When chopped porcine pulmonary arteries were incubated with calcium ionophore A23187 (1) in the presence of indomethacin there was a time dependent generation of a substance which produced contractions of superfused strips of guinea-pig ileum smooth muscle (GPISM) which were indistinguishable from those induced by LTD4. This material however had a different retention time from LTD4 when subjected to HPLC and co-chromatographed with synthetic LTE4. In addition to LTE4 a substance which had properties indistinguisable from those of LTB4 when assayed on a combination of guinea-pig lung parenchymal strips (GPP) and GPISM (2) was generated from the pulmonary artery. This substance co-chromatographed with synthetic LTB4. The adventitia and intima were the richest source of LTE4, the adventitia releasing slightly more than the intima. The output of LTB4 and LTE4 was inhibited by 6,9-deepoxy-6,9-(phenylimino)-Δ6,8 prostaglandin I1 (U-60,257). Nordihydroguaiaretic acid (NDGA) inhibited the generation of LTE4. 相似文献

Using dithionite difference spectra we have detected cytochrome b in highly purified human neutrophils at a concentration of 0.08 nmol/mg protein. The presence of quinone was identified in lipid extracts at a concentration of approx. 0.06 nmol/mg protein. It was identified as ubiquinone-10 by mass spectrographic analysis. Simultaneous measurements of cytochrome oxidase indicated that these compounds could not be attributed to mitochondrial contamination. These results are compatible with the hypothesis that initiation of the respiratory burst in human neutrophils involves a multicomponent electron-transport system. 相似文献

Summary Dispersed cells from anterior pituitary glands of male rats were cultured embedded in collagen gels or on a plastic surface. After 6 or more days in culture, cells in collagen produced more prolactin than those on plastic. The cultures in collagen had fewer fibroblasts than those on plastic. Many cells cultured in collagen developed processes, and a few cells in every culture had long extended processes that sometimes branched and resembled those of neurons. About 60% of the cells in culture including cells with well developed processes bound the monoclonal antibody A2B5, an antibody that binds to neuronal cells in culture. Fibroblasts did not have detectable binding of A2B5. Some cells with short processes reacted with antiserum to prolactin or ACTH, but the cells with neuronlike processes did not. Collagen gels may provide a matrix in which cells from the anterior pituitary gland can differentiate into neuronlike cells. 相似文献

A proprietary aspirin, phenacetin, and caffeine preparation given to rats in a dose equivalent to that taken by patients with analgesic nephropathy produced papillary necrosis in 55%. There was a higher incidence in rats deprived of fluids overnight.Papillary necrosis was not noted in rats receiving twice as much phenacetin.These findings support the argument that phenacetin should not be singled out as the substance responsible for analgesic nephropathy in man. 相似文献

Flowers of 23 species representing six subfamilies of Araceae were studied by means of serial cross sections, special attention being given to vascular patterns and to taxa of supposed phylogenetic importance. Floral structure is shown to be extremely diverse with no unifying pattern common to all subfamilies. Conclusions include the following: (1) Lysichiton has a specialized gynoecial vascular pattern which differs from others encountered in the survey and which weighs against the primitive position attributed to this genus by Hutchinson. (2) Philodendron, with its multiple stylar canals, cannot have originated from subfamily Pothoideae, as Engler's phylogenetic concept would require of all Araceae; instead, it appears that several syncarpous evolutionary lines have evolved independently from extinct apocarpous members of the family. (3) In Acorus, stamens are introrse and dorsal carpellary bundles are lacking; these characters and others justify the recognition of Acorus as a separate subfamily Acoroideae. In addition, the survey revealed a peculiar deterioration of the inner ovary wall and the septa in several taxa, apparently a normal feature of floral development. Abstract The mycobacterial cell wall visualized by transmission electron microscopy (TEM) of thin sections of resin-embedded specimens is generally believed to consist of an electron-dense peptidoglycan, an electron-transparent arabinogalactan-mycolate layer and an electron-dense outer layer (OL). In addition, a pseudocapsule known as the ‘electron-transparent zone’ (ETZ) has been observed after phagocytosis of mycobacteria by macrophages. TEM of thin sections of Mycobacterium bovis BCG, Tice® substrain, revealed an OL bilayer, each of which measured 2–4 nm in diameter. The intermediate electron-transparent layer varied from 1 to about 250 nm in diameter and appears to be a previously observed oxygen-dependent amorphous integument that consists of hot water-extractable neutral polysaccharides, especially a recently characterized α-glucan, comprising about 12% of the dry cell weight. This and other recent studies of BCG have revealed cell-surface features that may provide a better understanding of the outer mycobacterial cell envelope. 相似文献

A 2041 bp DNA fragment isolated from the Sxr (sex reversed) region of the mouse Y Chromosome (Chr) was sequenced and characterized. The sequence, pY8/b, contains four exons that are highly similar to 525 contiguous bases from the cDNA of human ubiquitin activating enzyme El. Two of the exons contain stop codons, indicating that pY8/b is not part of a functional gene. Sequences related to pY8/b were amplified from the Y Chr of the inbred mouse strain, C57BL/6J. These sequences may be portions of the recently discovered functional equivalent of pY8/b. Despite a high degree of similarity with the human El gene, the functional equivalent of pY8/b is not the mouse El gene, because unlike El, the functional equivalent of pY8/b is expressed in a tissue-specific manner. These data are discussed with respect to theory on the evolution of the mammalian Y Chr, and in particular, to the prediction that functional genes on the Y Chr have a male-specific function. 相似文献

Gene transfer into somatic tissues is a tool for both the study of gene function in the basic science laboratory and for gene therapy and genetic immunization in the clinic. Biolistic processes can be used to deliver both viral and nonviral vectors into somatic tissues. This review discusses the advantages and disadvantages of three biolistic processes: jet injection, microparticle bombardment, and needle and syringe injection. Jet injection and needle and syringe injection can be used to deliver both viral and nonviral vectors. Both jet injection and microparticle bombardment can be used to target a broad range of tissues. Needle and syringe injection has been most widely used in muscle tissue. The choice of which biolistic process to use is dependent on the specific application. 相似文献
