Zinc (Zn) is an essential micronutrient required for growth and development of all organisms. Deficiency of Zn in humans is widespread, affecting 25% of world population and efforts are underway to develop crop plants with high levels of Zn in their edible parts. When strategies for enhancing Zn in crop plants are designed, it is essential to exclude cadmium (Cd), a toxic analogue of Zn. In the present work, a high affinity and high specificity zinc transporter gene (tzn1) from Neurospora crassa was cloned and introduced into Nicotiana tabacum with the objective of enhancing the potential of plants for zinc acquisition. When grown in hydroponic medium spiked with 65Zn, transgenic plants showed enhanced accumulation of Zn (up to 11 times) compared to control plants, which was confirmed further by environmental scanning electron microscopy coupled with Energy Dispersive X‐ray analysis. More importantly, no significant difference in uptake of Cd2+, Fe2+, Ni2+, Cu2+, Mn2+ and Pb2+ between the transgenic and control plants was observed. The present studies have shown that Neurospora crassa tzn1 is a potential candidate gene for developing transgenic plants for improving Zn uptake, without co‐transport of Cd and may have implications in Zn phytofortification and phytoremediation.  相似文献

THE Lyon hypothesis of X chromosome inactivation predicts that in mammalian females, somatic cells are mosaic with respect to whether the active X chromosome is of maternal or paternal origin and that this chromosomal mosaicism is heritable somatically1. Studies of cell clones derived from females who were heterozygous for genes at one of several X-linked loci2–6 have provided good evidence for such mosaicism. Proof that only one of the two X chromosomes, however, is active in any given cell rests on the demonstration that the cell or its clone expresses all of the X-linked genes from one parent and none from the other parent. For this purpose it is useful to examine cloned cells from female subjects for genetic markers representing allelic genes at two or more of the parental loci. This study was undertaken to determine whether genes at the X-linked loci for glucose-6-phosphate dehydrogenase (G6PD) and phosphoglycerate kinase (PGK) are consistently expressed in the eis position in cloned cells as would be expected from a single parental contribution.  相似文献

Time to flowering in the winter growth habit bread wheat is dependent on vernalization (exposure to cold conditions) and exposure to long days (photoperiod). Dominant Vrn-1 (Vrn-A1, Vrn-B1 and Vrn-D1) alleles are associated with vernalization independent spring growth habit. The semidominant Ppd-D1a mutation confers photoperiod-insensitivity or rapid flowering in wheat under short day and long day conditions. The objective of this study was to reveal the nature of interaction between Vrn-1 and Ppd-D1a mutations (active alleles of the respective genes vrn-1 and Ppd-D1b). Twelve Indian spring wheat cultivars and the spring wheat landrace Chinese Spring were characterized for their flowering times by seeding them every month for five years under natural field conditions in New Delhi. Near isogenic Vrn-1 Ppd-D1 and Vrn-1 Ppd-D1a lines constructed in two genetic backgrounds were also phenotyped for flowering time by seeding in two different seasons. The wheat lines of Vrn-A1a Vrn-B1 Vrn-D1 Ppd-D1a, Vrn-A1a Vrn-B1 Ppd-D1a and Vrn-A1a Vrn-D1 Ppd-D1a (or Vrn-1 Ppd-D1a) genotypes flowered several weeks earlier than that of Vrn-A1a Vrn-B1 Vrn-D1 Ppd-D1b, Vrn-A1b Ppd-D1b and Vrn-D1 Ppd-D1b (or Vrn-1 Ppd-D1b) genotypes. The flowering time phenotypes of the isogenic vernalization-insensitive lines confirmed that Ppd-D1a hastened flowering by several weeks. It was concluded that complementary interaction between Vrn-1 and Ppd-D1a active alleles imparted super/very-early flowering habit to spring wheats. The early and late flowering wheat varieties showed differences in flowering time between short day and long day conditions. The flowering time in Vrn-1 Ppd-D1a genotypes was hastened by higher temperatures under long day conditions. The ambient air temperature and photoperiod parameters for flowering in spring wheat were estimated at 25°C and 12 h, respectively.  相似文献

Genetic characterization of germplasm resources is necessary for their effective management and efficient utilization, especially for species like mulberry in which the available germplasm exhibits rich phenotypic diversity with almost no information about its genetic base. Here we present the first report on the isolation of six novel microsatellite markers of mulberry, developed from an enriched genomic library of Morus indica. These markers revealed a high degree of polymorphism (14–26 alleles per locus; polymorphic information content, 0.85–0.90) and a broad cross‐species affinity when tested on a set of 43 elite genotypes including 13 related Morus species. The data thus demonstrate their utility as potentially efficient genetic markers for germplasm characterization, crop improvement and molecular systematics of mulberry.  相似文献

γ-Secretase is a multiprotein intramembrane cleaving aspartyl protease (I-CLiP) that catalyzes the final cleavage of the amyloid β precursor protein (APP) to release the amyloid β peptide (Aβ). Aβ is the primary component of senile plaques in Alzheimer's disease (AD), and its mechanism of production has been studied intensely. γ-Secretase executes multiple cleavages within the transmembrane domain of APP, with cleavages producing Aβ and the APP intracellular domain (AICD), referred to as γ and ε, respectively. The heterogeneous nature of the γ cleavage that produces various Aβ peptides is highly relevant to AD, as increased production of Aβ 1-42 is genetically and biochemically linked to the development of AD. We have identified an amino acid in the juxtamembrane region of APP, lysine 624, on the basis of APP695 numbering (position 28 relative to Aβ) that plays a critical role in determining the final length of Aβ peptides released by γ-secretase. Mutation of this lysine to alanine (K28A) shifts the primary site of γ-secretase cleavage from 1-40 to 1-33 without significant changes to ε cleavage. These results further support a model where ε cleavage occurs first, followed by sequential proteolysis of the remaining transmembrane fragment, but extend these observations by demonstrating that charged residues at the luminal boundary of the APP transmembrane domain limit processivity of γ-secretase.  相似文献

The effect of four monoterpenes ‐ citronellol, citronellal, cineole and linalool ‐ on the germination, growth and physiology of Cassia occidentalis was investigated. All four monoterpenes reduced germination of C. occidentalis seeds but to varying extents. Citronellal and linalool completely inhibited germination beyond the concentrations of 55 and 110 μM, respectively, whereas in response to treatment of citronellol no germination was observed beyond 330 μM. Further, the growth of seedlings, measured in terms of seedling length and biomass, was also adversely affected. A reduction in chlorophyll content of the cotyledonary leaves of C. occidentalis was also noticed, indicating an adverse effect on photosynthesis. Likewise, respiratory ability of growing seeds was also impaired in response to all four monoterpenes, clearly indicating that monoterpenes affect energy metabolism. On the basis of overall phytotoxicity, potency of monoterpenes was in the order of citronellal > linalool > citronellol > cineole. The results from this study suggest that both citronellal and linalool possess strong phytotoxic potential and can thus serve as lead molecules for the synthesis of bioherbicides.  相似文献

In most tropical regions where wheat is grown under irrigation, high temperatures at sowing adversely affect crop establishment and subsequent seedling survival. The objective of this study was to compare wheat (Triticum aestivum) genotypes for their ability to germinate and grow at high temperatures during the seedling stage. Twenty-five seeds each of 14 spring wheat cultivars were placed on moist filter paper at different temperatures (5°C to 40°C) in a one-way thermogradient plate to determine the cardinal temperatures for germination. Rate of germination at each temperature for each genotype was computed as the inverse of time taken for 50% of the seeds to germinate. Rate of germination for each genotype at different temperatures was modelled with temperature to determine the base (tb), and optimum (topt) temperatures. Response of germination to temperature for each genotype was calculated as the slope of a linear regression of the rate of germination on temperature below topt. Genotypes differed in their optimum temperatures and Mexipak (= Kalyansona) had the lowest. Range in base temperature among the genotypes was between 0°C and 2°C differences but were not statistically significant though they might be biologically significant. Genotypes differed in their response to temperature with Gomam having the lowest rate, implying that it was slow to respond to increasing temperatures. Debeira and Cham 6 showed a similar response. Three lines which had performed well in spring wheat evaluation trials for moderate rainfall areas under heat stress had the highest response rate. It is concluded that combining higher optimum temperatures with faster response rates would result in better-adapted germplasm for regions where high temperatures persist at sowing.  相似文献

A new series of 2,4-diphenyl-6-aryl pyridines containing hydroxyl group(s) at the ortho, meta, or para position of the phenyl ring were synthesized, and evaluated for topoisomerase I and II inhibitory activity and cytotoxicity against several human cancer cell lines for the development of novel anticancer agents. Structure–activity relationship study revealed that the substitution of hydroxyl group(s) increased topoisomerase I and II inhibitory activity in the order of meta > para > ortho position. Substitution of hydroxyl group on the para position showed better cytotoxicity.  相似文献
