follows the law of the rhythmical halving of the nuclear volume. The cells exhibit more clearly identifiable maturation stages here, and the checkerboard nucleus is more distinct. The vascular endothelium is largely attenuated and moderately fenestrated; it lacks a distinct basement membrane. Organelle-rich adventitial cells are found in close apposition.     

Septate junctions in the cephalic epidermis of turbellarians (Bipalium)

Summary In the epidermis of turbellarians septate junctions of the pleated sheet type have been demonstrated in conventional thin sections and freeze fractured preparations.
