Antimicrobial and antiviral activities of the fractions from Scutia buxifolia stem bark and leaves were evaluated. Best antimicrobial results occurred with the ethyl acetate (EA) and n-butanolic (NB) fractions from the leaves against Micrococcus sp. (minimal inhibitory concentration—MIC = 62.5 μg/ml), and NB fraction from stem bark and leaves against Klebsiella pneumoniae and Enterococcus faecalis (MIC = 62.5 μg/ml). The most active fractions were selected and fractioned into silica column to perform an in vitro antibiofilm assay, which evidenced subfractions EA2 and EA3 as the more active against Candida albicans (biofilm inhibitory concentration—BIC = 582 ± 0.01 μg/ml) and Staphylococcus aureus (BIC = 360 ± 0.007 μg/ml), respectively. The NB (selectivity index—SI = 25.78) and the EA (SI = 15.97) fractions from the stem bark, and the EA (SI = 14.13) fraction from the leaves exhibited a potential antiviral activity towards Herpes Simplex Virus type 1 whereas EA2 and EA3 subfractions from leaves (SI = 12.59 and 10.06, respectively), and NB2 subfraction from stem bark (SI = 12.34) maintained this good activity. Phenolic acids and flavonoids (gallic acid, chlorogenic acid, caffeic acid, rutin, isoquercitrin, quercitrin and quercetin) were identified by HPLC and may be partially responsible for the antimicrobial and antiherpes activities observed. The results obtained in this study showed that Scutia buxifolia has antibiofilm and anti-herpetic activities and that these properties are reported for the first time for this species. 相似文献

Brazil is one of the most important endemic areas for leishmaniasis worldwide. Protected areas that are tourist attractions likely present an important risk of transmission of cutaneous leishmaniasis (CL). Furthermore, with the geographical expansion of visceral leishmaniasis (VL), several studies have recorded the occurrence of its vector, Lutzomyia longipalpis, and cases of human and canine VL in such tourist areas. The Parque Estadual do Sumidouro is an environmentally protected area located in the Brazilian Cerrado biome and in an important area endemic for leishmaniasis in the state of Minas Gerais. The purpose of this study was to monitor the sand fly fauna in areas of tourist activity in the park. Sampling was performed every month, from September 2011 to August 2013, using CDC light traps at six sites of differing environmental characteristics. Sampled specimens were identified following Galati (2003), and females were submitted to molecular techniques for the detection and identification of Leishmania DNA. A total of 4,675 sand fly specimens of 25 species belonging to nine genera were collected. The most abundant species were Micropygomyia quinquefer, Lutzomyia renei and Pintomyia pessoai, although only Pi. pessoai is implicated in the transmission of Leishmania braziliensis. The species accumulation curve reached saturation on the 16th sampling event. Species richness, diversity and evenness differed among the sampled areas. The seasonal curve was not determined by a single unique species, and no single species was the most abundant in all environments sampled. The main vector of Leishmania (Leishmania) infantum, Lutzomyia longipalpis, accounted for only 5.35% of the specimens collected. Proven or suspected vectors of Leishmania (Viannia) braziliensis were recorded, and one female of the cortellezzii complex tested positive for Le. braziliensis DNA. Even with a low infection rate (0.62%), these data indicate the circulation of the parasite and reinforce the need for entomological and epidemiological surveillance in the park and its surroundings. 相似文献

The effect of glycerol on xylose-to-xylitol bioconversion by Candida guilliermondii was evaluated by its addition (0.7 and 6.5 g/l) to semidefined media (xylose as a substrate). The glycerol concentrations
were chosen based on the amounts produced during previous studies on xylitol production by C. guilliermondii. Medium without glycerol addition (control) and medium containing glycerol (53 g/l) in substitution to xylose were also evaluated.
According to the results, the addition of 0.7 g/l glycerol to the fermentation medium favored not only the yield (Y
P/S = 0.78 g/g) but also the xylitol productivity (Q
P = 1.13 g/l/h). During the xylose-to-xylitol bioconversion, the formation of byproducts (glycerol and ethanol) was observed
for all conditions employed. In relation to the cellular growth, glycerol as the only carbon source for C. guilliermondii was better than xylose or xylose and glycerol mixtures, resulting in a maximum cellular concentration (5.34 g/l). 相似文献

Kappaphycus alvarezii is a seaweed of great economic importance for the extraction of kappa carrageenan from its cell walls. The most common strains are dark red, brown, yellow, and different gradations of green.
It is known that ultraviolet radiation (UVR) affects macroalgae in many important ways, including reduced growth rate, reduction
of primary productivity, and changes in cell biology and ultrastructure. Therefore, we examined the brown strain of K. alvarezii exposed to ultraviolet-B radiaton (UVBR) for 3 h per day during 28 days of cultivation. The control plants showed growth
rates of 7.27% d−1, while plants exposed to UVBR grew only 4.0% d−1. Significant differences in growth rates and in phycobiliproteins between control and exposed plants were also found. Compared
with control plants, phycobiliprotein contents were observed to decrease after UV-B exposure. Furthermore, the chlorophyll
a (Chl a) contents decreased and showed significant differences. UVBR also caused changes in the ultrastructure of cortical and subcortical
cells, which included increased thickness of the cell wall and number of plastoglobuli, reduced intracellular spaces, changes
in the cell contour, and destruction of chloroplast internal organization. Reaction with Toluidine Blue showed an increase
in the thickness of the cell wall, and Periodic Acid-Schiff stain showed a decrease in the number of starch grains. By the
significant changes in growth rates, photosynthetic contents and ultrastructual changes observed, it is clear that UVBR negatively

Cestrum strigilatum (Solanaceae) is a South American shrub with B chromosomes. Bs show a univalent behavior when a single B is present, have non-Mendelian segregation, and are poor in genes and rich in repetitive DNA. In this study, the histone H3 at serine 10 (H3/Ser10) phosphorylation pattern was investigated during mitosis and meiosis of C. strigilatum collected from the wild and was compared in A and B chromosomes. The results revealed that H3/Ser10 phosphorylation of A chromosomes occurred only in the pericentromeric region in both mitosis and meiosis, whereas in the B univalent, phosphorylation appeared in almost the whole extent of the chromosome, except in the terminal portion of the long arm. In meiosis II, the phosphorylation of A chromosomes was similar to that in the first division of meiosis, but the Bs did not show H3/Ser10 phosphorylation. Our results suggest that phosphorylation at the pericentromeric region may be associated with chromosome motility during cell divisions and with the cohesion of B chromatids in a univalent structure in meiosis I. 相似文献

In this work, we review the physiological and molecular mechanisms that allow vascular plants to perform photosynthesis in extreme environments, such as deserts, polar and alpine ecosystems. Specifically, we discuss the morpho/anatomical, photochemical and metabolic adaptive processes that enable a positive carbon balance in photosynthetic tissues under extreme temperatures and/or severe water‐limiting conditions in C3 species. Nevertheless, only a few studies have described the in situ functioning of photoprotection in plants from extreme environments, given the intrinsic difficulties of fieldwork in remote places. However, they cover a substantial geographical and functional range, which allowed us to describe some general trends. In general, photoprotection relies on the same mechanisms as those operating in the remaining plant species, ranging from enhanced morphological photoprotection to increased scavenging of oxidative products such as reactive oxygen species. Much less information is available about the main physiological and biochemical drivers of photosynthesis: stomatal conductance (gs), mesophyll conductance (gm) and carbon fixation, mostly driven by RuBisCO carboxylation. Extreme environments shape adaptations in structures, such as cell wall and membrane composition, the concentration and activation state of Calvin–Benson cycle enzymes, and RuBisCO evolution, optimizing kinetic traits to ensure functionality. Altogether, these species display a combination of rearrangements, from the whole‐plant level to the molecular scale, to sustain a positive carbon balance in some of the most hostile environments on Earth. 相似文献
