An optimized protocol was developed for in vitro plant regeneration of a medicinally important herb Hedychium coronarium J. Koenig using sprouted buds of rhizomes. The rhizomes with sprouted bud were inoculated on Murashige and Skoog (Physiol Plant 15:473–497, 1962) medium (MS) supplemented with either N6-benzyladenine (BA) alone (1.0–4.0 mg L−1) or in combination with 0.5 mg L−1 naphthalene acetic acid (NAA). Of these combinations, MS supplemented with a combination of 2.0 mg L−1 BA and 0.5 mg L−1 NAA was most effective. In this medium, best shoots (3.6) and roots (4.0) regeneration was observed simultaneously with an average shoot and root length of 4.7 cm and 4.2 cm respectively. Regeneration of shoots and roots in the same medium at the same time (One step shoot and root regeneration) reduced the time for production of in vitro plantlets and eliminates the media cost of rooting. Cent-percent (100 %) success in plant establishment was observed in both gradual acclimatization process as well as when plants were directly transferred to outdoor in clay pots containing a mixture of garden soil and sand (2:1) without any sequential acclimatization stage.  相似文献

Increased Abeta42 production has been linked to the development of Alzheimer disease. We now identify a number of compounds that raise Abeta42. Among the more potent Abeta42-raising agents identified are fenofibrate, an antilipidemic agent, and celecoxib, a COX-2-selective NSAID. Many COX-2-selective NSAIDs tested raised Abeta42, including multiple COX-2-selective derivatives of two Abeta42-lowering NSAIDs. Compounds devoid of COX activity and the endogenous isoprenoids FPP and GGPP also raised Abeta42. These compounds seem to target the gamma-secretase complex, increasing gamma-secretase-catalyzed production of Abeta42 in vitro. Short-term in vivo studies show that two Abeta42-raising compounds increase Abeta42 levels in the brains of mice. The elevations in Abeta42 by these compounds are comparable to the increases in Abeta42 induced by Alzheimer disease-causing mutations in the genes encoding amyloid beta protein precursor and presenilins, raising the possibility that exogenous compounds or naturally occurring isoprenoids might increase Abeta42 production in humans.  相似文献

Gene-based tests of association can increase the power of a genome-wide association study by aggregating multiple independent effects across a gene or locus into a single stronger signal. Recent gene-based tests have distinct approaches to selecting which variants to aggregate within a locus, modeling the effects of linkage disequilibrium, representing fractional allele counts from imputation, and managing permutation tests for p-values. Implementing these tests in a single, efficient framework has great practical value. Fast ASsociation Tests (Fast) addresses this need by implementing leading gene-based association tests together with conventional SNP-based univariate tests and providing a consolidated, easily interpreted report. Fast scales readily to genome-wide SNP data with millions of SNPs and tens of thousands of individuals, provides implementations that are orders of magnitude faster than original literature reports, and provides a unified framework for performing several gene based association tests concurrently and efficiently on the same data. A series of 2-(thienyl-2-yl or -3-yl)-4-furyl-6-aryl pyridine derivatives were designed, synthesized, and evaluated for their topoisomerase I and II inhibition and cytotoxic activity against several human cancer cell lines. Compounds 1019 showed moderate topoisomerase I and II inhibitory activity and 2029 showed significant topoisomerase II inhibitory activity. Structure–activity relationship study revealed that 4-(5-chlorofuran-2-yl)-2-(thiophen-3-yl) moiety has an important role in displaying topoisomerase II inhibition.  相似文献

Vermiconversion of industrial sludge for recycling the nutrients   总被引:1,自引:0,他引:1  
The aim of the present study was to investigate the transformation of sugar mill sludge (PM) amended with biogas plant slurry (BPS) into vermicompost employing an epigeic earthworm Eisenia fetida. To achieve the objectives experiments were conducted for 13 weeks under controlled environmental conditions. In all the waste mixtures, a decrease in pH, TOC, TK and C:N ratio, but increase in TKN and TP was recorded. Maximum worm biomass and growth rate was attained in 20% PM containing waste mixture. It was inferred from the study that addition of 30-50% of PM with BPS had no adverse effect on the fertilizer value of the vermicompost as well as growth of E. fetida. The results indicated that vermicomposting can be an alternate technology for the management and nutrient recovery from press mud if mixed with bulking agent in appropriate quantities.  相似文献

Abstract

A colony maintenance system is described. This method allowed development of neonate larvae through to the adult stage, to be completed in the container without changing the diet or handling of various stages. It reduced larval inoculation time by c. 85% (as compared with rearing larvae individually). This reduced overall rearing costs.  相似文献

Background

Alzheimer??s disease (AD) is the leading cause of dementia among the elderly. Disease modifying therapies targeting A?? that are in development have been proposed to be more effective if treatment was initiated prior to significant accumulation of A?? in the brain, but optimal timing of treatment initiation has not been clearly established in the clinic. We compared the efficacy of transient pharmacologic reduction of brain A?? with a ??-secretase inhibitor (GSI ) for 1?C3?months (M) treatment windows in APP Tg2576 mice and subsequent aging of the mice to either 15M or 18M.

Results

These data show that reducing A?? production in a 2-3M windows both initiated and discontinued before detectable A?? deposition has the most significant impact on A?? loads up to 11M after treatment discontinuation. In contrast, initiation of treatment for 3M windows from 7-10M or 12-15M shows progressively decreasing efficacy.

Conclusions

These data have major implications for clinical testing of therapeutics aimed at lowering A?? production, indicating that; i) these therapies may have little efficacy unless tested as prophylactics or in the earliest preclinical stage of AD where there is no or minimal A?? accumulation and ii) lowering A?? production transiently during a critical pre-deposition window potentially provides long-lasting efficacy after discontinuation of the treatment.  相似文献

In E. marginata 12 nonarticulated laticifer initials arise in the cotyledonary node of the young embryo during the early heart stage. The initials arise progressively in the developing embryo, the first laticifers differentiating simultaneously with or shortly before the elements of the pro-cambium. The laticifers occupy a position lateral to the six procambial strands which are formed in the embryo. Upon subsequent growth each laticifer becomes vacuolated and nuclear division unaccompanied by cytokinesis results in the formation of a coenocytic protoplast. The enlarging laticifer produces several branches, one growing into the cotyledon, another growing down along the hypocotyl penetrating toward the root meristem, and one or several growing along intercellular spaces of adjacent cells. No fusion of these branches with one another or adjoining parenchyma cells was observed.  相似文献
